Unit 4 ReviewUnit 4 Review

1 - Define the term plasmid and explain its significance for bacteria and recombinant technology.

Go to http://www.microbiologyprocedure.com/microbial-genetics/gene-cloning-procedure.htm or

http://faculty.plattsburgh.edu/donald.slish/Transformation.html

Explain what a plasmid is and why it is so vital for recombinant technology.

Extra – what other vectors are there besides plasmids?

2 - Describe how genomic and plasmid DNA can be isolated from cells.

Go to http://www.iupui.edu/~wellsctr/MMIA/isolating_dna/dna_isolation_rev.swf

Watch the animation. In a paragraph or two, in your own words,

describe the steps and their reasons in the DNA isolation process.

3 - Explain how the creation of sticky ends by restriction

enzymes is useful in producing a recombinant DNA molecule.

Look at the picture above, in your own words, explain why sticky ends are necessary when splicing together a recombinant DNA molecule.

4 - Explain each of the steps taken to isolate a gene of interest and insert it into bacterial DNA to produce the desired protein.

• Watch the tutorial http://www.sumanasinc.com/webcontent/animations/content/plasmidcloning.html

• Create step by step instructions, assume you are creating these instructions for a lab manual, explaining what must be done in each step and why, to isolate a specific gene ( we are assuming it is the gene to make you orange and blue!) and insert it into an E.Coli cell.

5 - Identify ways to determine if genetic transformation has been successful.

• Using our notes (PPT), give threethree protocols for markers of successful transformation in an E. coli.

• How can we tell our transformation occurred?

8 - Relate bacterial transformation to real life applications used by society.

• Speak to other students in the class, each person did a brochure on a real life application of bacterial transformation.

• Give one example of real life applications in…..1. Agriculture – plants 2. Agriculture – animals3. Medicine – production of medicines4. Medicine – gene therapy5. Transgenic organisms for research purposes
