What is Organ-on-Chip?
Create the smallest functional modules of healthy or diseased tissues / organs ‘on a chip’ / microscope slide
using microfluidics, microelectronics and microfabrication
• Human stem cells from healthy individuals or patients (adult stem cells, organoids, iPSC)
• 3D culture interaction between different cell types, including immune cells, under controlled conditions
• Human genetics • Manipulation of the (patho)physio-logical
environment: biochemical + physical factors • Long term culture: time for disease to develop • Real time monitoring
Why Organ-on-Chip?
The problem: • Limited availability of human model systems for preclinical research. • Many drug failures/withdrawals due to unreliability of predicted
efficacy in humans + unanticipated toxicity
Need for:
1. human disease models (including immune response) for development of therapies - Drugs and physical therapies (e.g. hyperthermy/radiotherapy) - Novel drugs and off-label
2. human organ models for toxicity assessment and cosmetics
Animal models: not sufficiently representative for human disease and toxicity
