METHODSNinety-eightsamplesincludingcerebro-spinal(6),pleural(18),ascitic(5),synovial(54),pericardic(2)andbronchoalveolarliquid(BAL,13)werecollectedover10weeksinaprospectivestudyatNantesUniversityHospital (figure 1). The samples were analyzed in duplicate by the Yumizen®H2500 to determine thenumber of leukocytes and erythrocytes, the partial formula (% of mononuclear cells (%MN) and % ofpolymorphonuclearcells(%PN))andtodetectabnormalcells.ResultswerecomparedtoaconventionalmicroscopiccountandtotheXN10(Sysmex®)referenceanalyzerinbodyfluidmode.Both instrumentswerecalibratedpriortothesamplesanalysisusingexternalcalibrators.Passing-Bablokregressionwasusedtoanalyzeresults.

Seventy-fouroutof98sampleswereusedtocompareleukocyteanderythrocytecount.ElevensampleswereexcludedbecausetheywereoutsidetherangeoflinearityoftheanalyzersandtheBALwereexcludedduetoviscosityrelatedflags.Thecorrelationcoefficientbetweenthe2analyzerswas0.991for leukocyte (figure2)and0.955betweenYumizen®H2500andtheconventionalchambercountingmethod(figure3).Discrepantresultswereobservedinlipemicsamplesandsamplescontainingcrystals.Forerythrocytecounts,thecorrelationcoefficientwas0.996betweenbothanalyzers.

Seventy-onesampleswereretained.Comparisonoftheanalyzersforthe%ofPMNyieldedaslopeof0.931,interceptof+0.18andacorrelationcoefficientof0.933(figure4A).Thecoefficientwas0.914betweenthemanualmodeandYumizen®H2500.Thepresenceofabnormalplasmacellsinthesamplecauseddiscrepantresultsbetweenbothanalyzers(figure4B,figure6)andbetweentheYumizen®H2500andmanualcount.The%MNcorrelationcoefficientbetweenthetwoanalyzers,andbetweentheYumizen®H2500andthemanualmodewasrespectively0.933(figure5)and0.917.

LeukocyteandErythrocytecount Differentialcount

ThisstudyconfirmedthegoodanalyticalperformanceoftheYumizen®H2500incomparisonwithconventionalmicroscopiccountaswellaswiththeXN10analyzer.Inaddition,datafromthisstudyallowedthescattergramthresholdsusedfortheleukocytecountandthedifferentialtobeoptimized

Automatedanalysisofbiologicalfluidsisnowrequiredtoensuretherobustnessandreliabilityofresultsreporting.Inthisstudy,weevaluatedtheperformanceofthebodyfluidcycleoftheYumizen®H2500.
