ApplicationPumping animal cell cultures

Gentle, cell-sensitive pump systems areoften required in cell cultivation.Ms Fan Sozzi-Guo and Prof. Dr.Graf-Hausner of Zurich TechnicalUniversity Winterthur, andDr. Joanne Laukart of ISMATEC SAstarted a biological study bycomparing different pump systems fortheir influence in pumping animal cellcultures. The experimental target was toguarantee a gentle cell delivery withremaining high cell viability and lowcell damage.

MethodCultivation of hybridoma cells inspinner bottles.

The cells are continuously pumpedthrough an external loop at a constantflow rate until identifiable cell death isobserved. Comparative parameters werethe number of pump rollers (2 or 3),spring-loaded vs. non-spring-loadedrollers and tubing bed / rollergeometry (convex vs. straight rollers).

Gentle Cell sensitive Pumping

ConclusionThanks to the 2 convex rollers and theconcave tube-bed the cells can escapethrough a gap towards the tubing wallbefore the roller totally closes thesqueezed tubing.

Comparisons to gear, piston andcentrifugal pumps proved that theseperistaltic pumps are the onlysuitable and sterilisable pumpsystem for gently pumping mediacontaining living cells.
