PE-Cy7

PE-Cy7* is a tandem fluorochrome composed of R-phycoerythrin(PE) (excited by the 488-nm line of a laser and serves as an energydonor), coupled to the cyanine dye Cy7™ (acts as an energyacceptor and fluoresces at 785 nm).1 PE-Cy7-labeled antibodiescan be used with FITC and PE-labeled reagents on single-laserflow cytometers with no significant spectral overlap between PE-Cy7 and FITC.1

APC-Cy7

APC-Cy7* is a tandem fluorochrome composed ofAllophycocyanin (APC) (excited by laser lines from 595 to 647nm and serves as an energy donor), coupled to the cyanine dyeCy7™ (acts as an energy acceptor and fluoresces at 785 nm).1,2

Alexa Fluor® 488

Alexa Fluor® 488 is a green fluorescent dye with a fluorescencespectrum almost identical to that of FITC, with excitation/emission maxima of 494/519 nm and a fluorescence lifetime of~4.1 nanoseconds. Alexa Fluor® 488 can be analyzed in the sameinstrument channel as FITC.

Alexa Fluor® 647

Alexa Fluor® 647 can be excited by long-wavelength excitationsources, such as a red HeNe laser (at 633 nm), a krypton-ion laser (at 647 nm), or laser diodes. The fluorescence quenching isminimal, and Alexa Fluor® 647 can be analyzed in the sameinstrument channel as APC.

Figure 2. Expression of IFN-γ by stimulated human peripheral blood lym-phocytes. Human PBMCs were stimulated for 4 hrs with PMA (5 ng/ml,Sigma, Cat. No. P-8139) and Ionomycin (500 ng, Sigma, P-8139) in the pres-ence of Brefeldin A (BD GolgiPlug™, Cat. No. 555029). Cells were har-vested, fixed, permeabilized, and stained with anti-human CD8-PE (Cat. No.555367) and either anti-human IFN–γ-PE-Cy7 antibody (Cat. No. 557643)(Panel A), or immunoglobulin isotype control-PE-Cy7 (Cat. No. 557646)(Panel B). (Donor 100)

Figure 3. Expression of IFN-γ by stimulated human peripheral bloodlymphocytes. Human PBMCs were stimulated for 4 hrs with PMA (5 ng/ml,Sigma, Cat. No. P-8139) and Ionomycin (500 ng, Sigma, P-8139) in thepresence of Brefeldin A (BD GolgiPlug™, Cat. No. 555029). Cells wereharvested, fixed, permeabilized, and stained with anti-human CD8-PE-Cy7 (Cat. No. 557746) and either anti-human IFN–γ Alexa Fluor® 488 (Cat.No. 557718) (Panel A), or immunoglobulin isotype control-Alexa Fluor®488 (Cat. No. 557702) (Panel B). (Donor 200)

Figure 4. Expression of IL-4 by stimulated CD4+ BALB/c spleen cells.Splenocytes from BALB/c mice were enriched for CD4+ cells by positiveselection using anti-CD4 coated plates (GK1.5, Cat. No. 555335)and either APC-Cy7-conjugated mouse anti-human CD195 (2D7/CCR5,Cat. No. 557755) (Panel A) or an APC-Cy7-conjugated mouseimmunoglobulin isotype control (MOPC-21, Cat. Figure 6. Multicolor analysis of human peripheral blood mononuclearcells stained with anti-human-CD3, anti-human-CD4, and anti-phospho-p44/42 (T202/Y204) Alexa Fluor® 647. PBMCs were depleted of adherentcells and stimulated by CD3/CD28 crosslinking (15 min at 4°C) orPMA/Ionomycin treatment (500 ng/ml for 30 min at 37°C). Followingtreatment, cells were washed, fixed, permeabilized, and stained with allthree antibodies simultaneously. Phospho-p44/42 (T202/Y204) was up-reg-ulated in only the CD3+/CD4+ double positive cells.

BD™ Phospho-Specific Solutions

Introducing BD™ PhosFlow Reagents

Direct fluorescent conjugates for multiparameter, phospho-specific flow cytometry. Now the company synonymous with flowcytometry brings you the finest set of anti-body conjugates for the study of proteinphosphorylation.

BD Biosciences Pharmingen is developingan extensive line of phospho-specific antibodies for multi-color flow cytometry.Featuring the high quality BD TransductionLaboratories™ brand of antibodies, conju-gated to Alexa Fluor® dyes and PE, thesereagents allow you to study severalparameters simultaneously in 3-, 4-, or even 6-color experiments.

• Study multiple phosphorylationevents in a single experiment

• Evaluate the effects of proteinphosphorylation in complex cellpopulations

• Monitor intracellular kinases inconjugation with surface markers

• Study the phosphorylation ofERK1/2, p38 MAPK, Stats 1, 3, 5,and 6, and more
