An improved bacterial culturing system which is advantageously used on-site at waste water treatment plants. The system rapidly and effic- iently produces various types of bacteria having enhanced activity and controllable charac- teristics. The bacteria are used to break down un- desired or unwanted chemical species contained in the waste water stream. The system includes a unique culturing or primary basin in com- munication with an acclimation or acclimator basin. Bacteria grown in the culturing basin are partially exposed to the materials they are later intended to break down in the acclimation basin.

DNA fragments (seeds) having the charac- teristics of amplicons, which are useful for am- plifying genes of interest, have been isolated from Marek's disease viruses of poultry. Con- catmers of the seeds and the associated genes have potential as vaccines or delivery vectors when cotransfected and replicated in the pre- sence of helper viruses. The amplicons are also useful for inserting associated genes into the hel- per viruses, which in turn could be used as ex- pression vectors. Candidate genes for use with the subject amplicons include those which encode immunogenic proteins and other benefi- cial economic traits desired in commercial poultry lines.
