Tissue-specific ChIP is a powerful method of elucidating

mechanisms by which cell microenvironment can affect gene

regulation. Conventional chromatin immunoprecipitation (ChIP)

protocols, including our popular Magna ChIP and EZ Magna ChIP

assay kits, work best using homogenous populations of cultured

cells. In contrast, the Magna ChIP G Tissue Kit is specifically

designed for tissue samples, which are more scientifically reliant,

but also more complex, heterogeneous mixtures of cells that are

less amenable to ChIP.

Magna ChIP G Tissue Kit Includes:

• Tools necessary to microdissect tissue sections

that are < 1 mm3 in size

• Tissue stabilization and lysis reagents

• Optimized reagents and protocols for chromatin

Chromatin from various mouse tissues was subjected to immunopre-cipitation with Anti-RNA Polymerase II, clone CTD4H8 (Catalogue No. 05-623B) and Anti-SP1 (Catalogue No. 07-645) using the Magna ChIP G Tissue kit. The IgG used for relative comparison was either Rabbit IgG (Catalogue No. 12-370) or Mouse IgG (Catalogue No. 12-371). Quantitative PCR was performed using primers specific for either the GAPDH promoter, Dhfr promoter, or an upstream region (negative con-trol). Data above is presented as fold relative enrichment to IgG.

Anti-RNA Polymerase II, clone CTD4H8 (Catalogue No. 05-623): 1 µg of mouse monoclonal affinity purified antibody immunoprecipitated with chromatin of various mouse tissue and qPCR assayed with primers specific for mouse GAPDH promoter: Forward: 5’ CAC CAT CCG GGT TCC TAT AAA TAC 3’, Reverse: 5’ CAG CAT CCC TAG ACC CGT ACA 3’

Anti-SP1 (Catalogue No. 07-645): 1 µg of mouse monoclonal affinity purified antibody immunoprecipitated from chromatin of various mouse tissue and qPCR assayed with primers specific for mouse Dhfr promoter: Forward: 5’ CAC GCC TCA ACC TGT GCG GGA 3’, Reverse: 5’ GCG GGG ATA AAA TCC TAC CAG CC 3’.
