The 3 years of Multiannual Programme founded by Ministry of Agriculture and Rural Development Poland Project: Broadening of barley gene pool The resistance of modern barley cultivars ( Hordeum vulgare , L.) has a narrow genetic background. Powdery mildew , caused by Blumeria graminis f. sp. hordei, is the most ecomicaly important barley disease. Landraces originate from regions with traditional agriculture are rich but still low operated source of genetic diversity. Resistence to powdery mildew is the aim of numerous breeding and scientific programmes. http://bankgenow.edu.pl/en/ Phenotypic tests for powdery mildew resistance of 4 000 landraces Resistnt line selection F 2 mapping population Identification of molecular markers linked to resistant trait selected lines with broad spectrum resistance for powdery mildew: i) Bgh255-3-3 ii) Bgh569-3-3 iii) Bgh5617-1-1 iv) Bgh39408-3-5 Identifying new sources of resistance and new molecular markers linked to resistance genes opens new dimensions for breeders and research programmes resistant line × susceptible cultivar: i) Bgh255-3-3 × Manchuria ii) Bgh569-3-3 × Manchuria Fitopathological tests of F 2 plants and F 3 families . Numerous of PCR markers based on SSRs and DArTSeq oligos (Diversity Arrays Technology Pty. Ltd.) was used to verify linkage with powdery mildew resistance. Barley line Bgh255-3-3 genetic map Powdery mildew resistant gene is located on 2HS chromosome The nearest marker: DArT5258111 – 3.2cM distance: chr2H 3653981 to 3654025* *Phisical location for Ensembl Plants release 36 http://plants.ensembl.org Genetic map calculated in JoinMap 4.0 softwere (Kyazma B. V.) Bgh255-3-3 and Bgh569-3-3 response to diverse collection of powdery mildew pathotypes. Compared with lines’ set carried various resistant. Bgh255-3-3 and Bgh569- 3-3 lines are resistant to all of used pathotypes. Over 70.000 accesions in long term storage (2 nd among European Union) Over 20.000 unique items in Herbarium collecion Well equipped molecular lab Qualified staff with passion to biodiversity Scores: 0, 1, 2- resistant, 3, 4- susceptible, according to Mains’s and Dietz’s 5-level scale (1930).
