    10.1. Ngoi nc (phn tch, nh gi tnh hnh nghin cu thuc lnh vc ca ti trn th gii,lit k danh mc cc cng trnh nghin cu, ti liu c lin quan n ti c trch dn khi nhgi tng quan)

    Vi to l cc sinh vt n bo, sinh trng bng quang t dng nh quang hp hoc d dng,hoc c hai (Liam Brennan, Philip Owende, 2009). Vi to c tc sinh trng nhanh, cho sinh khinhiu, din tch nui trng nh, tr lng du ln, khng cnh tranh t nng nghip, khng i himi trng nc sch, thm ch chng cn sinh trng trong mi trng nc thi nn thn thin vimi trng. Trung bnh 1kg sinh khi to th tiu th c 1,83kg CO2 (Attilio Converti, 2009).

    Nm 2003 Hanhua Hu, Kunshan Gao nghin cu v sinh trng v thnh phn axit bo caNannochloropsis sp. vi cc ngun cacbon khc nhau. Kt qu cho thy mc sc kh 2% CO2kch thch sinh trng ca N. oculata ln nht.

    Nm 2006 Hanhua Hu, Kunshan Gao tip tc nghin cu v sinh trng v thnh phn axit bot do vi cc nng CO2 khc nhau. Kt qu nghin cu cho thy hm lng CO2 l rt quantrng. Trong cc nng CO2 kho st th nng 2% CO2 chnh l nng im ti u nui cyN. oculata. Tuy nhin loi ny c tri qua qu trnh hun luyn thch nghi vi nng CO2 2%trc, sau nui cy trn mi trng c nng CO2 cao vn sinh trng kh tt. V vy c thkt hp s nui cy N. oculata ly lipid sn xut nhin liu sinh hc v hp th kh CO2 ci thin mitrng.

    Nm 2009, Sarmidi Amin nghin cu sn xut kh v du sinh hc t vi to. Kt qu cho thy snphm t vi to c c tnh tng t nh cc sn phm t du thc vt v du c, c th thay th ccho du m.

    Nm 2009, Attilio Converti v cng s nghin cu v nh hng ca nhit ti sinh trngv hm lng lipid ca N. oculata v Chlorella vulgaris. Kt qu nghin cu cho rng N. oculata

    thch nghi trong mt ph nhit kh rng nhng li nhy cm khi sinh trng ti cc nhit khcnhau, nhit thch hp 20-250C. Nu nhit b h thp hoc b nng cao th sinh trng ca loito ny b hn ch.

    Nm 2009, Sheng Yi Chiu v cng s nghin cu nh hng ca nng CO2 trong dngkh b sung vo mi trng nui cy ln sn lng sinh khi v s tch ly lipid N. oculata. Ktqu cho thy mc sc kh 2% CO2 kch thch sinh trng ca N. oculata cao nht.

    Nm 2011, Subhash Nigam nghin cu nh hng ca nng nitrogen ti sinh trng vhm lng lipid thu c.. Kt qu cho thy khng ch thiu ht Nit m d tha qu nhiu Nit(dng NH4Cl) cng c ch sinh trng.

    T nm 1983, mt trm thc nghim sn xut thm canh v lin tc Spirulina platensis cxy dng Bnh Thun. Sn xut bn cng nghip loi to Lam ny cho 5 tn bt to Spirulinakh trong 9 thng, tng ng vi 7-10g cht kh/m 2/ngy hoc khong 20 tn/ha/nm (NguynHu Thc, 1987)

    Nm 2005, mt s tnh pha Bc pht hin v phn lp c Labyrinthula c hm lng DHAcao (Hon Lan Anh v cng s,2005)

    Nm 2006, Hong Th Ninh Hin v cng s nghin cu iu kin nui cy ti u chng vito binLabyrn thula sp. trn mi trng rn.

    Nm 2010, Nguyn Th Minh Thanh v cng s cng b v sng lc cc loi vi to bin quangt dng c phn lp t vng bin ca Vit Nam nhm lm ngun nguyn liu cho sn xut nhinliu sinh hc.

    Gn y, nm 2012 TS Nguyn Th Thanh Xun ging vin trng i hc Bch Khoa, ihc Nng c nhn gii thng LOral Unessco v s pht trin ca ph n trong khoahc vi ti : Nghin cu m hnh nui trng vi to to ngun sinh khi ng dng trong sn xutnhin liu sinh hc.

    Cho ti nay, trong nc c nhiu nghin cu v nui vi to ly sinh khi nhng ch yu tptrung vo cc mc ch: lm thc n cho lun trng ca cc n v nui trng thy sn, lm thc n

    cho vt nui, xut khu, lm thc n b dng cho ngi v lm m phm.Nghin cu nui vi tonhm thu lipid lm nguyn liu sn xut nhin liu sinh hc (NLSH) cn hn ch.

    Sn xut NLSH ang c c th gii quan tm. Chnh ph Vit Nam k n 117 pht trinNLSH mt dng nng lng mi ti to c thay th mt phn NLHT truyn thng, gp phnm bo an ning nng lng v bo v mi trng.

    Ngun sn xut NLSH c th ly t m ng vt, du n thi, du ca cc cy c du. Khi sdng nhng nguyn liu ny gp phi cc hn ch sau:

    nhm thu lipid sn xut NLSH c cc u im sau:- Sn xut thu sinh khi ln trong thi gian ngn nn tit kim chi ph.

    - Ch ng v ngun nguyn liu do tc sinh trng ca vi to nhanh.

    - Khng i hi din tch t trng nn khng nh hng ti ngnh nng, lm nghip.

    Trong cc loi vi to bin th N. oculata l mt trong nhng loi vi to cho hm lng lipit cao(khong 22,7 29,7%). Mc d hm lng lipid ca N. oculata thp hn mt s loi nh Chlorellaemersonio, Chlorella sp.,Nannochloris sp.,. NhngN. oculata theo phng php t dng c scsinh sn cao, lng sinh khi trong ngy ln (Teresa M. Mata, Antnio A. Martins, Nidia. S, 2009 vAttilio, 2009). Hn na, nng sut lipid ca N. oculata c th thay i theo iu kin nui cy(Sukenik A., Carmeli Y., 1989). iu ny c ngha l N. oculata ang l mt tim nng v hmlng lipid nu tm c iu kin nui cy ph hp.

    Hin nay Vit Nam cha c nghin cu no i su vo nghin cu ti u ha cc iu kin nuicy vi to bin nhm thu lipit cung cp nguyn liu cho sn xut NLSH, c bit loi c tim nngnhN. oculata.

    14. CCH TIP CN, PHNG PHP NGHIN CU

    14.1. Cch tip cn:

    - Nui cy vi toNannochloropsis Oculata trn cc iu kin dinh dng khc nhau. La chniu kin ti u cho sinh khi ln nht vi hm lng lipid cao nht.

    - a ra iu kin ti u trn mi trng lng nui Nannochloropsis Oculata vi mc ch thulipid.

    14.2. Phng php nghin cu:

    Ni dung 1:Ti u ha mt s iu kin nui cyN. Oculata vi mc ch thu lipid

    a. B tr th nghim

    Nng sut lipid c tnh l khi lng lipid c sn xut ra trn mi n v th tch canhtrng vi to mi ngy, ph thuc vo tc sinh trng ca to mi ngy v hm lng lipid ctrong vi to. Hm lng lipit trong t bo li ph thuc vo cc yu t nh: ngun Nit, nhit , mn... kho st cc yu t ny chng ti b tr th nghim nh sau:

    Cc l nui cy c nghin cu c lp, mi l gm 5 bnh tam gic dung tch 1,5l c b trnh s di y (hnh 1). Cc l khc nhau v yu t kho st. C th nh sau:

    - L 1: Kho st v nhit . Nhit tng ng trong cc bnh nui l: 18, 20, 25, 28, 300C.

    - L 2: Kho st v mn. Nng kho st trong 5 bnh tng ng t: 0; 1%; 2%; 2,5%; 3%;4%

    - L 3: Kho st v cng chiu sng 2000, 2500, 3000, 3500, 4000, 4500 (lux).

    - L 4: Kho st v ngun Nit. ngun Nit kho st trong 5 bnh tng ng: (NH4) 2(SO)4,NH4NO3, NaNO3, ure, N-P-K

    Ni dung 2: Thu sinh khi v xc nh hm lng lipid

    a. thu sinh khi: Sau khi xc nh c thi im thu sinh khi bng phng php o quang tinhnh thu sinh khi bng ly tm hoc dng cht kt ta phn nhm. Sinh khi c ra li vi ncct 3 ln ri em sy kh 800C nghin thu c sinh khi kh. Phn nc lc c ti sdng cho cc nui cy tip sau.

    b. Xc nh hm lng lipid theo phng php Soxlet:

    - Vic xc nh hm lng lipid s c tin hnh i vi mi bnh nui cy trong mi l. Ktqu c ghi nhn c lp phn tch v so snh.

    - Cc bc tin hnh: cn chnh xc 5g mu sy n khi lng khng i 1050C chuynmu vo ti giy c sy n khi lng khng i cn li xc nh khi lng ca ti vmu t gi mu vo bnh chit chit t 6-8h vi dung mi chit l ete etanol ly gi mu rav cho bay hi ht dung mi sy kh n khi lng khng i.
