3. PROPOSAL SUMMARY
Inflammatory diseases are a complex and heterogeneous group of diseases,
which are a major cause of morbidity and mortality and an important socioeconomic
problem. Despite scientific and technological advances of recent decades, the current
treatment options for inflammatory diseases are still limited and in some cases,
inefficient. These facts are mainly related to limited understanding of the factors and
mechanisms underlying these diseases. Additionally, the lack of sensitive and specific
methods for the early and accurate diagnosis of each disease and their deviations further
complicates the choice of an effective treatment. Accordingly, the development of
translational research is imperative to integrate the basic and clinical scientific
knowledge and also identify new therapeutic targets and methods for diagnosis of
chronic inflammatory diseases. The Center for Research in Inflammatory Diseases
(CRID) was created with these goals. The researchers of the center came from various
biomedical fields, including basic (geneticists, molecular and cell biologists,
immunologists, pharmacologists, pathologists), and clinical (rheumatologists, clinical
immunologists, infectologists, dermatologists) researchers, as well as investigators on
the field computational medicinal chemistry and chemical synthesis. Thus, the
integrative researches that will be realized by the CRID will enable not only the
discovery of new mechanisms involved in inflammatory diseases, but also the synthesis
of new medicinal molecules and diagnostic markers.
The specific goal of CPDI will be to advance the understanding of
physiopathology of inflammatory diseases (infectious: Leishmaniasis and sepsis,
autoimmune: rheumatoid arthritis, multiple sclerosis, psoriasis, pemphigus, and
vascular atherosclerotic: atherosclerosis) in order to: 1) recognize and understand the
mechanisms involved, such as molecular (genetic and intracellular signaling),
immunological (innate and specific responses), pathological and pharmacological
(experimental models); 2) identify new biological targets to develop pharmacological
(synthetic and from natural source) and immune (antibody) therapies tools; 3) search for
possible diagnosis markers and prognostic clues; and 4) apply this new knowledge to
design and synthesize new molecules to treat inflammatory diseases. The new
molecules will be tested for their efficacy in experimental models and submitted to
concept tests in samples from human beings. In the case of potential therapeutical use,
we intend to seek for patents and perform stability, pharmacokinetic and pre-clinical
toxicology trials and coordinate possible early clinical trials. The stages of the
development of potential drugs will be performed in partnership with public and private
companies, which have already expressed interest in partnerships.
