Introduction
Genevac evaporators are designed to combine rapid and safe evaporation with high capacity and a wide variety of sample formats. In order to optimise the evaporation performance achieved it is important to select the correct sample holder for your particular application.
Genevac sample holders are manufactured from solid high grade aluminium to very precise specifications and are mass balanced to ensure smooth running of the evaporator.
This guide will help you make the right choice.
The Range
This guide includes the current range of Genevac sample holders which are coloured bright blue for identification purposes. These sample holders are designed for use in an HT series II system, an EZ-2 series system, or may be used in either.
Due to height and weight limitations, a small number of holders have limited compatibility and are not suitable for use with certain systems. Any holder which is subject to a limitation of compatibility, it is highlighted within this brochure. An example of such a limitation is shown below:
Not for use with EZ-2 MK1
In this instance, the sample holder must not be used in an EZ-2 MK1. The EZ-2 Mk1 can be identified by the large circular window in the chamber lid, EZ-2 mk2 and EZ-2 Mk3 has a solid lid with a small tear drop window.
Blue sample holders are not interchangeable with the older range of holders and swings. To choose the correct new holder, refer to the: Sample Holder Part Number Cross Reference.
Should you require an old design of holder, a holder for use in a Genevac HT series 1 system, or a holder for a tube which is not listed, please contact your local Genevac representative or sales agent.

Minimum Order Quantities
The minimum order quantity for Genevac sample holders is two, larger orders should be for an even number (paired sets) of holders. This also applies to associated parts such as swings, heat transfer plates, SampleGenie assemblies etc.
Nomenclature
Tube holder A holder which accommodates tubes with rounded ends
Vial Holder A holder which accommodates vials with flat ends
Bottle holder A holder which accommodates bottles with flat ends
Flask holder A holder which accommodates round bottom flasks. Holders for tubes with conical or tapered ends are identified individually.
Heat Transfer
A major factor in determining the speed of evaporation is the rate at which heat can transfer to the sample. Greater heat transfer rates mean faster evaporation.
Here are two ways that Genevac HT series II and EZ-2 series evaporators optimise heat transfer:
• High quality aluminium rotors and holders. Recommended for its properties of high thermal conductivity and low weight, Genevac systems use aluminium sample holders which have particularly efficient heat transfer properties.
• High quality engineering. Genevac samples holders are manufactured to very precise specifications. Tubes fit very snugly into the tube holders as any gaps would slow down the rate of heat transfer. This is why it is important to provide us with a sample of tubes when placing orders for sample holders.
Protective Coatings
Standard aluminium sample holders are Hard Anodised (water sealed) for protection against all normal organic solvents, including strong acids such as TFA, HCl and thionyl chloride.
HCl or Thionyl Chloride
To evaporate HCl or thionyl chloride, it is necessary to purchase a system (comprising of: evaporator, condenser and pump) which is HCl and thionyl chloride resistant. These systems are specifically built to resists attack from HCl and thionyl chloride.

Glassware Labels
There are two common methods of labelling glassware in laboratories; both can cause problems in the evaporator.
Paper or film labels The label effectively increases the tube diameter and the problem is exaggerated if the label overlaps itself. The tube or vial no longer fits snugly into the holder, high spots cause stress in the glass and compromise efficient heat transfer from the holder.
Ceramic Etched Ceramic labels etch the glass and may weaken it. Genevac recommend testing new tubes for breakage before buying large numbers.
Sample Capacity
Genevac uses the latest computer aided design technology to ensure evaporators have maximum internal capacity within a minimal instrument footprint. This means high throughput evaporation is possible from an instrument taking up a small amount of bench top space.

Genevac offers a comprehensive range of sample holders. Some are designed for use with swings, others are one piece holders which fit straight onto the evaporator rotor. Maintenance
Inspect and clean the sample holders at least monthly. Always dry the sample holders thoroughly after cleaning, never use wet sample holders in a Genevac system.
The following inspection routine is mandatory following tube breakage or solvent spillage.
Inspection
Visually inspect the swings and sample holders and clean off any debris, especially in the sample holder wells. High points, caused by debris, lead to stress in the glassware and may result in breakage.
The most common cause of repeat glassware breakage is a fragment from a previously broken tube. Solvent can stick glass fragments to the holder making it difficult to remove. Any residual solvent or sample material must be cleaned off.
Superficial surface damage (scratches) do not affect the performance of a sample holder. However, if there is any structural damage (if any part of the sample holder is bent or deformed) do not use it. Contact Genevac Service for evaluation.
Repeated use with strong acids is likely to cause colour fading of the sample holder over time. This is normal and does not affect the integrity of the holder.
Cleaning
Loose dirt and debris can be removed using a brush, a compressed-air line may also be used if this is available. Adhered dirt, debris or sample / solvent residue should be cleaned off using methanol or acetone, soak the sample holders if necessary. The sample holders should then be washed in clean water and fully dried before use.
Tube Breakage
The precision design of all Genevac sample holders contributes, not only to their excellent heat transfer properties and superior evaporation performance, it also ensures that tubes have maximum protection from breakage during evaporation. For additional protection against glassware breakage, some flask holders and some tube holders are designed for use with thermal cushions. Thermal cushions are made from a filled polymer with excellent heat transfer and cushioning properties.
Use only borosilicate tubes with a wall thickness greater than 1.2 mm in Genevac evaporators. Do not use old, damaged or scratched glassware. In laboratories where tubes or vials are reused, each tube or vial must be regularly inspected for damage. Discard any scratched or cracked items.

Microplate Heat Transfer Plates
Heat transfer plates are available for use in open swings, side bridge swings, and most Fast Stack holders.
This range of sample holders provide improved evaporation results when using deep well microtitre plates. Heat Transfer Plates are designed to support the wells of the plate, fitting precisely to the base of the wells, and where possible, to the sides of each well.
Heat transfer plates confer three main benefits:
• Excellent heat transfer rates, up to 50% reduced evaporation times when using micro plates.
• Even sample drying
• Support for micro plates when used in open swings, preventing potentially harmful direct heating by lamps, and preventing plate deformation by heat and G-force.
Heat transfer plates must be used when using microtube cluster racks, e.g. Micronics or ABgene racks, in Genevac open swings.

Gilson and Waters Fraction Collection Racks
Genevac offer a range of innovative options that help interface fraction collection systems directly to the Genevac HT series evaporator. Currently Gilson 215 and Waters 2767 systems are supported.
To interface with the fraction collections system, Genevac provide a simple sample holder carrier which fits directly on to the fraction collection bed. Genevac sample holders fit into the carrier to make up a complete fraction collection rack. Once fractions are collected, the sample holders can be placed directly into the Genevac evaporator with no need to label, mark or manipulate individual tubes. Sample holders can be marked to indicate their location within the fraction collection rack.
There are a range of options, some use standard sample holders, some use specially designed holders which keep the spacing between tubes uniform, and reduce fraction collector reprogramming. Waters 2767 users will require the adaptor plate when using 16 mm diameter tubes.

SampleGenie
Designed for situations where large volumes need to be dried or concentrated into a small vial. SampleGenie™ comprises sample tube and sample holder technology which enables large volume samples to be dried or concentrated directly into a variety of small vials. Where HPLC purification fractions are used, fractions are combined into one tube and and dried or fast lyophilised into the final vial.
