PS-III-4 GENETICS OF RESISTANCE TO MASTITIS Genetica de la resistencia a la mastitis ROBERT H. MILLER* UNITED STATES Mastitis is technically defined as an inflammation of the mammary gland. As a practical matter, it is expected that causation is bacterial infection of the gland. Additionally, a subclinical form of mastitis is widespread, in which inflammation is not obvious. Mastitis is a problem in livestock other than dairy cows but information on its extent in beef cows, sheep and pigs is scant. Table 1 gives rough estimates of losses to the U.S. dairy industry, totalling $2 billion annually. Loss of milk due to damaged secretory tissue is the major economic effect. Other important losses are cows culled prematurely, treatment costs, and unusable milk due to presence of antibiotics. Mastitis is caused by a wide variety of organisms, even including yeasts. The microorganisms most commonly involved are Streptococcus agalactiae, Staphylococcus aureus, Streptococcus dysgalactiae, and Streptococcus uberis. Also often found are so-called environmental pathogens such as E. coli and Klebsiella species. Additionally, Corynebacterium bovis is often found, but the cow seems not be be unfavorably impacted. Some pathogens readily exist outside the cow (coliform) but some do not. Mastitis is difficult to study because whether a cow becomes infected evidently involves a combination of events and conditions; timing also is important. Pathogens are generally passed from cow-to-cow, but presence of the organism is not the only requirement. Intriguingly, introduction of pathogens at teat end is by no means sufficient to induce infection with high probability. Factors conditioning the likelihood of infection may be categorized either as environmental or as cow-related. The environmental factors relate to degree of exposure to pathogens, and are often associated with herd management. Herd mastitis control regime is important -- sanitation, milking machine use, therapy practices, and exposure to infected herdmates. Season is also important -- infection is more probable during summer in the U.S. Cow factors are inherent resistance, age, stage of lactation, and physiological state (stress, nutritional status, etc.). National statistics on mastitis are not available in the U.S., but one can state that the disease is widespread. Frequency depends on the measure and length of time a cow is monitored. A d d i t i o n a l l y , a s u b c l i n i c a l f o r m of m a s t i t i s is w i d e s p r e a d , in w h i c h i n f l a m m a t i o n is n o t o b v i o u s .

    M a s t i t i s is a p r o b l e m in l i v e s t o c k o t h e r t h a n d a i r y c o w s b u t i n f o r m a t i o n on its e x t e n t in b e e f c o w s , s h e e p a n d p i g s is s c a n t .     M a s t i t i s is c a u s e d by a w i d e v a r i e t y of o r g a n i s m s , e v e n i n c l u d i n g y e a s t s . T h e m i c r o o r g a n i s m s m o s t c o m m o n l y i n v o l v e d are S t r e p t o c o c c u s a g a l a c t i a e , S t a p h y l o c o c c u s a u r e u s , S t r e p t o c o c c u s d y s g a l a c t i a e , a n d S t r e p t o c o c c u s u b e r i s . A l s o o f t e n f o u n d a r e s o - c a l l e d e n v i r o n m e n t a l p a t h o g e n s s u c h as E. c o l i a n d K l e b s i e l l a s p e c i e s . A d d i t i o n a l l y , C o r y n e b a c t e r i u m b o v i s is o f t e n f o u n d , b u t the c o w s e e m s n o t be be u n f a v o r a b l y i m p a c t e d . S o m e p a t h o g e n s r e a d i l y e x i s t o u t s i d e the c o w ( c o l i f o r m ) b u t s o m e do not.

    M a s t i t i s is d i f f i c u l t to s t u d y b e c a u s e w h e t h e r a c o w b e c o m e s i n f e c t e d e v i d e n t l y i n v o l v e s a c o m b i n a t i o n of e v e n t s a n d c o n d i t i o n s ; t i m i n g a l s o is i m p o r t a n t . P a t h o g e n s a r e g e n e r a l l y p a s s e d f r o m c o w - t o - c o w , b u t p r e s e n c e of t h e o r g a n i s m is n o t t h e o n l y r e q u i r e m e n t . I n t r i g u i n g l y , i n t r o d u c t i o n of p a t h o g e n s at t e a t e n d is b y n o m e a n s s u f f i c i e n t to i n d u c e i n f e c t i o n w i t h h i g h p r o b a b i l i t y .

    F a c t o r s c o n d i t i o n i n g the l i k e l i h o o d of i n f e c t i o n m a y be c a t e g o r i z e d e i t h e r as e n v i r o n m e n t a l or as c o w - r e l a t e d . T h e e n v i r o n m e n t a l f a c t o r s r e l a t e to d e g r e e of e x p o s u r e to p a t h o g e n s , a n d a r e o f t e n a s s o c i a t e d w i t h h e r d m a n a g e m e n t . H e r d m a s t i t i s c o n t r o l r e g i m e is i m p o r t a n t -- s a n i t a t i o n , m i l k i n g m a c h i n e use, t h e r a p y p r a c t i c e s , a n d e x p o s u r e to i n f e c t e d h e r d m a t e s . S e a s o n is a l s o i m p o r t a n t -- i n f e c t i o n is m o r e p r o b a b l e d u r i n g s u m m e r in the U. S. C o w f a c t o r s a r e i n h e r e n t r e s i s t a n c e , a g e , s t a g e of l a c t a t i o n , a n d p h y s i o l o g i c a l s t a t e ( s t r e s s , n u t r i t i o n a l s t a t u s , e t c . ) .

In o n c e - o n l y v i s i t s to 67 W i s c o n s i n h e r d s , S h o o k and M i l l e r ( 1 9 7 8 ) f o u n d p a t h o g e n s in 14 . 2 % of m i l k s a m p l e s ( p o o l e d u d d e r m i l k ) . In N o r w a y S o l b u (19 7 8 ) r e p o r t e d 1 5 . 6 % o f l a c t a t i o n s w e r e t r e a t e d f o r c l i n i c a l m a s t i t i s . C o m p a r a b l e f i g u r e s f r o m S w e d e n w e r e 7 . 9 6 % a n d 1 1 . 6 5 % in 2 b r e e d s ( P h i l i p s s o n et al., 1 9 8 0 ) . In a s u r v e y of 1 3 , 3 1 3 q u a r t e r s a m p l e s in 61 F i n n i s h h e r d s , L i n d s t r o m et a l . ( 1 9 8 1 ) r e p o r t e d b a c t e r i a l r e c o v e r y in 7.7% of s a m p l e s . O v e r 6 m o n t h s 2 3 . 6 % of c o w s w e r e i n f e c t e d . In a s u r v e y of 29 h e r d s in S w e d e n , B r o l u n d ( 1 9 8 1 ) c l a s s i f i e d 4 4 . 6 % o f m i l k s a m p l e s as i n f e c t e d , a n d 4 0 . 3 % as " l a t e n t " i n f e c t i o n s . In g e n e r a l , if o n e f o l l o w s a c o w o v e r a l i f e t i m e , f e w c o w s w i l l fail, to s h o w at l e a s t on e c l i n i c a l e p i s o d e .

    A c o m p l i c a t i n g f a c t o r is t h a t c o w s b e c o m e m o r e s u s c e p t i b l e w i t h age. F r e q u e n c y of c l i n i c a l m a s t i t i s is at l e a s t t w i c e as g r e a t in m a t u r e c o w s as in 1st l a c t a t i o n s . D e t e c t i o n of u d d e r i n f e c t i o n is r a r e in h e i f e r s b e f o r e f i r s t c a l v i n g .

    C u r r e n t m e t h o d s of m a s t i t i s c o n t r o l in t h e U.S. a r e b o t h s a n i t a t i o n a n d t h e r a p y . D a i r y m e n a r e r e c o m m e n d e d to s a n i t i z e m i l k i n g e q u i p m e n t a f t e r e a c h m i l k i n g a n d to d i p e a c h c o w ' s t e a t s in a d i s i n f e c t a n t s o l u t i o n a f t e r m i l k i n g . A n t i b i o t i c t h e r a p y is w i d e s p r e a d , e s p e c i a l l y in the U.S. w h e r e d a i r y m e n c a n e a s i l y o b t a i n a n t i b i o t i c s . It is r e c o m m e n d e d t h a t c o w s w i t h c u r r e n t or r e c e n t c l i n i c a l m a s t i t i s be t r e a t e d w i t h a n t i b i o t i c at t h e t i m e of d r y i n g off. In s o m e a r e a s d a i r y m e n a p p l y a n t i b i o t i c to a l l c o w s as t h e y go dr y. T h e l a s t r e s o r t of c o n t r o l is to c u l l i n f e c t e d c o w s (as m u s t o f t e n be d o n e s i n c e c e r t a i n o r g a n i s m s c a n n o t b e o v e r c o m e by a n t i b i o t i c s ) . A n o t h e r f i e l d p r o g r a m is the m o n t h l y c o u n t i n g of s o m a t i c c e l l c o n c e n t r a t i o n of p o o l e d u d d e r m i l k of i n d i v i d u a l cows.

    W h i l e m a n y d a i r y m e n r e d u c e d i s e a s e f r e q u e n c y to a l o w l e v e l by t h e s e p r a c t i c e s , I a m n o t a w a r e of a n y d e f i n i t i v e r e p o r t s of r a p i d l y d e c l i n i n g m a s t i t i s r a t e s in n a t i o n a l p o p u l a t i o n s . T h e r e are a d d i t i o n a l d r a w b a c k s to t h e s e c o n t r o l m e t h o d s : (1) m i l ks u p p l i e s c a n b e c o n t a m i n a t e d b y r e s i d u e s (2) o v e r - u s e of a n t i b i o t i c s m a y l e a d to r e s i s t a n t s t r a i n s ( c u r r e n t c u r e r a t e s are g e n e r a l l y n o t v e r y h i g h ) (3) f a r m e r s m u s t c o n s t a n t l y r e p e a t r o u t i n e a c t i o n s .

    I m m u n i z a t i o n a g a i n s t m a s t i t i s h a s p r o v e d to be a n i n t r a c t a b l e p r o b l e m , a l t h o u g h i m m u n e r e s p o n s e is s u r e l y an i m p o r t a n t d e f e n s e m e c h a n i s m . O n e r e a s o n is the w i d e v a r i e t y o f c a u s a t i v e o r g a n i s m s . S u c c e s s f u l i m m u n i z a t i o n a g a i n s t o n e s p e c i e s is n o t l i k e l y to be r e w a r d i n g , e x c e p t f o r s p e c i e s u n r e s p o n s i v e to a n t i b i o t i c s . T h e r e is on e v i e w t h a t s a y s if one t y p e of o r g a n i s m is r e m o v e d in a s u s c e p t i b l e co w, it w i l l be r e p l a c e d b y a n o t h e r .

    T h e s e f a c t o r s s u g g e s t the p o s s i b i l i t y a n d v a l u e of g e n e t i c c o n t r o l . B h a t i n a n d G a n g u l i ( 1 9 7 5 ) r e p o r t e d t h a t v a r i o u s b r e e d s a n d t h e i r c r o s s e s d i d n o t d i f f e r in b l o o d c o n c e n t r a t i o n s of the v a r i o u s i m m u n o g l o b u l i n (Ig) c l a s s e s .     A b a s i c q u a n d a r y in s t u d y of g e n e t i c r e s i s t a n c e is t h a t the a n i m a l p o s s e s s e s a c e r t a i n i n h e r e n t a b i l i t y to r e s i s t i n f e c t i o n b e f o r e e x p o s u r e to t h e p a t h o g e n . O n e c a n e s t a b l i s h as a g o a l the a b i l i t y to d e t e c t or to m e a s u r e this i n h e r e n t r e s i s t a n c e c a p a b i l i t y b e f o r e e x p o s u r e . T h i s is a n o t h e r w a y to s a y t h a t w e w o u l d l i k e to k n o w the g e n o t y p e f o r r e s i s t a n c e (or the g e n o t y p e s f o r the v a r i o u s r e s i s t a n c e m e c h a n i s m s ) .

    T h e l i t e r a t u r e c o n t a i n s n u m e r o u s r e p o r t s of i n v e s t i g a t i o n s of " m a r k e r s " of r e s i s t a n c e ( e n z y m e s , u s u a l l y ) , b u t w e a r e far a w a y f r o m d e t e c t i n g r e s i s t a n c e c a p a b i l i t y . In p r a c t i c e , we f a l l b a c k u p o n r e t r o s p e c t i v e s t u d i e s of d a t a in w h i c h c e r t a i n a n i m a l s b e c a m e i n f e c t e d a n d o t h e r s d i d not. U s u a l l y we h a v e l i t t l e or no idea of w h a t r i s k or e x p o s u r e an a n i m a l e x p e r i e n c e d .

    It is t h e o r e t i c a l l y p o s s i b l e to e s t i m a t e g e n e t i c d i f f e r e n c e s a m o n g c o w s in a b i l i t y to r e s i s t by e x p e r i m e n t a l l y s u b j e c t i n g e a c h a n i m a l to a u n i f o r m a p p l i c a t i o n of l i v e p a t h o g e n s i n t o the u d d e r , a n d o b s e v i n g w h e t h e r i n f e c t i o n o c c u r s . In d a i r y c a t t l e w h e r e g e n e t i c i m p r o v e m e n t is m a i n l y t h r o u g h sire s e l e c t i o n , o n e w o u l d h a v e to a s s e m b l e p r o g e n y of m a n y b u l l s in o n e l o c a t i o n to c o n d u c t s u c h an e v a l u a t i o n . T h e r e p o r t of G r o o t e n h u i s J[1976) is the o n l y k n o w n a t t e m p t of t h i s k i n d ( o n l y 2 p r o g e n y g r o u p s ) .

    U l t i m a t e l y , o n e m u s t s e l e c t a m e a s u r e of m a s t i t i s w h i c h c a n be u s e d in a m i l k r e c o r d i n g s y s t e m , p r e f e r a b l y b a s e d o n a n a l y s i s of m i l k s a m p l e s . F r o m s u c h d e t e r m i n a t i o n s o n e m u s t j u d g e w h e t h e r the c o w w a s i n f e c t e d or n o t , k n o w i n g l i t t l e or n o t h i n g a b o u t e x p o s u r e to p a t h o g e n s .

    T h e m o s t d e s i r a b l e f i e l d m e a s u r e of m a s t i t i s is the t e s t i n g ot a s e p t i c a l l y c o l l e c t e d m i l k s a m p l e s for b a c t e r i a l p r e s e n c e .I d e a l l y , t h i s w o u l d be b y q u a r t e r , s i n c e e a c h q u a r t e r m a y h a v e a d i f f e r e n t s t a t u s ( a l t h o u g h m o s t i n f e c t e d c o w s h a v e o n l y o n e q u a r t e r a f f e c t e d ) . T h e d r a w b a c k to b a c t e r i o l o g i c d e t e r m i n a t i o n s is that c o l l e c t i n g s a m p l e s for b a c t e r i a l a n a l y s i s b y f a r m e r s or m i l k r e c o r d i n g p e r s o n n e l is n o t v e r y f e a s i b l e .     L a r g e s t u d i e s of b a c t e r i o l o g y a r e r a r e ( a l l c o w s m u s t be s a m p l e d , n o t j u s t s u s p e c t s ) .     T h e s e c o n d m o s t d e s i r a b l e c r i t e r i o n of i n f e c t i o n is the r e c o r d i n g of e a c h c l i n i c a l c a s e ( t r e a t m e n t ) of m a s t i t i s . O n e m i g h t h a v e e x p e c t e d t h a t b a c t e r i o l o g i c a l r e s u l t s w o u l d h a v e a h i g h e r h e r i t a b i l i t y t h a n c l i n i c a l , i n a s m u c h as t h e f o r m e r d e t e c t s s u b c l i n i c a l i n f e c t i o n s , w h i c h a r e t h e m a j o r i t y of c a s e s ( G i e s e c k e , 197 9 ) . Y o u n g et al. ( I 9 6 0 ) r e p o r t e d a low (. 29) g e n e t i c c o r r e l a t i o n b e t w e e n c l i n i c a l f r e q u e n c y a n d b a c t e r i o l o g y .

    T h e d r a w b a c k to u s i n g a c l i n i c a l m e a s u r e is t h a t it is d i f f i c u l t to g e t f a r m e r s to r o u t i n e l y r e c o r d t r e a t m e n t s , p a r t i c u l a r l y in l a r g e h e r d s . H o w e v e r , t h e s t r u c t u r e o f d i s e a s e c o n t r o l w o r k in some c o u n t r i e s m a k e s t h e r e c o r d i n g of c l i n i c a l m a s t i t i s f e a s i b l e . M o r e i m p o r t a n t l y , c o n s i d e r a t i o n of c e l l t y p e s a n d t h e i r r o l e s m a k e s it c l e a r that S C C at all t i m e s r e f l e c t s p r e s e n c e of c e l l s a c t i v e in d e f e n s e s ( m a c r o p h a g e s , P M N ) . In s e l e c t i o n o n e p r e s u m a b l y w o u l d aim to l o w e r SCC. H o w e v e r , f r o m t h e s t a n d p o i n t of d e f e n s e , an i n c r e a s e d n u m b e r of t h e p h a g o c y t i c c e l l s is d e s i r a b l e . T h e r e are m a n y r e p o r t s s h o w i n g t h a t h i g h e r c e l l c o u n t d e c r e a s e s r a t e of i n f e c t i o n to s u b s e q u e n t e x p o s u r e ( S c h a l m et a l ., 1971; P o u t r e l a n d L e r o n d e l l e , 1978; S c h a l m et al., 1 9 7 6 ) . T h i s s u g g e s t s o n e m u s t b e v e r y c a r e f u l n o t to d i s c r i m i n a t e a g a i n s t c o w s or p r o g e n i e s w i t h h i g h e r n o r m a l c o u n t s . S i r e s e l e c t i o n is l a r g e l y o n f i r s t l a c t a t i o n s w h e r e m a s t i t i s i n f e c t i o n r a t e is low.

    In a d d i t i o n , s t u d i e s s h o w t h a t S C C d i s t r i b u t i o n s of i n f e c t e d a n d u n i n f e c t e d c o w s o v e r l a p . S h o o k a n d M i l l e r ( 1 9 7 8 ) a n d L i n d s t r o m and S y v a j a r v i (19 8 1 ) f o u n d r a t h e r h i g h e r r o r r a t e s in c l a s s i f y i n g s i n g l e m i l k s a m p l e s as i n f e c t e d or u n i n f e c t e d . T h i s is n o t s u r p r i s i n g w h e n one c o n s i d e r s that n u m b e r of q u a r t e r s i n f e c t e d a n d s p e c i e s of o r g a n i s m s p r e s e n t v a r y g r e a t l y a m o n g c o m p o s i t e m i l k s f r o m i n f e c t e d c o w s . S p e c i e s h a v e v a r y i n g i m p a c t s o n SCC.

    W h i l e S C C s h o u l d b e v i e w e d c a u t i o u s l y as a n i n d i c a t o r of m a s t i t i s , i n f o r m a t i o n on m i l k r e c o r d e d c o w s m a y h a v e o t h e r us e s . S o m e m e t h o d s ( a n t i b i o t i c s ) m a y be d e c r e a s i n g in e f f e c t i v e n e s s .
