Immunology- nobel prizes
Since 1901 there have been 19 Nobel Prizes for
immunology-related research.
كان آخرها جائزة نوبل ف الطب
للمعالجة المناعة للسرطان

Immunity
Innate خلقة( الطبعة/فطرة/متأصلة)
(Nonspecific) 1o line of defense:
0-12 h
Adaptive التلاؤمة (Acquired/Specific) 2o line of defense:
days to weeks
![Page 8: ةعبنملا ملع Immunology](https://reader031.vdocuments.net/reader031/viewer/2022022112/6212318794e4e166771e7247/html5/thumbnails/8.jpg)
Innate immunity vs Adaptive Immunity
-No memory
o-No time lag ; mediated
•by cells and proteins that
•are always present
•-Not antigen specific
•A lag period
•Antigen specific
•Development

Neutrophils in innate immune response
• Most abundant WBCs (~50-60%)
• Efficient phagocytes : microphage
Phago = to eat; cyte = cell
• find, eat and digest microbes !
• Most important cells of the innate immune system
![Page 23: ةعبنملا ملع Immunology](https://reader031.vdocuments.net/reader031/viewer/2022022112/6212318794e4e166771e7247/html5/thumbnails/23.jpg)
How do neutrophils eat and digest microbes ?
Granules
Lysozyme – digests bacterial cell wall; other antimicrobial proteins
![Page 24: ةعبنملا ملع Immunology](https://reader031.vdocuments.net/reader031/viewer/2022022112/6212318794e4e166771e7247/html5/thumbnails/24.jpg)

Kills cancer cells

NK cells choose cells to kill
Uninfected cell / Normal cell
Microbe infected cell / cancer cell
Some cell surface proteins are missing
NK cells have two types of Receptors: inhibitory and activating. Inhibitory receptors recognize self class I MHC molecules, which are expressed on all nucleated healthy cells (CD47 in RBCs) Activating receptors
recognize modified molecules on stressed or infected cells.

How does the killer kill ?
Kills both host cells and microbes
Release of granules with perforins and proteases

Receptors of Innate Immunity (PRRs) • Toll-Like receptors (TLRs) : in
different cellular compartments
• NOD-like receptors (NLRs): in cytosol; they recognize a wide variety of substances, including products of necrotic cells, and some microbial products.
• C-type lectin receptors (CLRs):
membrane of macrophages and Dendritic cells; recognition of fungi
• RIG-like receptors (retinoid acid-inducible gene receptor): intracellular; for viral RNA
![Page 35: ةعبنملا ملع Immunology](https://reader031.vdocuments.net/reader031/viewer/2022022112/6212318794e4e166771e7247/html5/thumbnails/35.jpg)
Toll-like receptors (TLRs)
(مستقبلات جزس الانذار) ببلتولالمستقبلات الشبيهة
• Transmembrane proteins: receptors of innate immunity present on cell membrane and other cell compartments.
• 10 known types in humans.
• Conserved across vertebrates (Inherited)
• Present on macrophages / other cells (epithelial cells…)
• Important part of innate immune system

TLRs – What do they do ?
• They look out for microbes (or their components)
• They bind to the microbes (or their components)
• They trigger a cascade of events to kill or protect against pathogens
• THEY ARE INNATE IMMUNE SENSORS
![Page 37: ةعبنملا ملع Immunology](https://reader031.vdocuments.net/reader031/viewer/2022022112/6212318794e4e166771e7247/html5/thumbnails/37.jpg)
