Das Center of Drug Absorption and Transport (C_DAT) ist ein nationales Kompetenzzentrum, in dem interfakultäre Forschergruppen der Ernst-Moritz-Arndt-Universität Greifs-wald und der Universitätsmedizin Greifswald (Abteilungen für allgemeine und klinische Pharmakologie, Abteilung für Biopharmazie und pharmazeutische Technologie und Abtei-lung für pharmazeutische Biotechnologie) die Prozesse der Absorption sowie des Transports von Arzneistoffen hin zu Orten ihrer erwünschten (und unerwünschten) Wirkung im Körper des Menschen erforschen.
Strukturell ist das C_DAT aus den Instituten für Pharmako-logie und Pharmazie hervorgegangen. Der baulichen Vereini-gung ging eine Phase der intensiven Forschungskooperati-on der heute im C_DAT beheimateten Gruppen voraus, die seit Mitte der Neunzigerjahre durch Forschungsförderung der BMBF-Programme NBL1-3 ermöglicht wurde. Hierbei wurden die Grundlagen für die aktuelle Forschungsausrich-tung erarbeitet. Die gewonnenen Erkenntnisse der Transpor-ter-Pharmakologie führten zur internationalen Etablierung der beteiligten Arbeitsgruppen und im Jahr 2007 zur erfolg-reichen Einwerbung des Unternehmen-Region-Förderpro-gramms (BMBF) InnoProfile (2007-12). Dieses ermöglichte erstmalig die Etablierung einer interfakultären Nachwuchs-forschergruppe.
In Zusammenarbeit mit kleinen und mittleren Unter nehmen der Region aus dem Life-Science Bereich wurden hierbei die Transporter-Protein vermittelnden Absorptions- und Ver-teilungsvorgänge von Arznei stoffen im menschlichen Orga-nismus erforscht und praxisrelevante Forschungsmethoden und Applikationskonzepte entwickelt.
Aufgrund des großen wissenschaftlichen Erfolgs der inter-fakultären Kooperation, erfolgte die bauliche Verstetigung der gewachsenen Forschungsstrukturen dann im Jahr 2008 durch die Genehmigung des C_DAT-Forschungszentrums als „Bau von nationaler Bedeutung“ durch die gemeinsame Wissen schaftskonferenz von Bund und Ländern. Nach nur dreijähriger Planungs- und Bauzeit konnte das C_DAT im November 2011 in Betrieb genommen werden.
The Center of Drug Absorption and Transport (C_DAT) is a national center of expertise in which research groups of the Ernst-Moritz-Arndt University of Greifswald and the University Medical Center Greifswald (Departments of General and Cli-nical Pharmacology, Department of Biopharmaceutics and Pharmaceutical Technology, Department of Pharmaceutical Biotechnology) investigate the processes of absorption and transport of drugs through to their desired (and unwanted) sites of effect in the human body.
Structurally, the C_DAT emerged from the Departments of Pharmacology and Pharmacy. The structural organization was preceded by a phase of intensive research cooperation of today‘s research groups of C_DAT, made possible since the mid-nineties of the last century by the BMBF research funding programs NBL1-3. Here, the basis for the current research orientation was developed. The findings of the transporter pharmacology led to the international recog-nition of the working groups involved and in 2007 to the successful raising of company-region development program (BMBF) InnoProfile (2007-12), which enabled for the first time the establishment of an interdepartmental research group.
In cooperation with small and medium-sized enterprises in the region from the life science sector in this case the trans-porter protein-mediated absorption and distribution opera-tions of pharmaceutical substances in the human organism have been researched and developed practice-relevant re-search methods and application concepts.
Due to the huge scientific success as caused by implemen-tation of modern research structures, the C_DAT, a building of national importance, has been approved in 2008. After only three years of planning and construction, the C_DAT could be put into operation in November 2011.

In the Clinical Study Unit, investigator initiated studies (IITs) and studies for external sponsors are
conducted in healthy subjects according ICH_GCP. The sponsor for IITs is the University Medicine
Greifswald. The clientele of healthy subjects (mostly students, 20-30 years) is genotyped on demand
for clinical relevant enzyme and transporter polymorphisms. The unit has long-lasting experience in all
kinds conducting pharmacokinetic studies (bioequivalence, metabolism, drug interactions, pharma-

Arzneistoffanalytik
Die Core Unit Arzneistoffanalytik übernimmt die quantitative Bestimmung von Arzneistoffen und deren Metaboliten, Biomarkern und Mo-
dellsubstanzen aus klinischen, tierexperimentellen, zellbiologischen und pharmazeutisch-technologischen Untersuchungen. Der Protein-
gehalt klinisch relevanter Arzneistofftransporter und Phase-I/II-Enzyme wird in Zellen und Gewebe massenspektrometrisch quantifiziert.
Die Core Unit beteiligt sich im therapeutisch-toxikologischen Service an der Krankenversorgung.
Alle Methoden (inkl. Proteomanalytik) werden entsprechend den FDA/EMA-Empfehlungen validiert und qualitätskontrolliert. Für Analy-
sen im Rahmen klinischer Studien ist die Core Unit GLP-zertifiziert.

The core unit Drug Analysis provides methods for the quantitative determination of drugs and their
metabolites, biomarkers and probe substances in clinical, animal-experimental, cell-biological and
pharmaceutical-technological studies. The protein content of clinically relevant drug transporters and
phase-I/II enzymes can be quantified in cells and tissue using mass spectrometry. The core unit
provides also a therapeutic-toxicological service in patient care.
All methods (incl. proteome analytics) are validated and quality controlled according to the FDA/EMA
recommendations. The unit is GLP-certified for drug analysis in clinical studies.

Die Core Unit Molekular- und Zellbiologie untersucht Arzneistofftrans-
porter durch Charakterisierung der DNA (Sequenzierung, Genotypisie-
rung), Quantifizierung von mRNA-Expression (real time RT-PCR) und
Proteingehalt (Massenspektrometrie), Visualisierung der zellulären Lo-
kalisation (Fluoreszenz- und Konfokalmikroskopie) sowie Beurteilung
der Transportfunktion (validierte in-vitro Zellmodelle). Sie untersucht
regulatorische Veränderungen durch endogene Stoffe und Xenobiotika
(Reportergenuntersuchungen, Proliferationsassays, Überexpressions-
systeme).
Für klinisch relevante efflux und uptake Transporter und deren geneti-
sche Varianten stehen validierte Zellmodellen (HEK293, MDCKII) zur
Verfügung (incl. inside-out Lipovesikel). Die Routineplattform wird zur
Beurteilung des zellulären Transports von Arzneistoffen in Kompeti-
tions-, Uptake- und TransWell-Assays verwendet.
In Analogie GLP-Konzept arbeiten wir nach einem internen Qualitäts-
standard Good Cell Culture Practice.
Transportstudien mit primären Hepatozyten (Mensch, Affe, Ratte,
Hund, Minipig, Pferd) sind in Zusammenarbeit mit der PRIMACYT Cell
Culture Technology GmbH (Schwerin) möglich.
Molekular- und Zellpharmakologie Molecular and Cell Pharmacology
The Core Unit Molecular and Cell Pharmacology studies drug transpor-
ters by evaluation of the DNA (sequencing, genotyping), quantificati-
on of mRNA-expression (real time RT-PCR) and protein content (mass
spectrometry), imaging of cellular localization (fluorescence and con-
focal microcopy) and characterization of the function (validated in-vitro
cell-models). The influence of endogenous compounds and xeno-
biotics is measured using reporter gene and proliferation assays and
overexpression cell systems.
For clinically relevant efflux and uptake transporters and their genetic
variants, validated cell models (HEK293, MDCKII) are available (incl.
inside-out lipovesicles) to evaluate cellular transport in competition-as-
says, uptake-assays and TransWell™ assays.
In the unit, the internal quality standard Good Cell Culture Practice is
realized by analogy to the GLP-conception.
Transport studies with primary hepatocytes (human, monkey, rat, dog,
mini pig, horse) are available in cooperation with the PRIMACYT Cell
Culture Technology GmbH (Schwerin).

alle festen, halbfesten und flüssigen Arzneiformen wie Pulver, Granulate, Pellets,
Kapseln, Tabletten, Suppositorien, Salben, Cremes, Gele, Lösungen, Suspensionen,
Augentropfen sowie Injektions- und Infusionszubereitungen im Labormaßstab und
für eine Vielzahl von Zubereitungen auch im Technikumsmaßstab hergestellt wer-
den. Weiterhin sind neben einer modernen analytischen Geräteausstattung mit z. B.
HPLC-Anlagen mit verschiedenen Detektionsmöglichkeiten, AAS etc. alle wesentli-
chen Prüfverfahren für die Untersuchung des Freisetzungsverhaltens von festen und
halbfesten Darreichungsformen sowie transdermalen therapeutischen Systemen
vorhanden.
Besondere Expertise besteht darüber hinaus in der Vorhersage des Wirkstofffrei-
setzungsverhaltens von oralen, okkularen, vaginalen, rektalen oder parenteralen
Darreichungsformen im Organismus. Darüber hinaus entwickeln wir Formulierungen
und prädiktive Testmethoden für primär schwer lösliche Arzneistoffe in modifiziert
freisetzenden Darreichungsformen.
Die vorhandenen Geräte und Kenntnisse sind damit ideal auf die Entwicklung von
neuen Konzepten für Darreichungsformen und zum frühzeitigen Erkennen von po-
tenziellen Problemen bei der Wirkstofffreisetzung geeignet.

The Institute of Biopharmaceutics and Pharmaceutical Technology has all of the
necessary equipment and resources to manufacture solid, semisolid and liquid do-
sage forms including powders, granules, pellets, capsules, tablets, suppositories,
ointments, creams, gels, solutions, suspensions, eye drops as well as injectables
and infusions in laboratory scale and in many cases also in pilot plant scale. Ana-
lytical resources include state of the art analytical equipment, e.g. HPLC with a ran-
ge of detectors and AAS amongst others. Beside this, the institute has capability to
perform all relevant drug release experiments for solid and semisolid dosage forms
and transdermal therapeutic devices.
We can offer special expertise in predicting in vivo drug release from oral, ocular,
vaginal, rectal and parenteral formulations. Moreover, we develop formulation strate-
gies and predictive screening methods for poorly soluble compounds and modified
release formulations.
Overall, apparative equipment and expertise are tailored for developing new con-
cepts in the design of pharmaceutical dosage forms and delivery systems and en-
able a prediction of (unwanted) in vivo dosage form performance in early stages of
formulation development.
