Chromatographic Column Selection
There is a wide range of packing materials in HPLC, and the selection of them has great influence on the
separation performance, so it is very necessary to know of them for appropriate selection.
Silica and silica-based packings
A. Normal phase chromatography
The stationary phases for normal phase chromatography are usually silica and some polar bonded-phase
functional groups, such as Amino Propyl (NH2, APS), Propyl Nitrile (CN, CPS).
As the Si-OH in the surface of silica and other polar functional groups are highly polar, the sequence of
being separated is in line with their polarity in respective analytes, that is to say, the weaker its polarity,
the quicker being separated.
The mobile phases for normal phase chromatography is much weaker than stationary phases, such as
Hexane, Chloroform, and Methylene Chloride.
B. Reversed phase chromatography
The stationary phases for reversed phase chromatography are silica-based bonded phases with
non-polar functional groups on the surface of the silica.
The mobile phases for reversed phase chromatography are polar mixtures of water or buffered solvents
with methanol or acetonitrile.
The most polar analyte will be first dashed out among the sample, while the less polar ingredient will be
strongly retained.
Typical packings for reversed phase chromatography are C18 (ODS), C8 (MOS), C4 (Buty1), C6H5
(Phenyl).
Polymeric packings
Polymeric packings are mainly polystyrene-divinylbenzene or polymethacrylate,and etc,which can be
used from pH 1-14 as their leading advantage.
Comparing with silica-based C18 packing, these packings are more hydrolphobic; polymeric packings
with large pore are quite efficient to the separation of samples like protein.
The main disadvantage of current polymeric packings is the low column efficiency compared with sili-
ca-based packings.

Other inorganic packings
Other HPLC columns with inorganic packings have been commercialized, but they are limited for special
use because of unique property, for example, graphitic carbon is gradually becoming a member of re-
versed phase packings. Separation with this kind of packings is different from silica-based alkyl-bonded
phases, for the graphite surface is the retention base, you don’t need any other surface modifications.
Normally, this column packing has stronger retention than alkyl-bonded silica or porous polymeric pack-
ings. Graphitic carbon can separate some geometrical isomer, and it can’t be dissolved by the HPLC
mobile phase, so it can be used in any pH range and temperature. Alumina can also be used in HPLC,
first, it has strong rigidity and could be made into stable chromatographic column bed; second, it can be
used with the mobile phase at pH 12. But alumina hasn’t been widely applied for its application area is
limited by the strong interactions with basic compounds. The new-type zirconia-based packings can also
be used in HPLC, but the only commercializd packing is polymer-coated porous microspherical zirconia,
which exihits at the environment of pH 1-14 and the highest temperature 100℃. As zirconia-based
packings have been studied for recent years, facing experiment difficulties, so their important purposes
and advantages are in process.
How to Guarantee Good Performance and Lifetime?
◇Read columns user manual carefully
◇Use well packed columns
◇Reduce pressure pulsation to avoid mechanical and thermal impact
◇Make use of guard column and on-line filter
◇Wash columns with strong solvents often
◇Completely filter samples and mobile phases, avoid impurity atoms and strong reserved com-
ponents
◇Use stable stationary phases (C18 is the most stable stationary phase)
◇Use organic buffer solvents on the operation of pH ranging 6-8
◇The best use temperature for columns is lower than 40℃
◇Silica-based columns should keep their mobile phase pH ranging 2.0-8.0
◇Add 200ppm NaN3 in water mobile phases and buffer solvents
◇If there are buffer solvents in the mobile phases, pay attention to use 95:5 water and organic
solvent to filter, and the organic solvent can’t less than 5%.
◇For storage or stay for a night, rinse out salt and buffer solvent and reserve with pure organic
mobile phase. (Ethylic-nitrile is best)

How to solve peak tailing problems
A. Peak tailing problems concerned with chemistry
1. Add 30mM triethylamine (for basic compounds) or acetic-amine (for acid compounds), or add ace-
tic-triethylamine for unknowns into mobile phase.
2. If it is still tailing, replace the acetic-triethylamine with dimethyl-octyl amine
3. Make the injection volume less than 1μg.
B. Peak tailing problems concerned with columns
1. Reversed columns can be flushed with 20 times volume solvents: 96%CH2Cl2, 4% CH3OH (methanol),
and 1%NH4OH; normal columns can be flushed with CH3OH (methanol).
2. Use of guard columns.
C. Peak tailing and widening concerned with HPLC system
1. Syringe volume is too big (normally ≤ 25μL )
2. The tubing volume between injector, column and detector is too big (best for tube < 20cm, ID0.007”).
3. The flow cell volume of the detector is too big.
How to store columns
Make the mobile phases in accordance with the actual use to avoid the microorganism produced by
buffered solvents and aqueous mobile phases, or add 200ppm NaN3 to restrain the growth of the bacteria
(Pay attention to its toxicity and potentially explosibility); or add 20% organic modifier in mobile phases
can also restrain the bacteria, and the reconstructive solvents can be helpful to mobile phase degassing.
Use 100% organic solvents (Ethylic-nitrile is best) to store columns instead of buffered solvents.
Columns flushed with buffered solvents should be purged with 15-20 times volume organic mobile
phases without buffers, and then store with 100% organic solvents.
Don’t flush columns bonded C18 with pure water.
Screw with end caps for the both ends of the columns to avoid the packing hardened cracked.

Guard Columns Selections
As a regular question of chromatography workers, how to select guard columns but not affect separation
analysis always made them think. Usually, make sure the sample is clean before selecting guard column.
For most analytical workers, one 1cm length guard column can fully function for guard use. If samples are
very dirty or often need to change the 1cm guard column, then chose 2cm or 3cm length guard columns.
The longer the guard column, the more packing in, and the less chance the pollutants enter the column,
while the longer of the retention time. Generally speaking, the inside diameter of guard column should be
same with the chromatographic column.
Ways of packing the guard columns are also very important. At present, there is a way of membrane
packing with a very simple and convenient application process, and can be installed in the lab. But the
biggest shortcoming is not economical, especially for China’s current status, for the expense for only one
time use is comparatively high. In addition, the guard columns with the way of membrane have limited
packing materials and provide limited guard function; as these columns are short because of the few
packing materials, they have little effect on retention time of the analytical samples.
Other guard columns are curtate columns, and there are three designs: direct-connection, finger-tight,
and integral. The integral design is that the manufacturers install the guard column together with analyt-
ical column directly according to the order; it’s very convenient but can’t be modified. Direct–connection
design is that the guard columns can be connected to any kind of analytical columns and installed any-
time by workers, besides; the chromatographic workers can select different guard column length in ac-
cordance with samples. The finger-tight design and the direct-connection design are same in structure;
the only difference is it can be installed by hands instead of wrench. In addition, from the structure point of
view, some guard columns can replace their drop-in cartridges, while some others cannot. At present,
most guard columns can replace their drop-in cartridges to reduce the guard columns cost.
Most people select guard column packings in accordance with analytical column packings, normally
speaking, they are same. But they aren’t necessarily same according to actual analytical work.
The principle of selecting guard column is: on the precondition of meeting analysis and separation, select
the shortest guard column and chose packings with the least reservation to samples.

silica gel as base material, Depending on the choice of the eluent, it can both be used under normal and

DG230 On-line Degasser
The DG230 In-line Degasser operates on the principle of Henry’s Law to remove dissolved gases from
HPLC eluent. Henry’s Law states that the mole fraction of a gas dissolved in a liquid is proportional to the
partial pressure of that gas in the vapor phase above the liquid. If the partial pressure of a gas on the
surface of the liquid is reduced, for examples, by evacuation, then a proportional amount of that gas
comes out of solution. The In-line Degasser uses a gas permeable polymer membrane channel to carry
the eluent through the vacuum chamber. When the eluent enters the vacuum chamber, the vacuum
maintains a large differential in gas concentration across the membrane. This accelerates the rate at

which the dissolved gases diffuse through the polymer membrane into the vacuum chamber. The gases
are them carried away by the vacuum pump.

Based on EC2006/EC2000 workstation, EC2000 GPC adds GC data-processing function. Its characters

Calculate calibration curve according to molecular weight averages and dispersion coefficient of mono-
disperse or broad standards , to Mark-Houwink (k and a) of narrow or unknown samples; eliminate ex-
periment error by calibrating results of parallel injection of arbitrary number and all kinds standards; re-
gress calibration curve with simple equation, quadratic equation or cubic equation, and auto-calculate
standard error and correlation coefficient; obtain standards average molecular weight and error by re-
versed calculating with fitting curve, and can visually judge and modify fitting results.

By means of molecular weight calibration curve and peak slicing area, calculate number-average molec-
ular weight, weight-average molecular weight, Z average molecular weight, Z+1 average molecular

calculate and analyze any range of the chromatogram by setting up slicing.
◇Molecular Weight Distribution
Calculate to obtain integrated molecular weight distribution curve, differential molecular weight distribu-

EC2000DAD Chromatography Data-processing Workstation
To guarantee DAD system a stable operation under Windows98/XP2000, we adopt virtual driver. With a
friendly interface and strong function DAD data-processing platform, users can conveniently observe and
process contour map, spectrogram, chromatogram and three-dimensional spectrogram-chromatogram.
With sufficient functions of chromatographic data processing and quantitative calculating, arbitrary re-
volving and parts magnifying, users can obtain more vivid visual effect and results analysis.
DAD workstation can provide all kinds of chromatograms, including single wavelength, arbitrary
