Agilent QPCR and QRT-PCR Kits
These kits are used to sensitively quantify RNA expression levels (QRT-PCR) or to determine the DNA concentration (QPCR) of a sample
•Brilliant® was our line of traditional QPCR and QRT-PCR kits. All of these kits with the exception of the Brilliant Multiplex Master Mix were discontinued in 2011
•Brilliant® Multiplex kit is designed specifically for multiple amplification reactions in a single tube
•Brilliant® II kits are formulated for higher sensitivity
•Brilliant® III kits are formulated for fast cycling allowing shorter run times
•Brilliant® Sidestep and Sidestep II kits allow for QPCR and QRT-PCR directly from cell lysate, with no purification step necessary
•miRNA, FFPE and NGS Library Quantification
Brilliant® II QPCR & QRT-PCR Reagent Kits
• Consistent results and versatility
• Compatible with multiple fluorescent detection chemistries, including:
-SYBR™ Green I dye
-TaqMan probes
-Molecular Beacons
-Others...
• Several different kit configurations allow customers to choose the one that is right for them,
master mix and core reagent kits, multiplexing kits
•Brilliant II kits contain chemically modified hotstart Taq (SureStart) -Reduces nonspecific primer annealing and extension
•Brilliant II kits include a higher concentration of Taq polymerase for improved sensitivity
compared to the original Brilliant kits

Competitor Q
Earliest Ct value
Amplification plot of cyclophillin target in 8 replicates at 100-fold dilutions from 100ng down to 0.01ng (100pg) of template. The improved sensitivity of
Brilliant II is evident in earlier Ct values compared to original Brilliant, Competitor A and Competitor Q. In addition, replicate reproducibility is better
with Brilliant II compared to most others , particularly at 0.01ng. cDNA from Human Reference RNA used as template.
Brilliant® II
Reference Dye (free ROX)
All the QPCR and QRT-PCR kits are available with Passive Reference dye
• Included for normalization
• Controls for pipetting variability during assay setup
• Required in some instruments to correct for well to well signal variation, but
optional in the Mx instruments
Brilliant II and Brilliant III High and Low ROX kits have the reference dye pre-
mixed in the Master Mix so less aliquoting is necessary and you can order
the catalog number that is right for their instrument.
Brilliant III Ultra-Fast
-Utilizes mutant Taq that is capable of fast extension (runs completed in
under 40 minutes)
-Enhanced chemically-mediated hotstart allows for shorter hotstart
activation step
-QRT-PCR kits use a Moloney-based RT for 1st strand synthesis with
optimal performance at a synthesis temperature of 50°C.
-Protocols optimized to run on fast-cycling QPCR instruments such as
the ABI StepOnePlus and BioRad CFX96

QRT-PCR
Absolutely RNA® FFPE Kit
For purification of RNA from formalin fixed, paraffin embedded tissue samples
Results in highly pure DNA-free total RNA that is suitable for QRT-PCR
Method removes modifications to the RNA that occur during the fixation process that would prevent amplification
Phenol-free, non-hazardous method
High-Specificity miRNA
QRT-PCR Detection Kit
• The first step of the kit involves elongation with with E. coli poly A polymerase (PAP) to generate a poly-A tail at the 3´ end of each RNA molecule.
• A 1st strand synthesis is then performed using AffinityScript™ RT.
• The cDNA synthesis reaction is primed using an RT adapter primer containing a polyT sequence with the addition of a universal reverse primer binding site at the 5’ end.
• QPCR is then carried out using a Universal Reverse Primer (included with the kit) that will anneal to the tag added in cDNA synthesis, and an miRNA-specific forward primer which provides the target specificity.
• The dsDNA-binding dye EVA Green is used for detection (detected with FAM filter set).

QPCR NGS Library Quantification Kits
• Product offers a means to accurate quantify libraries prior to Next
Generation Sequencing.
• When preparing DNA libraries for next generation sequencing, accurate
quantification of the library molarity is required to ensure efficient data
generation.
• Overly concentrated libraries will result in data that is often impossible to
resolve.
• Conversely, concentrations that are too low can reduce sequencing
coverage and read-length.
