Abstract
Molecular hydrogen (H2) was believed to be inert and nonfunctional in mammalian cells. We overturned this concept
by demonstrating that H2 reacts with highly reactive oxidants such as hydroxyl radical ((•)OH) and peroxynitrite
(ONOO(-)) inside cells. H2 has several advantages exhibiting marked effects for medical applications: it is mild enough
neither to disturb metabolic redox reactions nor to affect signaling by reactive oxygen species. Therefore, it should
have no or little adverse effects. H2 can be monitored with an H2-specific electrode or by gas chromatography. H2
rapidly diffuses into tissues and cells to exhibit efficient effects. Thus, we proposed the potential of H2 for preventive
and therapeutic applications. There are several methods to ingest or consume H2: inhaling H2 gas, drinking H2-
dissolved water (H2-water), injecting H2-dissolved saline (H2-saline), taking an H2 bath, or dropping H2-saline onto the
eyes. Recent publications revealed that, in addition to the direct neutralization of highly reactive oxidants, H2 indirectly
reduces oxidative stress by regulating the expression of various genes. Moreover, by regulating gene expression, H2
functions as an anti-inflammatory, antiallergic, and antiapoptotic molecule, and stimulates energy metabolism. In
addition to growing evidence obtained by model animal experiments, extensive clinical examinations were performed or
are under way. Since most drugs specifically act on their specific targets, H2 seems to differ from conventional
pharmaceutical drugs. Owing to its great efficacy and lack of adverse effects, H2 has potential for clinical applications
for many diseases.

Abstract
Hydrogen is an established anti-oxidant that prevents acute oxidative stress. To clarify the mechanism of
hydrogen's effect in the brain, we administered hydrogen-rich pure water (H(2)) to senescence marker protein-
30 (SMP30)/gluconolactonase (GNL) knockout (KO) mice, which cannot synthesize vitamin C (VC), also a well-
known anti-oxidant. These KO mice were divided into three groups; recipients of H(2), VC, or pure water
(H(2)O), administered for 33 days. VC levels in H(2) and H(2)O groups were <6% of those in the VC group.
Subsequently, superoxide formation during hypoxia-reoxygenation treatment of brain slices from these groups
was estimated by a real-time biography imaging system, which models living brain tissues, with Lucigenin
used as chemiluminescence probe for superoxide. A significant 27.2% less superoxide formed in the H(2)
group subjected to ischemia-reperfusion than in the H(2)O group. Thus hydrogen-rich pure water acts as an
anti-oxidant in the brain slices and prevents superoxide formation.

AIM: Reactive oxygen species (ROS) contribute to the development of periodontitis. As molecular hydrogen can act as a
scavenger of ROS, we examined the effects of treatment with hydrogen-rich water on a rat model of periodontitis.
MATERIAL & METHODS: A ligature was placed around the maxillary molars for 4 weeks to induce periodontitis, and the animals
were given drinking water with or without hydrogen-rich water.
RESULTS: The rats with periodontitis which were treated with pure water showed a time-dependent increase in serum ROS level.
Compared with the rats without periodontitis, the periodontitis-induced rats which were given pure water also showed
polymorphonuclear leucocyte infiltration and alveolar bone loss at 4 weeks. Hydrogen-rich water intake inhibited an increase in
serum ROS level and lowered expression of 8-hydroxydeoxyguanosine and nitrotyrosine in the periodontal tissue at 4 weeks. Such
conditions prevented polymorphonuclear leucocyte infiltration and osteoclast differentiation following periodontitis progression.
Furthermore, inflammatory signalling pathways, such as mitogen-activated protein kinases, were less activated in periodontal
lesions from hydrogen-rich water-treated rats as compared with pure water-treated rats.
CONCLUSION: Consuming hydrogen-rich water might be beneficial in suppressing periodontitis progression by decreasing gingival
oxidative stress.

Summary :
The purpose of this study was to investigate skeletal muscle fatigue with hydrogen-rich water (HRW) in
collegiate athletes. Ten collegiate basketball male players aged 20 ± 1.47 years were recruited in this
study. All subjects were free of any drugs and ergogenic acid such as ginseng and vitamins. Subjects
were randomly divided into two groups (HRW group and placebo group). Each group received HRW or
placebo (7 ml/kg body weight) for 14 days. Electromyography (EMG) responses of left quadriceps muscle-s
(rectus femoris; RF, vastus lateralis; VL, vastus medialis; VM) during 40 maximal eccentric, isokinetic
muscle action was examined, and then each group received additional HRW or placebo for 3 days.
The same protocol was repeated. EMG amplitude demonstrated that there was a significant negative
linear relationship (p<.05) for RF, VL and VM of % MVC in HRW group, while there was no significant
negative linear relationship for VL in placebo group. After three day resting period, EMG amplitude also
demonstrated that there was significant negative linear relationship (p<.05) in RF of %MVC, VM of %MDF in
HRW group, while there was no significant effect in RF of %MDF in placebo group.
Thus, the results showed that HRW enhanced skeletal muscle fatigue in collegiate athletes.
It is concluded that HRW may be effective to improve the recovery time from skeletal muscle fatigue after
