Step 1: Std. Calibrating curve
10 mg drug was dissolved in 100 ml pH 6.8 Phosphate Buffer, to prepared 100 ug/ml solution
0.2 ml solution is diluted with 10 ml Buffer 2 µg/ml solution
0.4 ml solution is diluted with 10 ml Buffer 4 µg/ml solution
0.6 ml solution is diluted with 10 ml water 6 µg/ml solution
0.8 ml solution is diluted with 10 ml Buffer 8 µg/ml solution
1.0 ml solution is diluted with 10 ml Buffer 10 µg/ml solution
1.2 ml solution is diluted with 10 ml Buffer 12 µg/ml solution
(Above all solutions are 10 µg/ml solution)
Step 2: Take absorbance of all solution and Plot graph in MS Excel.
Step 3: After taking absorbance, to plot graph in Excel, Click Insert Chart Scatter

Time: Time for dissolution is vary dosage from to dosage from.
Abs.: Taking solution after suitable time interval, to take absorbance of this solution, if absorbance is
more than 1, to dilute the solution.
Conc µg/ml: 1. To calculate Conc µg/ml with the help of Y = mx + c (Equation: std.
calibrating curve) this equation,
2. To convert this equation in x = Y-c / m (Y = Abs.)
DF: dilution factor is indicate number time to dilute the given solution (10, 100 and 1000).
Conc µg/ml: DF * Conc µg/ml to get Conc µg/ml.

Conc mg/ml: Conc µg/ml / 1000 to get Conc mg/ml.
Conc mg/5ml: Conc mg/ml * 5 to get Conc mg/5ml.
Conc mg/900ml: Conc mg/ml * 900 to get Conc mg/900ml.
CDR (cumulative drug release): to select reading in below the zero in CDR column *
First reading 0 in Conc mg/900ml to get CDR.
%CDR: (% cumulative drug release): (CDR*100) / Labelled Claim of drug to get
%CDR.
IMPORTANT NOTE:
1. In every calculation in Excel sheet, “Always starting the calculation with = this sine
(equal to sine)”, if you’re not used this sine calculation was not done.
2. After complete all calculation to press ENTER button to give your reading.
