The time after one new development is the time before the next
Today, biomedical research is inconceivable without continuous progress in
fluorescence microscopy. New fluorescence technologies often initiated new discoveries and
soon became standard applications, such as immunofluorescence,
which is now a routine diagnostic technique. Further examples are confocal microscopy and
live cell examinations with GFP.
The invention of fluorescence microscopy ...
In 1932, Leica designed the first epifluorescence microscope, whose technology is still used
today. Then, in 1965, epifluorescence filter cubes were developed at Leica in
cooperation with Professor Ploem; they were adopted by other manufacturers and have
remained the generally accepted standard right up to the present day.
As Leica is in constant contact with leading research scientists and laboratories
all over the world, our instruments offer special customer benefits which we combine with
innovative technologies and the quality of Leica optics.
... and the revolution in light microscopy
The latest example of our pioneering spirit and probably one of the most fascinating
developments ever is the high-resolution Leica TCS 4Pi microscope,
which can image cell structures that were previously beyond the resolving power of
conventional light microscopy. Together with Professor Stefan Hell,
Director of the Max Planck Institute for Biophysical Chemistry in Gttingen, Leica Microsystems
has developed a method which leads to a substantial reduction of focal volume.
This improves the resolution along the optical axis three- to seven-fold,
opening completely new horizons for research scientists.
Unlimited Progress

If only all decisions were so simple
Despite the complexity of a fluorescence microscope, the criteria for its quality are
crystal clear. There are three main aspects for assessing any fluorescence microscope:
Optics, i.e. the brilliance and wealth of contrast of the images Ease of use, i.e. well thought-out operation concept
Practical orientation and application potential, i.e. a wide and flexible range of equipmentBrilliance: a visible difference
Leica microscopes have always been famous for their optics. And rightly so:
Leica images feature outstanding f luorescence, razor-sharp contours and breathtaking contrast.
The finest structures are clearly visible even when the excitation intensity is
attenuated; unique brilliance ensures top imaging precision.
Ease of use: when jobs get harder, tools must get easier
It may sound paradoxical, but the more difficult the scientific challenges, the easier the
microscope should be to use. This is the only way to be able to concentrate fully on the
subject of research and obtain quick results even in extremely time-consuming research series.
Practical orientation: reproducibility and precision for all applications
From climate chambers to sample manipulation to filter changers: all components
interact in perfect harmony, guaranteeing simple work routines, reproducible images and, most importantly,
the knowledge that you can rely a hundred percent on your results.

Answers To Every Question
Fluorescence
stereomicroscopy and
macroscopy:
Select, sort and assess
in 3D fluorescence.
Our stereomicroscopes
allow 3D imaging of
specimens and therefore
precise manipulation of
fluorescing objects.The upright fluorescence
macroscopes, on the
other hand, offer parallax-
free precision and a free
working distance that is
long enough to enable
viewing of specimens
the size of a mouse.
Fluorescence microscopy:
Our range of fluorescence
microscopes is extremely
wide. From the DM1000
standard routine microscope
to the high-end research
microscopes DM6000 and
DMI6000, youll be sure tofind the right system for any
application: we offer an
extensive choice of different
cameras and software
solutions. All the systems
feature brilliant optics, so
theres no need to sacrifice
high precision whatever the
size of your budget.
Confocal microscopy:
The essential characteristics of
confocal microscopy are top image
quality through optical sections and
outstanding three-dimensionalreconstruction of data sets.
We offer many different configurations
in this area of microscopy including
the worlds first filter-free Spectral
Confocal Microscope.

Digital Photography and Data Analysis
Experimental fluorescence
techniques:
Fluorescence Recovery after
Photobleaching (FRAP),
Fluorescence Resonance
Energy Transfer (FRET),
Photoactivation,
Fluorescence LiftetimeImaging (FLIM),
Fluorescence Correlation
Spectroscopy (FCS), etc.
enable the representation of
molecular interactions and
dynamics, answering
questions on cell functions.
Data analysis:
The software-supported
quantitative evaluation of
image data e.g. for FCS
or FLIM round off the
scientific information
yield of the experiment.
An essential requirement for reliable data analysis is excellent optical imaging quality as guaranteed by our
digital cameras. Choose between various models to suit your application needs.
Besides actual image information, interest is increasingly focusing on the development of new experimentalfluorescence techniques for exploring cell functions. These require powerful software solutions that are
precisely matched to your specifications and perfectly tuned to our cameras and systems. Thus equipped,
you can rely on smooth and safe data analysis and management for all your projects.
Image processing:
In traditional fluorescence microscopy,
images frequently seem slightly
blurred due to the fact that information
coming from outside the focal planeis included in the image. Image quality
can be significantly enhanced by
processing with mathematical
algorithms (deconvolution).
