무균 의약품 제조
원칙(Principle)
The manufacture of sterile products is subject to special requirements in order to
minimize risks of microbiological contamination, and of particulate and pyrogen
contamination. Much depends on the skill, training and attitudes of the personnel
involved. Quality Assurance is particularly important, and this type of manufacture
must strictly follow carefully established and validated methods of preparation and
procedure. Sole reliance for sterility or other quality aspects must not be placed on
any terminal process or finished product test.
무균 제품 제조 시에는 미생물, 미립자, 발열성 물질 오염을 최소화 하기 위해 특별한
기준이 필요하다. 많은 부분이 관련 작업자의 기술, 훈련, 기본 자세에 달려 있다. 품질
보증 체계(Quality Assurance)가 특히 중요하며, 이런 종류의 제조 작업 시에는
체계적으로 정립되고 밸리데이션된 절차와 방법을 정확하게 준수해야 한다. 최종 제품의
무균 상태나 여타 품질 측면을 최종 공정이나 최종 제품 시험에만 의존해서는 안 된다.

This guidance does not lay down detailed methods for determining the
microbiological and particulate cleanliness of air, surfaces etc. Reference should be
made to other documents such as the EN/ISO Standards.
이 가이드라인은 공기, 표면 등의 미생물과 미립자 청정도를 결정짓는 구체적인 방법을
제시하지 않는다. 이에 대한 사항은 EN/ISO 표준 같은 다른 문서를 참고한다.
공통(General)
1. The manufacture of sterile products should be carried out in clean areas
entry to which should be through airlocks for personnel and/or for
equipment and materials. Clean areas should be maintained to an
appropriate cleanliness standard and supplied with air which has passed
through filters of an appropriate efficiency.

2. The various operations of component preparation, product preparation and
filling should be carried out in separate areas within the clean area.
Manufacturing operations are divided into two categories; firstly those
where the product is terminally sterilised, and secondly those which are
conducted aseptically at some or all stages.
원자재 준비, 제품 조제, 충전 등의 작업은 청정 지역 내의 별도 지역(separate
areas)에서 실시해야 한다. 제조 작업은 제품을 사후 멸균 하는 경우와 제조
공정 전부 또는 일부 단계를 무균적으로 실시하는 경우 등 두 종류로 구분할 수
있다.
3. Clean areas for the manufacture of sterile products are classified according
to the required characteristics of the environment. Each manufacturing
operation requires an appropriate environmental cleanliness level in the
operational state in order to minimise the risks of particulate or microbial
contamination of the product or materials being handled.
필요 환경 특성을 바탕으로 무균 제품 제조를 위한 청정 지역을 분류한다. 각각의
제조 작업마다 취급 대상 제품 또는 물품의 미생물 또는 미립자 오염 위험을
최소화하기 위해, 작업 상태(operational state)에서 적정 청정도 수준을
유지해야 한다.
In order to meet “in operation” conditions these areas should be designed
to reach certain specified air-cleanliness levels in the “at rest” occupancy
state. The “at-rest” state is the condition where the installation is installed
and operating, complete with production equipment but with no operating
personnel present. The “in operation” state is the condition where the
installation is functioning in the defined operating mode with the specified
number of personnel working.
작업 상태(in operation condition)의 청정도 기준을 만족시키기 위해서는, 우선
휴지 상태(at rest condition)에서 지정 청정도 기준에 맞도록 작업 지역을
설계해야 한다. 휴지 상태(at rest state)라 함은 생산 설비의 설치 등 모든
설치가 완료되고 작업자 없이 설비가 작동 중인 상태를 의미한다. 작업 상태(in

operation state)라 함은 지정 작업자가 있는 상태에서 정해진 바에 따라 작업이
진행 중인 상태를 말한다.
The “in operation” and “at rest” states should be defined for each clean
room or suite of clean rooms.
각각의 청정 작업실 또는 청정 작업실 그룹별로 "작업 상태" 및 "휴지 상태"의
기준을 규정해야 한다.
For the manufacture of sterile medicinal products 4 grades can be
distinguished.
무균 의약품 제조 작업과 관련하여, 일반적으로 4개 청정도 등급이 있다.
Grade A: The local zone for high risk operations, e.g. filling zone, stopper
bowls, open ampoules and vials, making aseptic connections. Normally
such conditions are provided by a laminar air flow work station. Laminar
air flow systems should provide a homogeneous air speed in a range of
0.36 – 0.54 m/s (guidance value) at the working position in open clean
room applications. The maintenance of laminarity should be demonstrated
and validated.
A 등급: 충전 지역, 마개 보관통, 노출 상태의 앰플과 바이알, 무균 연결 조작
같이 위험성이 높은 작업이 진행되는 곳. 일반적으로 층류 작업대(laminar air
flow work station)를 구비하여 이런 조건을 확보한다. 층류 시스템은 개방 청정
작업실 상황에서 작업 부위에 0.36 - 0.54 m/s(참고치)의 균질한 풍속을
제공해야 한다. 층류 상태의 유지를 증명하고 밸리데이션해야 한다.
A uni-directional air flow and lower velocities may be used in closed
isolators and glove boxes.
폐쇄형 아이솔레이터 및 글로브박스에서는 단일 방향 공기 흐름을 확보하면서
보다 느린 풍속 상태로 작업할 수도 있다.
Grade B: For aseptic preparation and filling, this is the background
environment for the grade A zone.
B 등급: 무균 준비 및 충전 작업을 하는 경우에, A 등급 지역의 배경 환경에
해당된다.

4. Clean rooms and clean air devices should be classified in accordance with
EN ISO 14644-1. Classification should be clearly differentiated from
operational process environmental monitoring. The maximum permitted
airborne particle concentration for each grade is given in the following
table.
청정실 및 청정 공기 장치의 등급을 EN ISO 14644-1에 따라 분류한다. 작업
상태의 공정 환경 모니터와 등급 분류를 명확히 구분해야 한다. 등급별 최대 허용
부유 미립자 농도는 다음 표와 같다.

5. For classification purposes in Grade A zones, a minimum sample volume of
1 m3 should be taken per sample location. For Grade A the airborne
particle classification is ISO 4.8 dictated by the limit for particles ≥5.0 μm.
For Grade B (at rest) the airborne particle classification is ISO for both
considered particle sizes. For Grade C (at rest & in operation) the airborne
particle classification is ISO 7 and ISO 8 respectively. For Grade D (at rest)
the airborne particle classification is ISO 8. For classification purposes
EN/ISO 14644-1 methodology defines both the minimum number of
sample locations and the sample size based on the class limit of the

largest considered particle size and the method of evaluation of the data
collected.
A 등급 지역의 청정도 분류 시에 검체 채취 위치별로 최소 1 m3의 검체를
취해야 한다. A 등급의 부유 미립자 기준은 ≥5.0 μm의 미립자 한도 기준을
적용한 ISO 4.8이다. B 등급(휴지 상태)인 경우에는 부유 미립자 기준이 두 가지
미립자 크기 모두에 대하여 ISO 5를 적용한다. C 등급(휴지 상태 & 작업 상태)인
경우에는 부유 미립자 기준이 각각 ISO 7과 ISO 8이다. 등급 분류 목적에서의
고려 대상 최대 입자 크기의 기준에 근거한 검체량과 최소 검체 채취 위치 수,
그리고 수집한 데이터의 평가 방법이 EN/ISO 14644-1에 제시되어 있다.
6. Portable particle counters with a short length of sample tubing should be
used for classification purposes because of the relatively higher rate of
precipitation of particles ≥5.0 μm in remote sampling systems with long
lengths of tubing. Isokinetic sample heads shall be used in unidirectional
airflow systems.
등급 분류 시에는 검체 튜빙의 길이가 짧은 휴대용 미립자 계수 장치를 사용한다.
긴 튜빙을 구비하여 먼 거리에서 검체를 취하는 시스템인 경우에는 ≥5.0 μm
입자의 침전율이 상대적으로 높아지기 때문이다. 단일 방향 기류 시스템에서는
등속성 샘플 헤드를 사용한다.
7. “In operation” classification may be demonstrated during normal
operations, simulated operations or during media fills as worst-case
simulation is required for this. EN ISO 14644-2 provides information on
testing to demonstrate continued compliance with the assigned cleanliness
classifications.
"작업 상태" 등급은 정상 작업, 시뮬레이션 작업, 또는 메디아필 과정 중에
증명할 수 있다. 이 경우에는 "최악의 경우" 시뮬레이션이 요구되기 때문이다.
지정 청정도 기준의 지속적 준수를 증명하는 테스트에 관한 정보가 EN ISO
14644-2에 기술되어 있다.

8. Clean rooms and clean air devices should be routinely monitored in
operation and the monitoring locations based on a formal risk analysis

study and the results obtained during the classification of rooms and/or
clean air devices.
청정실과 청정 공기 장치를 작업 상태에서 모니터하며, 작업실 및/또는 청정 공기
장치의 등급 분류 과정에서 획득한 결과와 공식 리스크 분석 조사 결과에
근거하여 모니터 위치를 정한다.
9. For Grade A zones, particle monitoring should be undertaken for the full
duration of critical processing, including equipment assembly, except
where justified by contaminants in the process that would damage the
particle counter or present a hazard, e.g. live organisms and radiological
hazards. In such cases monitoring during routine equipment set up
operations should be undertaken prior to exposure to the risk. Monitoring
during simulated operations should also be performed. The Grade A zone
should be monitored at such a frequency and with suitable sample size
that all interventions, transient events and any system deterioration would
be captured and alarms triggered if alert limits are exceeded. It is
accepted that it may not always be possible to demonstrate low levels of
≥5.0 μm particles at the point of fill when filling is in progress, due to the
generation of particles or droplets from the product itself.
A 등급 지역은 설비 조립을 포함하여 핵심 가공 기간 전체에 걸쳐 미립자
모니터를 실시한다. 다만 미립자 계수 장치를 손상시키거나 위험을 발생시킬 수
있는 공정 중의 오염물(예, 살아 있는 생물체와 방사학적 위험)을 감안해
타당성이 있는 경우에는 예외를 인정한다. 이런 경우에는 정규 설비 셋업 작업
시의 모니터를 리스크 노출 이전에 실시해야 한다. 시뮬레이션 작업 도중의
모니터도 실시해야 한다. 모든 개입 상황, 일회적 이벤트, 시스템 상태 저하를
포착하고 경계 기준을 초과하면 경보가 발생될 수 있는 정도의 빈도로 적합한
검체량을 채취하여 A 등급 지역 모니터를 실시한다. 제품 자체에서 발생하는
미립자나 물방울 때문에 충전 작업이 진행 중인 상태에서 충전 지점의 ≥5.0 μm
미립자가 낮은 수준임을 증명하기가 항상 가능하지 않을 수도 있음을 인정한다.
10. It is recommended that a similar system be used for Grade B zones
although the sample frequency may be decreased. The importance of the
particle monitoring system should be determined by the effectiveness of
the segregation between the adjacent Grade A and B zones. The Grade B

zone should be monitored at such a frequency and with suitable sample
size that changes in levels of contamination and any system deterioration
would be captured and alarms triggered if alert limits are exceeded.
검체 채취 빈도를 감소시킬 수는 있지만, B 등급 지역도 유사한 방식으로 모니터
할 것을 권고한다. 인접한 A 등급 지역과 B 등급 지역 사이 분리의 효과를
감안하여 미립자 모니터 시스템의 중요도를 결정한다. 시스템 상태 저하와 오염
수준의 변화를 포착하고 경계 기준을 초과하면 경보가 발생될 수 있는 정도의
빈도로 적합한 검체량을 채취하여 B 등급 지역의 모니터를 실시한다.
11. Airborne particle monitoring systems may consist of independent particle
counters; a network of sequentially accessed sampling points connected by
manifold to a single particle counter; or a combination of the two. The
system selected must be appropriate for the particle size considered.
Where remote sampling systems are used, the length of tubing and the
radii of any bends in the tubing must be considered in the context of
particle losses in the tubing. The selection of the monitoring system should
take account of any risk presented by the materials used in the
manufacturing operation, for example those involving live organisms or
radiopharmaceuticals.
부유 미립자 모니터 시스템은 독립적인 미립자 계수 장치, 하나의 미립자 계수
장치가 있고 매니폴드를 통해 순차적으로 여러 검체 채취 포인트가 연결되는
네트워크 시스템, 또는 이 둘의 조합으로 구성될 수 있다. 모니터 대상 미립자
크기에 적절한 시스템을 선정한다. 원격 검체 채취 시스템을 사용한다면,
미립자가 튜빙에 남아 정확히 측정되지 않을 가능성이 있으므로, 튜빙의 길이와
튜빙 가운데 구부러진 부분의 반경을 검토해야 한다. 제조 작업에 사용되는
물품에 의한 리스크, 예를 들어 살아 있는 생물체나 방사성 의약품 관련 리스크를
고려하여 모니터 시스템을 선정한다.
12. The sample sizes taken for monitoring purposes using automated systems
will usually be a function of the sampling rate of the system used. It is not
necessary for the sample volume to be the same as that used for formal
classification of clean rooms and clean air devices.
자동화 시스템을 이용하여 모니터 목적으로 채취하는 검체량은 일반적으로 그
시스템의 검체 채취 속도를 감안하여 정한다. 청정실 및 청정 공기 장치의 공식

등급 분류 시에 취했던 검체량과 모니터 목적의 검체량이 반드시 같을 필요는
없다.
13. In Grade A and B zones, the monitoring of the ≥5.0 μm particle
concentration count takes on a particular significance as it is an important
diagnostic tool for early detection of failure. The occasional indication of
≥5.0 μm particle counts may be false counts due to electronic noise, stray
light, coincidence, etc. However consecutive or regular counting of low
levels is an indicator of a possible contamination event and should be
investigated. Such events may indicate early failure of the HVAC system,
filling equipment failure or may also be diagnostic of poor practices during
machine set-up and routine operation.
A 등급과 B 등급 지역의 경우에 ≥5.0 μm 미립자 모니터는 문제점을 조기에
파악할 수 있는 중요한 진단 도구이므로 특별한 의미를 갖는다. 전자 노이즈,
미광, 계수 손실 등으로 인하여 ≥5.0 μm 미립자의 수가 이따금 잘못된 수치로
나타날 수 있다. 하지만 낮은 수준이 연속적으로나 주기적으로 발생한다면, 이는
오염 징후이므로 조사를 실시해야 한다. HVAC 시스템의 이상이나 충전 설비
이상을 조기에 알려 주는 징후일 수 있으며, 또는 기계 셋업과 정규 작업 과정의
잘못된 행위를 의미할 수도 있다.
14. The particle limits given in the table for the “at rest” state should be
achieved after a short “clean up” period of 15-20 minutes (guidance value)
in an unmanned state after completion of operations.
표의 "휴지 상태" 미립자 기준은 작업 완료 이후 사람이 없는 상태에서 15-
20분(참고치) 동안의 짧은 "클린업" 기간 이후에 달성되어야 한다.
15. The monitoring of Grade C and D areas in operation should be performed
in accordance with the principles of quality risk management. The
requirements and alert/action limits will depend on the nature of the
operations carried out, but the recommended “clean up period” should be
attained.
작업 상태의 C 등급과 D 등급 지역 모니터는 품질 리스크 관리 원칙에 따라
실시한다. 수행 작업의 특성을 감안하여 기준과 경계/조치 기준을 정하지만, 권장
"클린업" 기간이 달성 되어야 한다.

16. Other characteristics such as temperature and relative humidity depend on
the product and nature of the operations carried out. These parameters
should not interfere with the defined cleanliness standard.
온도나 상대 습도 같은 다른 특성 요소는 수행 작업의 특성과 제품을 감안하여
정한다. 이들 변수로 인해 지정 청정도 기준이 훼손되어서는 안 된다.
17. Examples of operations to be carried out in the various grades are given in
the table below (see also paragraphs 28 to 35):

18. Where aseptic operations are performed monitoring should be frequent
using methods such as settle plates, volumetric air and surface sampling
(e.g. swabs and contact plates). Sampling methods used in operation
should not interfere with zone protection. Results from monitoring should
be considered when reviewing batch documentation for finished product

release. Surfaces and personnel should be monitored after critical
operations. Additional microbiological monitoring is also required outside
production operations, e.g. after validation of systems, cleaning and
sanitisation.
무균 작업을 하는 곳에서는 낙하균(settle plate), 부유 미생물(volumetric air),
표면균(surface sampling; swabs & contact plates) 검사 등 미생물 모니터를
자주 실시해야 한다. 작업 중의 미생물 모니터(검체 채취 방법) 때문에 해당
지역의 청정도가 훼손되어서는 안 된다. 최종 제품 출하 승인을 위한 배치 문서
검토 시에 환경 모니터 결과를 점검해야 한다. 중요 작업이 끝난 뒤에는 표면과
작업자를 모니터 해야 한다. 생산 작업 이외에도 위생 처리, 세척, 시스템
밸리데이션 이후에 추가적인 미생물 모니터가 필요하다.

20. Appropriate alert and action limits should be set for the results of
particulate and microbiological monitoring. If these limits are exceeded
operating procedures should prescribe corrective action.

기준(action limit)을 설정해야 한다. 환경 관리 결과가 이 기준을 초과하는
경우에 취할 조치 사항도 규정되어 있어야 한다.
아이솔레이터 기술(Isolator technology)
21. The utilisation of isolator technology to minimize human interventions in
processing areas may result in a significant decrease in the risk of
microbiological contamination of aseptically manufactured products from
the environment. There are many possible designs of isolators and transfer
devices. The isolator and the background environment should be designed
so that the required air quality for the respective zones can be realised.
Isolators are constructed of various materials more or less prone to
puncture and leakage. Transfer devices may vary from a single door to
double door designs to fully sealed systems incorporating sterilization
mechanisms.
제조 작업 지역에서 작업자의 개입을 최소화하기 위해 아이솔레이터
기술(isolator technology)을 활용하면, 무균적으로 제조된 제품이 제조 환경의
미생물에 의해 오염될 위험성을 크게 줄일 수 있다. 아이솔레이터와 이송
장치(transfer devices)는 다양한 방식으로 디자인할 수 있다. 아이솔레이터와
주변 환경은 해당 지역의 필요 공기 품질 수준을 달성할 수 있도록 설계해야 한다.
아이솔레이터는 구멍이 나거나 누출되는 정도가 다양한 여러 종류의 재질로
만든다. 이송 장치(transfer devices)의 디자인도 단일문에서 이중문에
이르기까지 다양하며, 멸균 메커니즘을 통합한 완전 봉 시스템도 있다.
22. The transfer of materials into and out of the unit is one of the greatest
potential sources of contamination. In general the area inside the isolator
is the local zone for high risk manipulations, although it is recognised that
laminar air flow may not exist in the working zone of all such devices.
아이솔레이터 장치에 물품을 반입하거나 장치에서 물품을 반출할 때 오염이
발생할 가능성이 매우 크다. 모든 아이솔레이터 장치의 작업 지역에 층류가
존재하지 않을 수도 있음이 인정되고 있지만, 일반적으로 아이솔레이터 내부
지역은 고위험 조작을 위한 국소 지역이다.

23. The air classification required for the background environment depends on
the design of the isolator and its application. It should be controlled and
for aseptic processing it should be at least grade D.

24. Isolators should be introduced only after appropriate validation. Validation
should take into account all critical factors of isolator technology, for
example the quality of the air inside and outside (background) the isolator,
sanitisation of the isolator, the transfer process and isolator integrity.
적절한 밸리데이션을 거친 다음에 아이솔레이터를 도입해야 한다. 밸리데이션을
할 때는 아이솔레이터 내부와 외부(주위)의 공기 품질, 아이솔레이터의 위생 처리,

25. Monitoring should be carried out routinely and should include frequent leak
testing of the isolator and glove/sleeve system.
모니터를 일상적으로 실시해야 하며 아이솔레이터와 작업자와의 연결
부위(glove/sleeve system)에 대해서는 누출 시험을 자주 실시해야 한다.
BFS(blow/fill/seal) 기술(Blow/fill/seal technology)
26. Blow/fill/seal units are purpose built machines in which, in one continuous
operation, containers are formed from a thermoplastic granulate, filled and
then sealed, all by the one automatic machine. Blow/fill/seal equipment
used for aseptic production which is fitted with an effective grade A air
shower may be installed in at least a grade C environment, provided that
grade A/B clothing is used. The environment should comply with the viable
and non viable limits at rest and the viable limit only when in operation.
Blow/fill/seal equipment used for the production of products which are
terminally sterilised should be installed in at least a grade D environment.

27. Because of this special technology particular attention should be paid to, at

28. Preparation of components and most products should be done in at least a
grade D environment in order to give low risk of microbial and particulate
contamination, suitable for filtration and sterilisation. Where the product is
at a high or unusual risk of microbial contamination, (for example, because
the product actively supports microbial growth or must be held for a long
period before sterilisation or is necessarily processed not mainly in closed
vessels), then preparation should be carried out in a grade C environment.

29. Filling of products for terminal sterilisation should be carried out in at least
a grade C environment.
사후 멸균 제품의 충전 작업은 적어도 C 등급 환경에서 실시해야 한다.
30. Where the product is at unusual risk of contamination from the
environment, for example because the filling operation is slow or the
containers are wide-necked or are necessarily exposed for more than a few
seconds before sealing, the filling should be done in a grade A zone with at
least a grade C background. Preparation and filling of ointments, creams,
suspensions and emulsions should generally be carried out in a grade C
environment before terminal sterilisation.

31. Components after washing should be handled in at least a grade D
environment. Handling of sterile starting materials and components, unless
subjected to sterilisation or filtration through a micro-organism-retaining
filter later in the process, should be done in a grade A environment with
grade B background.
세척 이후의 원자재는 적어도 D 등급 환경에서 다루어야 한다. 무균 출발 물질과
원자재의 취급은, 이들이 이후 공정에서 멸균되거나 미생물 제거 효율이 있는
필터를 통해 여과를 거치지 않는 경우, A 등급 환경에서 실시해야 하며 주위
환경은 B 등급이어야 한다.

32. Preparation of solutions which are to be sterile filtered during the process
should be done in a grade C environment; if not filtered, the preparation of
materials and products should be done in a grade A environment with a
grade B background.

공정을 거치지 않는 경우에는 물품 및 제품의 조제 작업을 A 등급 환경에서

33. Handling and filling of aseptically prepared products should be done in a
grade A environment with a grade B background.
무균적으로 준비한 제품의 취급 및 충전은 A 등급 환경에서 해야 하며 주위
환경은 B 등급이어야 한다.
34. Prior to the completion of stoppering, transfer of partially closed containers,
as used in freeze drying should be done either in a grade A environment
with grade B background or in sealed transfer trays in a grade B
environment.
동결 건조 제품처럼 반타전된 제품을 완전 봉하기 전에 이송할 때는 A 등급
환경과 B 등급의 주위 환경에서 하거나, B 등급 환경에서 폐된 이송 트레이에
담아 이송해야 한다.
35. Preparation and filling of sterile ointments, creams, suspensions and
emulsions should be done in a grade A environment, with a grade B
background, when the product is exposed and is not subsequently filtered.
제품이 노출되고 이후에 여과를 거치지 않는다면, 무균 연고제, 크림제, 현탁제,
유제의 조제 및 충전은 A 등급 환경에서 실시하며 주위 환경은 B 등급이어야
한다.
작업자(Personnel)
36. Only the minimum number of personnel required should be present in
clean areas; this is particularly important during aseptic processing.
Inspections and controls should be conducted outside the clean areas as
far as possible.

37. All personnel (including those concerned with cleaning and maintenance)
employed in such areas should receive regular training in disciplines
relevant to the correct manufacture of sterile products. This training should
include reference to hygiene and to the basic elements of microbiology.
When outside staff who have not received such training (e.g. building or
maintenance contractors) need to be brought in, particular care should be
taken over their instruction and supervision.
청정 지역내 작업자(청소 및 유지관리 업무 관련 작업자 포함)에 대하여 무균
제품의 올바른 제조와 관련된 사항을 정기적으로 훈련시켜야 한다. 위생 및
미생물학 기본 개념에 관한 사항도 훈련 내용에 포함시킨다. 이런 훈련을 받지
않은 외부인(예, 건물 관리 또는 유지 관리 위탁 업체)이 작업장 안으로 들어갈
필요가 있는 경우, 작업장 안에서 준수해야 할 사항을 주지시키고 감독하는 등
특별한 주의 조치를 취해야 한다.
38. Staff who have been engaged in the processing of animal tissue materials
or of cultures of micro-organisms other than those used in the current
manufacturing process should not enter sterile-product areas unless
rigorous and clearly defined entry procedures have been followed.
현 제조 공정에 사용되는 것 이외의 다른 동물 세포나 미생물 배양 작업을 수행한
작업자는, 엄격하고 명확히 규정된 출입 절차를 준수하지 않으면 무균 제품 작업
지역에 들어갈 수 없다.
39. High standards of personal hygiene and cleanliness are essential.
Personnel involved in the manufacture of sterile preparations should be
instructed to report any condition which may cause the shedding of
abnormal numbers or types of contaminants; periodic health checks for
such conditions are desirable. Actions to be taken about personnel who
could be introducing undue microbiological hazard should be decided by a
designated competent person.

40. Wristwatches, make-up and jewellery should not be worn in clean areas.
손목 시계, 화장, 장신구를 착용하고 청정 지역에 들어가서는 안 된다.
41. Changing and washing should follow a written procedure designed to
minimize contamination of clean area clothing or carry-through of
contaminants to the clean areas.
복장이나 물품 반입에 의해 청정 지역이 오염되는 것을 최소화 할 수 있도록 절차
문서에 따라 갱의와 세척을 실시해야 한다.
42. The clothing and its quality should be appropriate for the process and the
grade of the working area. It should be worn in such a way as to protect
the product from contamination.
작업복과 작업복의 품질은 작업 지역의 환경 등급과 해당 공정에 적절해야 한다.
작업복은 제품이 오염되지 않게 하는 방식으로 착용해야 한다.
43. The description of clothing required for each grade is given below:
청정 등급별로 필요한 작업복은 다음과 같다.
• Grade D: Hair and, where relevant, beard should be covered. A
general protective suit and appropriate shoes or overshoes should be
worn. Appropriate measures should be taken to avoid any
contamination coming from outside the clean area.
D 등급: 머리카락과 해당되는 경우에 턱수염을 가려야 한다. 일반 보호
복장과 적절한 신발 또는 덧신을 착용해야 한다. 청정 지역 외부에서
들어오는 오염물질을 방지하기 위해 적절한 조치가 있어야 한다.
• Grade C: Hair and where relevant beard and moustache should be
covered. A single or two-piece trouser suit, gathered at the wrists
and with high neck and appropriate shoes or overshoes should be

worn. They should shed virtually no fibres or particulate matter.
C 등급: 머리카락과 해당되는 경우에 턱수염 및 콧수염을 가려야 한다.
적당한 신발이나 덧신, 그리고 목 위로 올라오고 손목 부위에서 묶을 수
있는 원피스나 투피스형 작업복을 착용해야 한다. 이들은 섬유질이나
미립자를 방출해서는 안 된다.
• Grade A/B: Headgear should totally enclose hair and, where relevant,
beard and moustache; it should be tucked into the neck of the suit; a
face mask should be worn to prevent the shedding of droplets.
Appropriate sterilised, non-powdered rubber or plastic gloves and
sterilised or disinfected footwear should be worn. Trouser-legs should
be tucked inside the footwear and garment sleeves into the gloves.
The protective clothing should shed virtually no fibres or particulate
matter and retain particles shed by the body.
A/B 등급: 모자는 머리카락과 해당되는 경우에 턱수염 및 콧수염을 완전히
가려야 하며, 작업복의 목 부위로 어 넣어야 한다. 얼굴에는 비말이
날리지 않도록 마스크를 써야 한다. 적절하게 멸균된 비분말형(non-
powdered) 고무 또는 플라스틱 장갑과 멸균 또는 소독된 신발을 착용한다.
바지 하단은 신발 안으로 쑤셔 넣고 소매는 장갑 속으로 넣는다. 이런 보호
복장은 섬유질이나 미립자를 방출하지 않아야 하며, 몸에서 떨어지는
미립자가 유포되지 않도록 해야 한다.
44. Outdoor clothing should not be brought into changing rooms leading to
grade B and C rooms. For every worker in a grade A/B area, clean sterile
(sterilised or adequately sanitised) protective garments should be provided
at each work session. Gloves should be regularly disinfected during
operations. Masks and gloves should be changed at least for every working
session.
일반 복장을 B 및 C 등급 작업실로 이어지는 갱의실에 반입해서는 안 된다. A와
B 등급 지역에서 일하는 모든 작업자에게 작업 기간마다 청정 무균(멸균 또는
적절하게 위생 처리한) 보호 복장을 제공해야 한다. 작업 중에는 주기적으로
장갑을 소독한다. 마스크와 장갑은 적어도 매 작업 시마다 교체해야 한다.
45. Clean area clothing should be cleaned and handled in such a way that it

does not gather additional contaminants which can later be shed. These
operations should follow written procedures. Separate laundry facilities for
such clothing are desirable. Inappropriate treatment of clothing will
damage fibres and may increase the risk of shedding of particles.
청정 지역 복장을 세척하고 취급할 때는, 오염물이 작업복에 붙어 있다가 나중에
떨어지는 일이 생기지 않도록 해야 한다. 세척과 취급 작업은 절차 문서에 따라
실시해야 한다. 별도의 세탁 시설을 갖추는 것이 바람직하다. 작업복을
부적절하게 처리하면 섬유에 손상을 주어 입자 방출 위험성이 커진다.
시설(Premises)
46. In clean areas, all exposed surfaces should be smooth, impervious and
unbroken in order to minimize the shedding or accumulation of particles or
micro-organisms and to permit the repeated application of cleaning agents,
and disinfectants where used.
청정 지역의 모든 노출 표면은 미생물이나 미립자의 방출 또는 축적을 최소화하고
반복적으로 세척제와 소독제를 사용할 수 있도록, 매끄럽고 불침투성이며
파손되지 않아야 한다.
47. To reduce accumulation of dust and to facilitate cleaning there should be
no uncleanable recesses and a minimum of projecting ledges, shelves,
cupboards and equipment. Doors should be designed to avoid those
uncleanable recesses; sliding doors may be undesirable for this reason.
먼지가 쌓이지 않고 청소하기 쉽도록 하기 위해, 청소하기 힘든 후미진 부위가
없도록 하고 돌출부와 선반, 컵보드, 설비는 최소화 해야 한다. 출입문도
청소하기 힘든 후미진 부위가 없도록 설계해야 한다. 이런 점에서 미닫이 문은
바람직하지 않을 수 있다.
48. False ceilings should be sealed to prevent contamination from the space
above them.
보조 천장(false ceiling)은 그 위의 공간에서 오염물질이 떨어지는 것을 방지하기
위해 봉해야 한다.
49. Pipes and ducts and other utilities should be installed so that they do not

create recesses, unsealed openings and surfaces which are difficult to
clean.
파이프와 덕트, 여타 유틸리티 시설은 청소하기 힘든 표면, 봉되지 않은 개구부,
후미진 곳이 생기지 않도록 설치해야 한다.
50. Sinks and drains should be prohibited in grade A/B areas used for aseptic
manufacture. In other areas air breaks should be fitted between the
machine or sink and the drains. Floor drains in lower grade clean rooms
should be fitted with traps or water seals to prevent backflow.
무균 제조 작업을 하는 A/B 등급 지역에 싱크대와 배수 시설을 해서는 안 된다.
다른 지역에는 기계 또는 싱크대와 배수 시설 사이에 에어 브레이크(air break)를
설치해야 한다. 낮은 등급의 청정 작업실은 역류 방지를 위해 바닥 배수구에
트랩(trap) 또는 수 봉(water seal)을 갖추어야 한다.
51. Changing rooms should be designed as airlocks and used to provide
physical separation of the different stages of changing and so minimize
microbial and particulate contamination of protective clothing. They should
be flushed effectively with filtered air. The final stage of the changing room
should, in the at-rest state, be the same grade as the area into which it
leads. The use of separate changing rooms for entering and leaving clean
areas is sometimes desirable. In general hand washing facilities should be
provided only in the first stage of the changing rooms.
갱의실은 갱의 단계에 따라 물리적으로 구분하고 미생물과 미립자에 의한 보호
복장의 오염을 최소화할 수 있도록 사용해야 하며 에어록 방식으로 설계해야 한다.
갱의실에는 여과된 공기를 효과적으로 공급해야 한다. 갱의실의 마지막 단계는
갱의실을 통해 들어가는 작업 지역의 휴지 상태 청정 기준과 동일해야 한다.
때로는 청정 지역에 들어갈 때와 나올 때의 갱의실을 별도로 두는 것이
바람직하다. 일반적으로 수세 시설은 갱의실의 첫 단계에만 설치해야 한다.
52. Both airlock doors should not be opened simultaneously. An interlocking
system or a visual and/or audible warning system should be operated to
prevent the opening of more than one door at a time.
에어록이 설치된 문은 양쪽이 동시에 열리지 않아야 한다. 동시에 양쪽 문이 같이
열리는 것을 방지하기 위해, 시각 및/또는 청각적 경보 장치를 설치하거나 인터록

53. A filtered air supply should maintain a positive pressure and an air flow
relative to surrounding areas of a lower grade under all operational
conditions and should flush the area effectively. Adjacent rooms of
different grades should have a pressure differential of 10 - 15 pascals
(guidance values). Particular attention should be paid to the protection of
the zone of greatest risk, that is, the immediate environment to which a
product and cleaned components which contact the product are exposed.
The various recommendations regarding air supplies and pressure
differentials may need to be modified where it becomes necessary to
contain some materials, e.g. pathogenic, highly toxic, radioactive or live
viral or bacterial materials or products. Decontamination of facilities and
treatment of air leaving a clean area may be necessary for some
operations.
모든 작업 조건에서 주위의 낮은 등급 지역에 대해 양압 상태의 공기 흐름이
유지되도록 여과된 공기를 공급해야 하며, 해당 지역을 효과적으로 환기할 수
있어야 한다. 서로 다른 등급의 인접 작업실 간에는 10 ~ 15 파스칼(참고치)
정도의 차압을 유지해야 한다. 고위험 지역, 즉 제품과 접촉하는 깨끗한 자재와
제품이 노출되는 지역의 보호를 위해 특히 주의해야 한다. 병원성 제품, 독성
제품, 방사성 또는 생 바이러스나 세균성 물품 또는 제품의 봉쇄 필요성이 있는
경우, 급기와 차압에 대한 권고 사항이 다양하게 조정될 수 있다. 작업에 따라
시설의 정화 작업과 청정 지역에서 배출되는 공기의 처리가 필요할 수도 있다.
54. It should be demonstrated that air-flow patterns do not present a
contamination risk, e.g. care should be taken to ensure that air flows do
not distribute particles from a particle-generating person, operation or
machine to a zone of higher product risk.
공기 흐름 패턴으로 인한 오염 위험성이 없다는 점을 증명해야 한다. 즉 공기
흐름 때문에 작업자, 작업 또는 기계에서 발생한 미립자가 제품 위험성이 큰
지역으로 퍼지지 않게 주의해야 한다.
55. A warning system should be provided to indicate failure in the air supply.
Indicators of pressure differences should be fitted between areas where
these differences are important. These pressure differences should be

recorded regularly or otherwise documented.
급기에 이상이 발생했을 경우에 이를 알려 주는 경보 장치가 설치되어 있어야
한다. 작업실간 차압이 중요한 경우, 실간 차압을 보여 주는 인디케이터가
구비되어야 한다. 차압을 정기적으로 기록하거나 다른 방식으로 문서화한다.
설비(Equipment)
56. A conveyor belt should not pass through a partition between a grade A or
B area and a processing area of lower air cleanliness, unless the belt itself
is continually sterilised (e.g. in a sterilising tunnel).
컨베이어 벨트는 터널식 멸균기처럼 벨트 자체가 연속해서 멸균되는 경우가
아니면, A 또는 B 등급 작업 지역과 이보다 낮은 등급의 작업 지역 사이를
통과해서는 안 된다.
57. As far as practicable equipment, fittings and services should be designed
and installed so that operations, maintenance and repairs can be carried
out outside the clean area. If sterilisation is required, it should be carried
out, wherever possible, after complete reassembly.
가능하면 설비, 장치물, 서비스 시설은 운전, 유지 관리, 보수를 청정 지역
외부에서 수행할 수 있도록 설계하고 설치한다. 멸균이 필요한 경우에는 가능하면
완전히 재조립한 후에 실시한다.
58. When equipment maintenance has been carried out within the clean area,
the area should be cleaned, disinfected and/or sterilised where appropriate,
before processing recommences if the required standards of cleanliness
and/or asepsis have not been maintained during the work.
설비 유지관리를 청정 지역 안에서 하는 경우, 그 작업 도중에 청정도와 무균
상태 유지가 되지 않았다면 제조 작업을 다시 시작하기 전에 그 작업 지역을
적절하게 청소, 소독, 및/또는 멸균해야 한다.
59. Water treatment plants and distribution systems should be designed,
constructed and maintained so as to ensure a reliable source of water of
an appropriate quality. They should not be operated beyond their designed
capacity. Water for injections should be produced, stored and distributed in

a manner which prevents microbial growth, for example by constant
circulation at a temperature above 70°C.
수처리 시설과 분배 시스템은 적정 품질의 믿을 수 있는 용수를 공급할 수 있도록
설계, 시공, 유지관리되어야 한다. 설계 용량 범위 이상으로 가동해서는 안 된다.
주사용수(WFI)는 미생물 성장을 방지할 수 있는 방식으로, 예를 들어 70C
이상에서 연속 순환하는 식으로 생산, 보관, 공급해야 한다.
60. All equipment such as sterilisers, air handling and filtration systems, air
vent and gas filters, water treatment, generation, storage and distribution
systems should be subject to validation and planned maintenance; their
return to use should be approved.
멸균기, 공조 및 여과 시스템, 배기 및 가스 필터, 용수 처리, 생산, 보관, 분배
시스템 등 모든 설비를 밸리데이션하고 계획에 의거하여 유지관리해야 한다. 이들
설비를 다시 사용하는 것도 승인을 받아야 한다.

61. The sanitation of clean areas is particularly important. They should be
cleaned thoroughly in accordance with a written programme. Where
disinfectants are used, more than one type should be employed.
Monitoring should be undertaken regularly in order to detect the
development of resistant strains.
청정 지역의 위생 관리가 특히 중요하다. 청정 지역은 문서화된 프로그램에
의거하여 철저하게 청소해야 한다. 소독제를 사용하는 경우에는 한 가지 이상의
소독제를 채택해야 한다. 소독제 내성균 발생 여부를 확인하기 위해 주기적으로

62. Disinfectants and detergents should be monitored for microbial
contamination; dilutions should be kept in previously cleaned containers
and should only be stored for defined periods unless sterilised.
Disinfectants and detergents used in Grades A and B areas should be
sterile prior to use.
소독제와 세정제의 미생물 오염 여부를 모니터 해야 한다. 희석액은 미리 세척한

63. Fumigation of clean areas may be useful for reducing microbiological
contamination in inaccessible places.

공정(Processing)
64. Precautions to minimize contamination should be taken during all
processing stages including the stages before sterilisation.
멸균 이전 단계를 포함하여 모든 제조 공정에서 오염 최소화를 위해 주의가
필요하다.
65. Preparations of microbiological origin should not be made or filled in areas
used for the processing of other medicinal products; however, vaccines of
dead organisms or of bacterial extracts may be filled, after inactivation, in
the same premises as other sterile medicinal products.
미생물 유래 물질의 취급 작업을 다른 의약품 작업 지역에서 하거나 충전해서는
안 된다. 하지만 사백신이나 세균 추출물 백신은 불활화 단계 이후에 다른 무균
의약품과 동일 시설에서 충전할 수 있다.
66. Validation of aseptic processing should include a process simulation test
using a nutrient medium (media fill). Selection of the nutrient medium
should be made based on dosage form of the product and selectivity,
clarity, concentration and suitability for sterilisation of the nutrient medium.
무균 공정 밸리데이션을 할 때는 영양 배지를 활용한 공정 시뮬레이션 테스트를
실시해야 한다(메디아필). 영양 배지의 종류는 해당 제품의 제형과 영양 배지의
멸균 적합성, 농도, 투명도, 선택성을 토대로 결정한다.
67. The process simulation test should imitate as closely as possible the
routine aseptic manufacturing process and include all the critical
subsequent manufacturing steps. It should also take into account various
interventions known to occur during normal production as well as worst-
case situations.

68 Process simulation tests should be performed as initial validation with
three consecutive satisfactory simulation tests per shift and repeated at
defined intervals and after any significant modification to the HVAC-system,
equipment, process and number of shifts. Normally process simulation
tests should be repeated twice a year per shift and process.
초기 밸리데이션의 일환으로 작업조(shift)별로 공정 시뮬레이션 테스트를 3회
연속 실시해 적합한 결과가 나와야 하며, 이후에도 주기적으로 실시해야 한다.
또한 HVAC 시스템, 설비, 공정, 작업조 인원의 중요 변경이 있을 때도 실시해야
한다. 일반적으로 공정 시뮬레이션 테스트를 1년에 2회, 작업조 및 공정별로
실시한다.
69. The number of containers used for media fills should be sufficient to enable
a valid evaluation. For small batches, the number of containers for media
fills should at least equal the size of the product batch. The target should
be zero growth and the following should apply:
메디아필 충전 수량은 유효한 평가를 가능하게 할 정도로 충분해야 한다. 소규모
배치인 경우에는 메디아필 충전 수량이 적어도 실제 제품 배치 규모와 동일해야

• When filling fewer than 5000 units, no contaminated units should be
detected.

a) One (1) contaminated unit should result in an investigation,
including consideration of a repeat media fill;

70. For any run size, intermittent incidents of microbial contamination may be
indicative of low-level contamination that should be investigated.
Investigation of gross failures should include the potential impact on the
sterility assurance of batches manufactured since the last successful media
fill.
어떤 작업 규모이건, 미생물 오염이 간헐적으로 발생한다면, 이는 낮은 수준의
오염이 있다는 징후로 볼 수 있고 조사를 실시해야 한다. 조사를 할 때는
메디아필이 성공적으로 끝났던 시점 이후에 제조된 배치의 무균성 보장에 대한
영향 가능성도 조사해야 한다.
71. Care should be taken that any validation does not compromise the
processes.
밸리데이션으로 인해 해당 공정이 훼손되지 않도록 주의해야 한다.
72. Water sources, water treatment equipment and treated water should be
monitored regularly for chemical and biological contamination and, as
appropriate, for endotoxins. Records should be maintained of the results of
the monitoring and of any action taken.
수원, 수처리 설비, 처리수를 대상으로 화학적 및 생물학적 오염 시험을
정기적으로 실시해야 하며, 또한 적절한 경우에는 엔도톡신 시험을 실시해야 한다.

73. Activities in clean areas and especially when aseptic operations are in
progress should be kept to a minimum and movement of personnel should
be controlled and methodical, to avoid excessive shedding of particles and
organisms due to over-vigorous activity. The ambient temperature and
humidity should not be uncomfortably high because of the nature of the
garments worn.
청정 지역과 특히 무균 조작을 수행하는 지역에서의 활동을 최소화해야 하며,
과도한 활동으로 인하여 미립자와 미생물이 지나치게 방출되지 않도록, 작업자의

74. Microbiological contamination of starting materials should be minimal.
Specifications should include requirements for microbiological quality when
the need for this has been indicated by monitoring.
출발 물질의 미생물 오염을 최소화해야 한다. 모니터 결과에 의거하여 필요하다고
판단되는 경우, 출발물질 규격에 미생물학적 품질 기준을 포함시켜야 한다.
75. Containers and materials liable to generate fibres should be minimised in
clean areas.
섬유질을 방출하기 쉬운 용기와 물품을 청정 지역에서는 최소화해야 한다.
76. Where appropriate, measures should be taken to minimize the particulate
contamination of the end product.
적절한 경우에는 최종 제품의 미립자 오염을 최소화할 수 있는 대책을 구비해야
한다.
77. Components, containers and equipment should be handled after the final
cleaning process in such a way that they are not recontaminated.
원자재, 용기, 설비를 최종 세척한 이후에는 다시 오염되지 않도록 취급해야 한다.
78. The interval between the washing and drying and the sterilisation of
components, containers and equipment as well as between their
sterilisation and use should be minimized and subject to a time-limit

appropriate to the storage conditions.

79. The time between the start of the preparation of a solution and its
sterilisation or filtration through a micro-organism-retaining filter should
be minimised. There should be a set maximum permissible time for each
product that takes into account its composition and the prescribed method
of storage.
용액의 조제 시작부터 멸균 또는 제균 여과 작업까지의 시간을 최소화해야 한다.
제품 조성과 지정 보관 방법을 고려하여 제품별로 최대 허용 기간을 설정한다.
80. The bioburden should be monitored before sterilisation. There should be
working limits on contamination immediately before sterilisation, which are
related to the efficiency of the method to be used. Bioburden assay should
be performed on each batch for both aseptically filled product and
terminally sterilised products. Where overkill sterilisation parameters are
set for terminally sterilised products, bioburden might be monitored only
at suitable scheduled intervals. For parametric release systems, bioburden
assay should be performed on each batch and considered as an in-process
test. Where appropriate the level of endotoxins should be monitored. All
solutions, in particular large volume infusion fluids, should be passed
through a micro-organism-retaining filter, if possible sited immediately
before filling.
멸균 하기 전에 바이오버든을 모니터 해야 한다. 멸균 방법의 효율성을 감안하여
멸균 직전의 오염 기준이 설정되어 있어야 한다. 무균 충전 제품과 사후 멸균
제품 모두, 각 배치에 대하여 바이오버든 분석을 실시한다. 사후 멸균 제품에
대하여 오버킬 멸균 변수를 설정한 경우, 적합한 주기로만 바이오버든 모니터를
실시할 수 있다. 변수 기반 출하 승인 시스템인 경우, 배치별로 바이오버든
분석을 실시하고, 이를 공정 검사로 간주한다. 적절한 경우에는 엔도톡신 수준도
모니터 한다. 모든 용액은(특히 대용량 수액제의 경우) 제균 여과를 해야 하며,

81. Components, containers, equipment and any other article required in a
clean area where aseptic work takes place should be sterilised and passed
into the area through double-ended sterilisers sealed into the wall, or by a
procedure which achieves the same objective of not introducing
contamination. Non-combustible gases should be passed through micro-
organism retentive filters.
무균 작업을 하는 청정 지역에서 사용할 원자재, 용기, 설비 및 기타 물품은
멸균을 한 다음에 벽을 가로 질러 부착되어 있으며 이중문을 장착한 멸균기를
통해 반입하거나, 오염 물질의 유입 차단이라는 목표를 달성할 수 있는 방법으로
반입해야 한다. 불연성 가스는 제균 여과 필터를 통과해야 한다.
82. The efficacy of any new procedure should be validated, and the validation
verified at scheduled intervals based on performance history or when any
significant change is made in the process or equipment.
새로운 절차는 그 효율성을 밸리데이션해야 하며, 공정 또는 설비에 주요 변경이
발생하거나 성능 이력을 감안하여 일정 주기로 밸리데이션 상태를 확인해야 한다.
멸균(Sterilisation)
83. All sterilisation processes should be validated. Particular attention should
be given when the adopted sterilisation method is not described in the
current edition of the European Pharmacopoeia, or when it is used for a
product which is not a simple aqueous or oily solution. Where possible,
heat sterilisation is the method of choice. In any case, the sterilisation
process must be in accordance with the marketing and manufacturing
authorisations.
모든 멸균 공정을 밸리데이션해야 한다. 채택하고 있는 멸균 방법이 유럽 약전
최신판에 기술된 것이 아닌 경우, 또는 해당 제품이 단순한 액상이나 유성 액체가
아닌 경우에는 특히 주의해야 한다. 가능하면 열 멸균(heat sterilisation) 방법이
가장 적당하다. 멸균 공정은 판매 및 제조 허가 사항과 일치해야 한다.
84. Before any sterilisation process is adopted its suitability for the product
and its efficacy in achieving the desired sterilising conditions in all parts of
each type of load to be processed should be demonstrated by physical
measurements and by biological indicators where appropriate. The validity

of the process should be verified at scheduled intervals, at least annually,
and whenever significant modifications have been made to the equipment.
Records should be kept of the results.
멸균 공정을 도입하기 전에 그 멸균 공정이 해당 제품에 적합하며 멸균 적재
유형별로 모든 부분에서 기대한 만큼의 멸균 조건을 달성하는데 효과적이라는
점을 물리적 측정 방법과 적절한 경우에는 생물학적 지시물(biological
indicator)을 사용하여 증명해야 한다. 해당 공정의 유효성을 적어도 1년에
한번씩 주기적으로 확인해야 하며 설비의 중요 변경이 있을 때도 실시해야 한다.
그 결과에 대한 기록서도 유지해야 한다.
85. For effective sterilisation the whole of the material must be subjected to
the required treatment and the process should be designed to ensure that
this is achieved.
효과적인 멸균을 위해 물품 모두에 대하여 필요한 처리를 해야 하며, 그에 맞도록
공정을 설계해야 한다.
86. Validated loading patterns should be established for all sterilisation
processes.
모든 멸균 공정에 대하여 밸리데이션된 적재 패턴을 설정해야 한다.
87. Biological indicators should be considered as an additional method for
monitoring the sterilisation. They should be stored and used according to
the manufacturer’s instructions, and their quality checked by positive
controls. If biological indicators are used, strict precautions should be
taken to avoid transferring microbial contamination from them.
생물학적 지시물(BI)은 멸균 공정 모니터를 위한 부가적인 방법으로 생각해야 한
다. 제조원의 설명서에 따라 BI를 보관하고 사용하며, 양성 대조로 BI의 품질을
점검한다. BI를 사용하는 경우에는 이로 인해 미생물 오염이 일어나지 않도록 특
히 주의해야 한다.
88. There should be a clear means of differentiating products which have not
been sterilized from those which have. Each basket, tray or other carrier of
products or components should be clearly labelled with the material name,
its batch number and an indication of whether or not it has been sterilised.

Indicators such as autoclave tape may be used, where appropriate, to
indicate whether or not a batch (or sub-batch) has passed through a
sterilisation process, but they do not give a reliable indication that the lot
is, in fact, sterile.
멸균된 것과 멸균되지 않은 것을 구분할 수 있는 명확한 방법이 있어야 한다.
제품 또는 원자재를 담는 바구니, 트레이, 기타 운반 용구에 품명, 배치 번호,
멸균 여부를 명확하게 기재한 표시서를 부착한다. 배치(또는 서브배치)가 멸균
공정을 거쳤는지 표시하기 위해 적절한 경우에는 멸균 테이프 같은 지시물을
사용할 수도 있다. 하지만 이것은 그 로트가 실제로 무균 상태라는 신뢰할 수
있는 표시는 아니다.
89. Sterilisation records should be available for each sterilisation run. They
should be approved as part of the batch release procedure.
매 멸균 작업 마다 멸균 기록서가 있어야 한다. 배치 출하 승인 절차의 일부로
멸균 기록서를 승인해야 한다.
열에 의한 멸균(Sterilisation by heat)
90. Each heat sterilisation cycle should be recorded on a time/temperature
chart with a sufficiently large scale or by other appropriate equipment with
suitable accuracy and precision. The position of the temperature probes
used for controlling and/or recording should have been determined during
the validation, and where applicable also checked against a second
independent temperature probe located at the same position.
열멸균 사이클을 충분히 큰 스케일의 시간/온도 차트에 기록하거나, 적합한
정확도와 정 도를 지닌 적절한 장비를 사용해 기록해야 한다. 관리 또는 기록을

91. Chemical or biological indicators may also be used, but should not take the
place of physical measurements.

92. Sufficient time must be allowed for the whole of the load to reach the
required temperature before measurement of the sterilising time-period is
commenced. This time must be determined for each type of load to be
processed.
적재물 전체가 필요 온도 수준에 도달한 다음에 멸균 기간 측정을 시작하도록
충분한 시간을 두어야 한다. 적재 유형별로 이 시간을 결정해야 한다.
93. After the high temperature phase of a heat sterilisation cycle, precautions
should be taken against contamination of a sterilised load during cooling.
Any cooling fluid or gas in contact with the product should be sterilised
unless it can be shown that any leaking container would not be approved
for use.
열멸균 사이클 고온 단계 이후에는 냉각 기간 동안에 멸균된 적재물이 오염되지
않도록 주의해야 한다. 누출 용기는 사용을 승인하지 않는다는 점을 증명할 수
없다면, 제품과 접촉하는 냉각용 액체나 가스도 멸균해야 한다.
습열(Moist heat)
94. Both temperature and pressure should be used to monitor the process.
Control instrumentation should normally be independent of monitoring
instrumentation and recording charts. Where automated control and
monitoring systems are used for these applications they should be
validated to ensure that critical process requirements are met. System and
cycle faults should be registered by the system and observed by the
operator. The reading of the independent temperature indicator should be
routinely checked against the chart recorder during the sterilisation period.
For sterilisers fitted with a drain at the bottom of the chamber, it may also
be necessary to record the temperature at this position, throughout the
sterilization period. There should be frequent leak tests on the chamber
when a vacuum phase is part of the cycle.
온도와 압력으로 공정을 모니터 해야 한다. 제어 장치는 모니터 장치 및 기록
차트로부터 독립되어 있어야 한다. 자동 제어 및 모니터 시스템을 사용하는 경우,
핵심 공정 기준이 충족된다는 점을 밸리데이션을 통해 확인해야 한다. 시스템과
사이클 이상을 시스템 자체가 기록하고 작업자가 확인할 수 있어야 한다. 멸균

한다. 챔버 하단에 배수 장치가 갖추어진 멸균기인 경우, 전체 멸균 기간 동안 이
부분의 온도를 기록할 필요가 있다. 멸균 사이클 중에 진공 단계가 있으면,
챔버에 대하여 누출 시험(leak test)을 빈번하게 실시해야 한다.
95. The items to be sterilised, other than products in sealed containers, should
be wrapped in a material which allows removal of air and penetration of
steam but which prevents recontamination after sterilisation. All parts of
the load should be in contact with the sterilizing agent at the required
temperature for the required time.
봉 용기에 담긴 제품 이외의 다른 멸균 대상 물품은, 공기의 제거와 스팀의
침투가 용이하고 멸균 이후 재오염을 방지할 수 있는 재질로 포장해야 한다.
적재되는 모든 물품은 필요 시간 동안 필요 온도에서 멸균 인자(sterilizing
agent)와 접촉 상태를 유지해야 한다.
96. Care should be taken to ensure that steam used for sterilisation is of
suitable quality and does not contain additives at a level which could cause
contamination of product or equipment.
멸균에 사용되는 스팀은 적합한 품질이어야 하며 제품이나 설비를 오염시킬
수준의 첨가물이 함유되지 않도록 주의해야 한다.
건열(Dry heat)
97. The process used should include air circulation within the chamber and the
maintenance of a positive pressure to prevent the entry of non-sterile air.
Any air admitted should be passed through a HEPA filter. Where this
process is also intended to remove pyrogens, challenge tests using
endotoxins should be used as part of the validation.
이 멸균 방법을 사용하는 경우에는 챔버 내의 공기가 순환되어야 하며 비무균
공기의 유입을 방지하기 위해 양압을 유지해야 한다. 공기는 HEPA 필터를
통해서만 유입되어야 한다. 이 공정이 발열성 물질 제거를 목적으로 하는
경우에는 밸리데이션의 일부로 엔도톡신을 이용한 챌린지 시험을 실시해야 한다.

98. Radiation sterilisation is used mainly for the sterilisation of heat sensitive
materials and products. Many medicinal products and some packaging
materials are radiation-sensitive, so this method is permissible only when
the absence of deleterious effects on the product has been confirmed
experimentally. Ultraviolet irradiation is not normally an acceptable method
of sterilisation.
방사선 멸균은 열에 민감한 물품과 제품의 멸균에 주로 사용된다. 많은 의약품과
일부 포장 자재는 방사선에 민감하다. 그래서 방사선이 제품에 유해한 영향을 미
치지 않는다는 점이 실험적으로 증명되었을 경우에만 이 방법을 사용한다. 자외선
조사는 일반적으로 적절한 멸균 방법이 아니다.
99. During the sterilisation procedure the radiation dose should be measured.
For this purpose, dosimetry indicators which are independent of dose rate
should be used, giving a quantitative measurement of the dose received
by the product itself. Dosimeters should be inserted in the load in sufficient
number and close enough together to ensure that there is always a
dosimeter in the irradiator. Where plastic dosimeters are used they should
be used within the time-limit of their calibration. Dosimeter absorbances
should be read within a short period after exposure to radiation.
멸균 과정 중에 방사능 조사량을 측정해야 한다. 이를 위해 선량율(dose
rate)과는 별도의 선량계 인디케이터(dosimetry indicators)를 사용하여 제품
자체가 받은 선량을 양적으로 측정할 수 있어야 한다. 충분한 수의 선량계를
적재물에 삽입해야 하며, 충분히 서로 착하도록 하여 방사선 조사
장치(irradiator)에 항상 선량계가 있도록 해야 한다. 플라스틱 선량계를 쓰는
경우에는 이들은 정해진 교정 기한 내에서만 사용한다. 방사선 노출 이후 단시간
안에 선량계 흡광도를 읽어야 한다.
100. Biological indicators may be used as an additional control.
부가적으로 생물학적 지시물을 사용할 수도 있다.
101. Validation procedures should ensure that the effects of variations in
density of the packages are considered.

102. Materials handling procedures should prevent mix-up between irradiated
and non-irradiated materials. Radiation sensitive colour disks should also
be used on each package to differentiate between packages which have
been subjected to irradiation and those which have not.
물품 취급 시에는 방사선 처리된 것과 처리되지 않은 것 사이의 혼동을
방지하도록 해야 한다. 방사선 처리된 것과 처리되지 않은 것을 구분하기 위해 각
포장물에 방사선에 민감한 유색 디스크(radiation sensitive colour disks)를

104. This method should only be used when no other method is practicable.
During process validation it should be shown that there is no damaging
effect on the product and that the conditions and time allowed for
degassing are such as to reduce any residual gas and reaction products to
defined acceptable limits for the type of product or material.

105. Direct contact between gas and microbial cells is essential; precautions
should be taken to avoid the presence of organisms likely to be enclosed in
material such as crystals or dried protein. The nature and quantity of
packaging materials can significantly affect the process.

required for this should be balanced against the opposing need to minimize
the time before sterilisation.

107. Each sterilisation cycle should be monitored with suitable biological
indicators, using the appropriate number of test pieces distributed
throughout the load. The information so obtained should form part of the
batch record.
멸균 사이클마다 적합한 생물학적 지시물(biological indicator)로 모니터 하며,
적재물 전체에 걸쳐 적정 수의 지시물을 사용해야 한다. 이를 통해 얻은 정보를

108. For each sterilisation cycle, records should be made of the time taken to
complete the cycle, of the pressure, temperature and humidity within the
chamber during the process and of the gas concentration and of the total
amount of gas used. The pressure and temperature should be recorded
throughout the cycle on a chart. The record(s) should form part of the
batch record.
멸균 사이클마다 전체 멸균에 소요된 시간, 챔버 내의 압력, 온도, 습도, 그리고

109. After sterilisation, the load should be stored in a controlled manner under
ventilated conditions to allow residual gas and reaction products to reduce
to the defined level. This process should be validated.

110. Filtration alone is not considered sufficient when sterilisation in the final
container is possible. With regard to methods currently available, steam
sterilisation is to be preferred. If the product cannot be sterilised in the
final container, solutions or liquids can be filtered through a sterile filter of
nominal pore size of 0.22 micron (or less), or with at least equivalent
micro-organism retaining properties, into a previously sterilised container.
Such filters can remove most bacteria and moulds, but not all viruses or
mycoplasmas. Consideration should be given to complementing the
filtration process with some degree of heat treatment.
최종 용기에 담긴 상태로 멸균할 수 있는 경우, 여과 공정만으로는 충분하지 않은
것으로 간주된다. 현재 적용 가능한 방법으로는 스팀 멸균(steam sterilization)이

111. Due to the potential additional risks of the filtration method as compared
with other sterilization processes, a second filtration via a further sterilised
micro-organism retaining filter, immediately prior to filling, may be
advisable. The final sterile filtration should be carried out as close as
possible to the filling point.

known volume of bulk solution and the pressure difference to be used

across the filter should be determined during validation and any significant
differences from this during routine manufacturing should be noted and
investigated. Results of these checks should be included in the batch
record. The integrity of critical gas and air vent filters should be confirmed
after use. The integrity of other filters should be confirmed at appropriate
intervals.
제균 여과용 필터는 사용하기 전에 완전성(integrity)을 확인해야 하며, 사용
직후에도 적절한 방법(예, bubble point, diffusive flow, pressure hold test)으로

114. The same filter should not be used for more than one working day unless
such use has been validated.
동일 필터를 1 작업일 이상 사용해서는 안 되며, 그 이상 사용하는 경우에는

115. The filter should not affect the product by removal of ingredients from it or
by release of substances into it.

116. Partially stoppered freeze drying vials should be maintained under Grade A
conditions at all times until the stopper is fully inserted.

117. Containers should be closed by appropriately validated methods.
Containers closed by fusion, e.g. glass or plastic ampoules should be

118. The container closure system for aseptically filled vials is not fully integral
until the aluminium cap has been crimped into place on the stoppered vial.
Crimping of the cap should therefore be performed as soon as possible
after stopper insertion.
무균 충전 바이알의 용기 마개 시스템은 타전 바이알의 알루미늄 캡 작업이
완료될 때까지 완전한 상태가 아니다. 그러므로 타전 작업 이후에 가능한

119. As the equipment used to crimp vial caps can generate large quantities of
non-viable particulates, the equipment should be located at a separate
station equipped with adequate air extraction.
바이알 캐핑 작업용 설비는 다량의 미립자를 발생시킬 수 있으므로, 별도의
작업대에 이 설비를 위치시켜야 하고 적절한 공기 추출 장치를 구비해야 한다.
120. Vial capping can be undertaken as an aseptic process using sterilised caps
or as a clean process outside the aseptic core. Where this latter approach
is adopted, vials should be protected by Grade A conditions up to the point
of leaving the aseptic processing area, and thereafter stoppered vials
should be protected with a Grade A air supply until the cap has been
crimped.

required conditions and minimising direct human interventions into the
capping operation.

123. Containers sealed under vacuum should be tested for maintenance of that
vacuum after an appropriate, pre-determined period.
진공 상태에서 봉된 용기는 미리 설정한 적정 기간 이후에 진공 상태의 유지
여부를 시험해야 한다.
124. Filled containers of parenteral products should be inspected individually for
extraneous contamination or other defects. When inspection is done
visually, it should be done under suitable and controlled conditions of
illumination and background. Operators doing the inspection should pass
regular eye-sight checks, with spectacles if worn, and be allowed frequent
breaks from inspection. Where other methods of inspection are used, the
process should be validated and the performance of the equipment
checked at intervals. Results should be recorded.
주사제는 이물의 오염이나 기타 이상 사항 여부를 개별적으로 검사해야 한다.
육안으로 이물 검사를 할 경우에는 적합한 조명과 배경 조건을 유지한 상태에서

125. The sterility test applied to the finished product should only be regarded as
the last in a series of control measures by which sterility is assured. The
test should be validated for the product(s) concerned.
최종 제품에 대한 무균 시험은 제품의 무균성 보장을 위한 일련의 관리 조치
가운데 마지막 단계로만 간주해야 한다. 무균 시험은 해당 제품별로
밸리데이션해야 한다.
126. In those cases where parametric release has been authorised, special
attention should be paid to the validation and the monitoring of the entire
manufacturing process.
변수 기반 출하 승인 방식(parametric release)을 승인 받은 경우에는 전 제조
공정의 밸리데이션과 모니터에 특히 주의해야 한다.
127. Samples taken for sterility testing should be representative of the whole of
the batch, but should in particular include samples taken from parts of the
batch considered to be most at risk of contamination, e.g.:
