Diagnostic activities for Aujesky’s Disease [Pseudorabies (PRV)]

Testing for Ajueszky’s Disease was completed on 553 samples originating within the United States (US) using gB ELISA, gI ELISA, Autolex and Virus Neutralization (VN), and 41 samples of international origin using the same test methods. Virus Isolation (VI) and PCR testing was completed on an additional 20 samples from the US and 41 international samples.The NVSL also produced the following reagents and proficiency test panels which were supplied to OIE Member Countries: PRV virus and positive control sera for VN, antisera for various serology tests, fluorescent antibody (FA) conjugate, and proficiency panels for latex agglutination, gB ELISA, gI ELISA, and VN test methods.

Diagnostic activities for Avian Influenza Virus (AIV) and Newcastle Disease (ND)

Diagnostic testing for AIV and ND was completed on 31,310 samples of national origin using the indirect test methods of agar gel immunodiffusion (AGID) Hemagglutination (HI) and neuraminidase-inhibition (NI) antibody subtype identification for AIV, and HI antibody identification for avian paramyxovirus (APMV). Another 247 samples of international origin were tested by AGID for AIV. NVSL tested another 27,807 diagnostic samples of domestic origin using the direct test methods of HI and NI virus subtype identification, real-time RT-PCR for IAV, ND and APMV; and virus isolation (VI), genomic sequencing and pathogenicity testing for AIV and APMV. A total of 78 samples of international origin were also tested using these techniques.Reagents produced and provided to OIE Member countries included; positive control antigens and type antisera for AIV HI testing; positive and negative control sera and antigen for AGID testing, IAV matrix gene and APMV-1 matrix gene positive amplification and extraction controls for real-time PCR,

H5 and H7 gene real-time PCR positive amplification controls, APMV-1 control positive antigen and positive antisera for HI testing.Two proficiency tests were also produced; a real-time RT PCR panel and an AGID proficiency test.NVSL also provided diagnostic support for three OIE Member countries, and tested a total of 221 samples for routine diagnostic testing, and another 9 samples for confirmatory diagnosis.

Diagnostic activities for Anthrax

A total of 9 samples were submitted to the NVSL for bacteriological culture in 2013, with all samples originating within the U.S.Gamma phage was also produced and distributed to OIE Member Countries for use in the gamma phage lysis diagnostic test method.

Diagnostic activities for bluetongue virus (BTV)

In the US, 697 diagnostic specimens were tested for BTV via indirect test methods [competitive ELISA, AGID, complement fixation (CF) and VN], and another 835 samples were screened using real-time reverse-transcriptase PCR (real-time RT-PCR), nested RT-PCR, and VI.Another 102 international samples were screened by the above direct methods and 232 international samples were screened using real-time RT-PCR, nested RT-PCR, VI, and sheep inoculation.Reagents produced were strong and weak positive reference sera; fluorescent antibody conjugate, Bluetongue virus antiserum for VN, ELISA and AGID ( serotype specific), multiple serotypes of bluetongue stock virus for VN assays, various serotypes for BTV RNA for PCR testing, and a BTV proficiency panel consisting of 20 serum samples for AGID and ELISA methods.Diagnostic testing was carried out for two OIE Member Countries on a total of 27 samples.

Diagnostic activities for contagious equine metritis (CEM)

In 2013 the US tested a total of 1588 samples of domestic origin and 551 samples of international origin using the CF indirect diagnostic test. Another 1167 tests were performed on domestic samples and 1139 tests performed on international samples using the direct methods of culture and identification, identity confirmation and pulse field gel electrophoresis (PFGE).Also in 2013 NVSL produced and provided the following reagents to OIE Member Countries: growth media for CEM (Eugon Agar and other selective agars), CEM high titer control sera for CF testing, CEM control strains for use in identification protocols, a culture and identification proficiency panel, and a CF test ring trial panel.NVSL provided diagnostic consulting to two OIE Member Countries in the form of consultation on the PFGE test.

Diagnostic activities for Eastern equine encephalomyelitis (EEE)

Within the United States (US), a total of 3096 indirect diagnostic tests [CF, hemagglutination inhibition (HI), Plaque Reduction Neutralization (PRN) and IgM Capture ELISA] were performed for EEE, and 66 diagnostic tests performed using RT-PCR. 124 indirect diagnostic tests and 3 direct diagnostic tests were performed on international diagnostic samples using the same methods.NVSL provided diagnostic assistance for five separate OIE Member countries, testing a total of 17 samples submitted during 2013.Reagents supplied to recipient OIE Member countries included EEE virus antigen and antiserum, and normal cell culture antigen for CF, HI and ELISA. EEE equine IgM antiserum for the IgM Capture ELISA test method was also provided.

Diagnostic activities for equine infectious anaemia (EIA)

3,135 diagnostic tests for EIA were performed nationally during 2013, using AGID, ELISA and Immunoblotting. Another 9,382 diagnostic tests were performed for international clients.

Reagents produced include: strong positive, weak positive and negative reference sera for AGID, ELISA and competitive ELISA (cELISA); and an EIA proficiency panel consisting of 20 serum samples for AGID, ELISA and cELISA.Diagnostic testing for diagnostic support was conducted on 170 additional samples for four different OIE Member Countries.

Diagnostic activities for foot and mouth disease (FMD)

A total of 352 indirect tests [ELISA, agar gel immunodiffusion (AGID)] and 923 direct tests [cell cultures, real-time RT PCRs] were performed for diagnostic purposes for foot and mouth disease (FMD) virus on samples originating within the U.S. An additional 18 diagnostic samples of international origin were screened for the detection of FMD using the previously mentioned indirect tests, and 130 international samples were tested using the direct methods listed.Diagnostic testing for diagnostic support was conducted on 9 additional samples for one OIE Member Country.NVSL produced, distributed and evaluated a proficiency test for FMD to 43 participant laboratories in the U.S. Additionally, reagents FMD PCR tests were supplied to 1 OIE Member Country.

Diagnostic activities for Leptospira

In 2013, the NVSL performed 5,867 tests [Leptospira microscopic agglutination test (MAT)] for detection of Leptospira sp. on samples from the US, and another 150 tests on international samples.The following diagnostic reagents were produced and supplied to other OIE Member Countries; reference positive and negative control sera for MAT, reference cultures for MAT, multivalent fluorescent antibody conjugate, and Leptospira culture media.

Diagnostic activities for Swine influenza virus (SIV)

A total of 82 HI serology tests were performed on samples originating outside of the US, and another 319 samples originating in the U.S. were tested by direct methods [RT-PCR, VI and sequencing].The NVSL produced and provided the following products to support OIE diagnostic testing activities in Member Countries: SIV H1N1 virus, antigen and antiserum; H3N2 virus, antigen, and antiserum; FA conjugate; PCR positive controls; PCR proficiency panels; and field isolates from surveillance testing.

Diagnostic activities for Venezuelan equine encephalomyelitis (VEE)

A total of 521 diagnostic samples originating within the US were tested by indirect methods for VEE, using the same tests described above for EEE, and another 103 diagnostic samples of international origin were tested. Three direct diagnostic tests were performed on samples of international origin using RT-PCR.Diagnostic reagents for VEE that were produced and supplied to recipient OIE Member Countries in 2013 were VEE virus antigen and antiserum for performing the HI, DF, ELISA and PRN test methods.Five OIE Member Countries requested assistance with diagnostic support for VEE, and the NVSL tested 19 samples as part of this support.

Diagnostic activities for Vesicular Stomatitis Virus (VSV)

5,854 samples of national origin were tested for VSV using indirect methods of cELISA, CF and VN in2013; another 558 samples of international origin were tested for diagnostic purposes using the same methods. Another 1016 samples of national origin were tested for diagnostic purposes using the direct diagnostic methods of VI, PCR and antigen capture ELISA.The NVSL also produced reagents for VN and CF that were distributed to several OIE Member Countries. Reagents include complement, modifying factor, normal mouse brain antigen, VSV antigens, and positive and negative control sera for NJ and IN strains of VSV, plus positive and negative control sera and viruses for both NJ and IN VSV VN test methods. A total of 15 proficiency panels for the VSV-CF serology test and 2 proficiency panels for VSV-VN test were provided to laboratories within the U.S, and 2 additional

VSV-VN test panels provided to 2 OIE Member Countries. No proficiency panels were produced in 2013.One OIE Member Country also requested assistance in the form of diagnostic testing of 9 samples for VSV.

Diagnostic activities for Western equine encephalomyelitis (WEE)

In 2013, NVSL tested 2030 diagnostic samples from the US via the indirect methods of CF, HI and PRN, and another 70 samples of international origin using the same assays. A total of 44 diagnostic samples were tested using the direct diagnostic method of RT-PCR from the US, and 3 international samples.WEE virus antigen and antiserum reagents were produced and provided to OIE Member Countries for use in HI, CF or ELISA testing.Five OIE Member Countries requested assistance with diagnostic support for WEE, and the NVSL tested 17 samples as part of this support.

Diagnostic activities for West Nile Virus (WNV)

During 2013, the NVSL performed 2041 indirect diagnostic tests [equine IgM capture ELISA, PRN, VN] on samples of domestic origin and another 624 indirect tests for samples originating from other countries. For direct diagnostic tests, another 61 RT-PCR tests were performed on domestic samples, and 4 RT-PCR tests done on international samples.NVSL produced the following reagents that were also supplied to OIE Member Countries: WNV RNA for PCR controls, WNV negative, equine IgM low positive, and equine capture IgM antisera for IgM capture ELISAs, antisera for the PRN and VN assays, and an equine IgM ELISA proficiency panel consisting of 10 serum samples/panel.NVSL provided diagnostic assistance to six OIE Member Countries and tested a total of 47 samples for WNV.

VACCINES

VICH Steering Committee Meeting

February 19-21, attended the 28th International Cooperation on Harmonisation of Technical Requirements for Registration of Veterinary Medicinal Products (VICH) Steering Committee, in Washington, DC, as the U.S. delegation representing the USDA on issues related to veterinary biologics.

Visit with Delegates from the People’s Republic of China

On March 21, representatives from NVSL and CVB met with visitors from the People’s Republic of China, as part of a visit coordinated by Iowa’s International Center, International Visitor Leadership Program. The purpose of the visit is to learn more about NVSL and CVB program missions, and the support they offer the OIE.

Facilitated Transfer of Donated Classical Swine Fever Virus

The CVB participated with the National Veterinary Stockpile to facilitate the movement of Classical Swine Fever Vaccine from Germany, through Miami International Airport, to Guatemala for use in their control program.

Visit with Delegates from Indonesia’s Agency of Drug and Food Control

On May 6, several representatives from the Indonesian National Agency of Drug and Food Control met with delegates from the CVB and NVSL to discuss the roles of U.S. Federal agencies in food and product safety regulation and supervision, as well as aspects of risk management, mitigation, evaluation, resolution and communication.

National Vaccine Advisory Committee attendance

Attended several meetings of the National Vaccine Advisory Committee in Washington, DC. The committee’s function is to encourage the availability of adequate supplies of safe and effective vaccination products, recommend research priorities, identify areas of government and non-government cooperation, provide direction to prevent adverse reactions to vaccines, and recommend measures to achieve optimal prevention of human infectious disease through vaccine development.

As part of a One Health Diagnostic Collaboration on avian influenza

A(H7N9) between USDA & the Centers for Disease Control and Prevention

(CDC), a human-origin avian influenza A(H7N9) virus [A/Anhui/1/2013] was

shared with CDC from China, and subsequently shared with animal health

partners at the NVSL and SEPRL. The virus was propagated and pathotyped

and in a cooperative effort working with national partners such as the

SEPRL as well as international influenza reference laboratories an updated

Eurasian-specific H7 assay was developed.
