ABSTRACT
Introduction Proliferation activity of keratinocyte, and skin hydration decreased with
aging process. Aging skin looks dull, dry, and sagging. Aquaporin 3(AQP3) is a protein thatplayed a a!or role in cells proliferation and skin hydration. AQP3 e"pression decreased
with aging, therefore AQP3 is a key protein as a novel #target$ for anti%aging treatent.
&etinoic acid (&A) was coonly used in treating photoaging and intrinsic aging, 'ut it
frequently showed any harful side effect including skin irritation. Centella asiatica and
chitosan are natural product 'eing used in wound healing and anti aging cosetic. Chitosan
nanoparticle has a potential activity as an active agent and as a carrierdelivery syste.
Aim he ai of our study is to copare and analy*e the effect of Centella asiatica
+thanolic +"tract encapsulated in Chitosan anoparticles (CA++-CP) and &A induced
proliferation activity and the e"pression of AQP 3 in oral uan +pideral
/eratinocytes (+/).
Methods 0icroculture etra*oliu est (0) was conducted to analy*e the
proliferation of keratinocytes, after treated with CA++-CP and &A in several dosages for
12, and 2 hours. 4unocytocheisry e"aination using anti%aquaporin anti'ody
a'516157 was conducted to e"aine the e"pression of AQP3, then quantitatively analy*ed 'y
4age%8 software.
Results After 12 hours incu'ated CA++-CP, showed increased proliferation
activity of keratinocytes, optial dose at 3.516 g9, and the e"pression of AQP3 at :,16
g9, while &A increased the e"pression of AQP3, optial dose at 5 g9 after 12 hours.
Conclusion CA++-CP increased the proliferation activity and the e"pression of AQP3
in the keratinocyte. ;urther clinical trial required in the future to evaluate the skin hydrationafter topical application.

Tu$uan uuan !enelitian kami adalah untuk membandin(kan dan men(analisa e/ek
ekstrak ethanol Centella asiaticadalam nano!artikel kitosan 3++7A8NP0 dan asam retinoat
3A6 untuk men(induksi akti*itas !roli/erasi dan eks!resi A4P2 !ada keratinosit e!idermis
kulit manusia 30+09.
