Research 2.0
As well as doing leadingedge biomedical research, the PRBB cen
tres also want their discoveries to reach as many interested people as possible. That’s why they are consolidating their presence in the social network: to get their message instantly to an everincreasing number of people from all over the world. The IMIM was pioneering with its Twitter account. Today, we can follow the CRG activities or become fans of the FPM in Facebook, get to know the CMRB or the UPF communities better through LinkedIn and watch CREAL videos on YouTube. The PRBB is also present in the web 2.0, collaborating in the science communication from the centres. The success has been spectacular: during 2011 our followers in Twitter have increased from 123 to 543 and our Facebook friends have doubled, reaching 533. Our videos on YouTube have been reproduced more than 1,200 times and the number of PRBB followers in LinkedIn has quadrupled. If you still don’t follow us, join us now!

Recerca 2.0
Si encara no ho feu, us animem a que ens seguiu!

A PRBB core facilities coalition supports research
On February 9, the PRBB residents had the chance to find out about the
latest technologies in the park 's core facilities, the applications and services they offer and how they can support research. During the event organised by Doris Meder, head of the CRG core facilities, most technical services at the park were presented: the CRG histology, biomolecular screening and protein technologies, bioinformatics and genomics units; the mixed CRG/UPF microscopy, FACS and proteomics units; the UPF peptide and genomics services, and the IMIM microarrays and molecular cytogenetics platforms. According to Meder, the core facilities allow to share knowledge
and costs and to make the best use of the experts who run them. “We constantly develop and implement new technologies according to the needs of the scientists, and we would love them to tell us what they need. All projects are welcome”, she encouraged. Researchers at the PRBB have access to about 15 different internal scientific and technical services. With the exception of the PRBB animal facility, all the services are managed by the research centres, mostly the CRG, UPF and IMIM. After a decision of the PRBB scientific committee, the services are offered to the park’s entire scientific community for the same internal service fee. In general, the capacities of the core facilities meet the internal needs of the park’s researchers, but they also provide services to external public and private entities ■

Listening to the language of neurones
Coming from the Rockefeller University in New York, the Belgian researcher Matthieu Louis le
ads the sensory systems and behaviour group at the CRG. This is the only lab in Barcelona, and one of the few in Spain, investigating Drosophila neuroscience. His team comprises eight people with backgrounds in molecular biology, engineering and physics. Their aim is to correlate neural circuit function with behaviour using fruit fly larvae. “The Drosophila larva has a large repertoire of complex behaviours and key cognitive functions. Yet its nervous system has10 million neurones fewer than humans”, explains the physicist. Therefore it is a very useful model system. The research group tries to understand how odours are encoded by the olfactory system. Features such as quality –does this smell like banana?– and intensity –is this a morsel of banana or a bunch?– of an odour are efficiently represented by only 21 olfactory sensory neurones, so that the larva can distinguish between hundreds of foodrelated odours. The researcher says that there must be a combinatorial code, yet it does not seem to be as trivial as the activation of different combinations of neurones by distinct odours. “We have evidence that the nature and the intensity of an odour is represented not only by the identity of the sensory neurones the odour activates, but also by the way each neurone is activated”, he explains.
From information processing to chemotaxis
Once a smell has been encoded, it has to be processed. To find the higherorder neurones involved in the integration of olfactory information, the group is undertaking a large behavioural screen. They are testing thousands of fly lines in which subsets of neurones are inhibited or overactivated. They then characterise how these perturbations affect chemotaxis, the orientation behaviour observed in response to an odour gradient. To decide whether to go straight ahead or turn, the larva monitors information about odour concentration changes. When a wildtype animal detects an intensity increase of an attractive odour, it keeps going forwards. But, as the group has recently described, if the odour intensity decreases the larva reorients through an activesampling mechanism: much like rats and dogs, the larva sweeps its head laterally to check intensity levels on either side. With their screen, the researchers are looking for mutants showing reorientation defects. To that end, they have developed their own computervision software. “We needed an algorithm to quantify subtle movements of the head and body posture with a high spacetime accuracy. As no tool like this existed, we spent a year developing one”, says the head of the group.
Predicting behaviour
“If we understand the neural logic of larval chemotaxis well enough, we should be able to synthetically produce predictable behavioural sequences”. Such a model could be useful for robotics. “Currently, dogs are trained to find mines. We could design robots that navigate spatially, searching for the chemical compounds present in explosives”. Many questions remain to be answered before this becomes reality. How does the larva integrate a series of stimuli (touch, light, heat, smell) that are received simultaneously before deciding what to do next? And how is sensory input converted into motor output? “There are many challenges ahead. But it is thrilling to witness the genesis of a decision in a mini brain, from elementary spikes in sensory neurones down to the coordinated contraction of dozen of muscles. Flies have much to teach us about the function of our own brain”, concludes the researcher ■

In a paper published in Nature Medicine, the researchers treated cells continuously with cetuximab until some developed resistance, and the same mutation was identified in all of them. The authors confirmed that this mutation had appeared in two out of ten subjects who had disease progression after cetuximab treatment. Because the mutation did not confer resistance to panitumumab, the work could help in deciding which cetuximabresistant patients could still benefit from using this other drug ■

Moltes coses!
Durant anys vam ser només tres persones: Lluís Espinosa, Julia InglésEsteve i jo.

“We are in a privileged place for doing research”
What was your PhD on?
Icharacterised haematopoietic progenitors to use in autologous bone marrow transplants for leukaemia patients.
Since then I have always worked with stem cells, and I’m most interested in the haematopoietic system.
After that you left for the US.In 1993 I went to the Fred Hutchinson Cancer Research Center in Seattle. There was a group pioneering transplant protocols who we collaborated with. There I studied how haematopoietic stem cells regulate and amplify. That got me totally hooked. My thesis had been fairly descriptive, but during this postdoc I really went into it in depth, I wanted to understand why those cells had an infinite capacity to replicate.
When did you set up your own group? After four years of postdoc I returned to Barcelona, to what was the IRO and
is now IDIBELL. The first three years were very hard, perhaps the most difficult of my career. I often wondered if it was really what I wanted to do and so I didn’t take any students on. For years there were only three of us, Lluís Espinosa, Julia InglésEsteve and me.
But you kept going, and you were still there 12 years later.Yes, in the end, working hard, and thanks to this great team of three, we made it. We started publishing and getting more money. Then the group grew and got the opportunity to come to the IMIM and the PRBB. Here there was a critical mass, more opportunities for collaboration with other researchers and very good central services. It was a great change.
What do you think about it three years later?I still think we are in a privileged place for doing research. The administration is something that could be improved, because it is very difficult to do any thing official, even things that seem trivial, like a foreign student coming to do a six month stay!
What’s your day-to-day work like? The truth is that since we came here and I have a larger group, I haven’t even stepped into the lab to do any experiments. I'm on top of everyone’s work, and I meet with my students to design and analyse experiments, but mostly I spend my time writing articles and managing, and about twice a month I have to go to a conference or meeting, examine a thesis, evaluate projects, and so on.
What does one need to do research?Loads of stuff! You need to be restless, be able to analyse, see things from different points of view, consider many things at the same time and integrate them, have original ideas and design and carry them out. Patience is also necessary, but above all, you should have fun and believe that what you do is worth it simply to understand how things work.
What do you do when you’re not working?I love playing tennis. It helps releasing tension and I do it every weekend, if I can. Also I go to the cinema and travel. This is, in fact, one of the advantages of doing research, you meet people from around the world and, for one thing or another, you have to travel quite a lot! ■

GONZALO GIRIBET, Monday March 5. Giribet, from the Department of Organis-mic and Evolutionary Biology, at Harvard University in Cambridge, USA, focuses on the evolution and biogeography of invertebrate animals. Current projects in-clude a multidisciplinary study to assemble a stable Animal Tree of Life, exploring phylogenomic data, mostly based on novel EST (expressed sequence tags) data
but also based on available genomic information, of most animal phyla. At the same time he has also developed new strategies for coding morphological information from extant and extinct metazoans and combined them with the phylogenomic data. He is also interested in philosophical aspects of sequence data analysis, emphasizing homology-related issues. He has been invited by Jaume Bertranpetit (UPF).
JANUSZ M. BUJNICKI, Monday March 19. Bujnicki, from the Laboratory of Bioin-formatics and Protein Engineering at the International Institute of Molecular and Cell Biology in Warsaw, Poland, is doing both theoretical and experimental research on nucleic acids and proteins. The current focus of his group is on RNA sequence-structure-function relationships (in particular 3D modelling), RNA-protein complexes, and enzymes
acting on RNA. He develops and uses computer programs for modelling of three-dimensional protein structures based on heterogeneous, low-resolution, noisy and ambivalent experimental data. He is also involved in genome-scale phylogenetic analyses, with the focus on identification of proteins that belong to particular families. His ultimate goal is to identify complete sets of enzymes involved in particular metabolic pathways (e.g. RNA modification, DNA repair) and to design proteins with new properties, in particular enzymes with new useful functions, which have not been observed in the nature. He has been invited by Jordi Villà-Freixa (UPF).

New stress-protection cell mechanism
Cell stress can inhibit cell proliferation, but the cells develop adaptive responses in order to maintain their
functionality in these conditions. According to the results of a study coordinated by José Aramburu from the CEXSUPF, the protein
mTOR plays an important role in this adaptation. The work published in Nucleic Acids Re-search reveals how mTOR remains active under moderate stress and modulates the expression of the genes involved in cell adaptation. By inhibiting mTOR, the authors have demonstrated that this protein is necessary in order to maintain the ability of cells to proliferate in stressful situations. The researchers exposed mouse fibroblasts to osmotic stress –a higher salinity than normal– sufficiently strong to elicit a response from the cell, but moderate enough to allow the cell to adapt. These conditions simulate the osmotic stress that can be observed in cases of severe dehydration and renal dysfunction ■

In addition, all journals require a section

on materials and methods, where details of the experiments undertaken and resources used must be presented. This is especially important as it enables other researchers to replicate the experiments. With the current “peer review” system, once the editors of the journal receive the article they look for two or three external experts to review it for free, and anonymously. The reviewers must check that the experiments were done correctly, that the conclusions are sound and realistic, and that the findings represent a real advance in the field. These experts, in general, are also active researchers. In this way, scientists monitor and correct each other. There are inherent difficulties with this system. Can researchers who are potential competitors of the authors be truly objective? The problem is that, in scientific research, only researchers working in the same field are capable of evaluating an article. The good news is that all scientists in the world are in the same boat: all are, at some time, authors and reviewers. Therefore, although the “peer review” system is not perfect, it is the best we have ■

DEBAT ACTUAL / CURRENT-AFFAIRS DEBATE
Should “dangerous” data be published?
recently, two research groups demonstrated that it is possible to create a more transmissible form of the h5n1 avian flu virus. articles explaining this research we-re sent to Science and Nature, but are currently on hold due to recommendations from the us national science advisory Board for Biosecurity (nsaBB). heated debate has arisen about this unprecedented censorship and about whether the research should be published in full, allowing others to replicate it, or whether this information would be too dangerous in the wrong hands. should scientific information, on occasions like this, be censored? how involved should the public and government bodies be in discussing the fate of papers like these? El·lipse asked several scientists at the prBB centres their opinion on the matter.
“It seems likely, although not yet sure, that the papers will be published in full. Censoring research results to avoid their possible use by terrorists is doomed to failure. It is naïve to think that this type of information will not end up being disseminated sooner or later (fortunately, we have the Internet). This aside, I believe that this controversial research on avian influenza is completely legitimate and necessary to both prevent and better manage future epidemics. Keeping scientific information from the public and justifying secret research are two sides of the
same –unacceptable– coin: how many, and which, governments are currently but secretly developing biological weapons?”
“These studies should be released to peerreviewed publications. Firstly, all the
S’haurien de publicar dades «perilloses»?mutations introduced into the new H5N1 laboratory strain, although not reported to have occurred in combination, are known of and already appear in natural strains. Secondly, the techniques for generating mutant viruses in vitro are also well known and available in many virology laboratories. Therefore, the risk of increasing the probability of an ‘intentional’ release of lethal viruses due to the procedures detailed in the studies is not founded. Whether an accidental release of such a virus could produce a new pandemic is also speculative. While ferrets have been used as a mammalian model for influenza, it is unclear how this ‘new’ strain might transmit in humans. Overall, these studies could lead to a better understanding of pandemic dynamics. Of course, the possibility of an accidental outbreak cannot be denied, but, as the authors of the papers state, ‘the benefits of sharing this stateoftheart data outweigh by far the potential risks’.”
Mark Isalan (CRG)
“The recent voluntary moratoria on releasing certain flu research have led to reactions ranging from ‘ban all dangerous research’ to ‘allow freedom for all scientific data’. In a world where you can order genes from DNA synthesis companies at the touch of a button, it would appear prudent to withhold such data on potentially lethal viruses. This seems a reasonable compromise as long as the people using the knowledge for useful purposes (e.g., making predictive vaccines) do get access to it. Interestingly, the DNA synthesis companies do several stages of automated and manual screening for orders from potential bioterrorists. So don’t try it!” ■

Aires de canvi al CRG Joves investigadors del PRBB, premiatsWinds of change at the CRG
These last months have seen many changes at the CRG. On an organisational level, a new programme
has come about through fusion of the former gene regulation and differentiation and cancer programmes. This gene regulation, stem cells and cancer programme will focus on the study of the mechanisms controlling gene expression, on how cells specialise and what makes them become cancerous. Thomas Graf and Juan Valcárcel will share the coordination of the new programme. This merger is part of a reorganisation proposed by the CRG’s scientific advisory board and management in order to optimise resources and create synergies bringing together research groups working in the same field. At the staff level, Eduard Sabidó, just arrived from Zurich, has become the new head of the CRG/UPF Proteomics Unit. The French molecular biologist Guillaume Filion, coming from Amsterdam, has also joined the centre. His research group on genome architecture will focus on understanding the regulatory genome, the largest part of the genome which does not code for proteins. On the other hand, Hernán LópezSchier and his group will be leaving in March after nearly six years at the CRG. The group is moving north to Munich, where LópezSchier will become director of the Department of Sensory Biology and Organogenesis at the IDGHelmholtz Zentrum ■
