Issue 1 www.esn-online.org Nematology News 33 rd Symposium of ESN, Ghent 2018 Registration now open! @ESNematologists a For the third time in ESN history the medieval city of Ghent welcomes nematologists for its biennial symposium (9-13 th September 2018). When we launched the idea of organising the 33 rd ESN symposium, memories from 1994, the previous time in Ghent, were shared with us. Vivid memories of a splendid symposium means we now have a challenge to try to do better. Since then Ghent has changed a lot. Historic buildings have been restored, the bar and restaurant scene gets high ratings in travel guides and the historic city centre is now one of Europe’s biggest pedestrian zones. In 2017 Belgian beer culture was acknowledged as a cultural world heritage by UNESCO. With Ghent University and the Flemish Research Institute for Agriculture, Fisheries and Food (ILVO) the Ghent region can also be considered as a hotspot for nematological research and education. Both institutes will jointly organise the 33 rd ESN symposium. Our team is enthusiastic and can also rely on the students of the International Master of Science in Agro- and Environmental Nematology, a unique master programme of Ghent University. We will create the atmosphere but we count on your contribution, both scientifically and socially, to make ESN2018 a success. ESN Braga: 28th August - 1st September 2016

ESN 2016 will take place at The University of Minho, Braga, Portugal (http://www.uminho. pt/). The main campus building will host the posters (which will be available for the full meeting), tea and coffee breaks and the trade stands as well as parallel sessions in four adjoining lecture theatres. This venue will be the focal point for the meeting and is a 10 minute walk from the of-ficial hotel of the symposium. Braga, with over 175,000 inhabitants, is one of the oldest cities in Portugal and is located in the North of the country approxi-mately 30 Km from the sea. It is close to the mountains of the Minho Region and very near to the National Park of Peneda-Gerês. Braga is 40 Km from Oporto international airport. This is served by many international airlines, including budget airlines. Options for travelling between Oporto and Braga include train, bus or taxi. Further information, including details of bursaries for ESN student members, will be provided in the next edition of this newsletter.

Dr.SaraSánchezMorenois a doctoronBiology by the UniversityofAlcalá, Spain.Shedeveloped her PhD at theNatural History Museum inMadridon nematodeecology inpollutedareasandconductedpost-doctoral researchattheUniversityofCalifornia at Davis, focusing on the role of soil nematodes in soil functioning inagroecosystems. Caves andcave-dwellingbiota havefascinatedscientistsforcenturies butthere is a lackofinformationonsubterraneanrealmsandtheecosystems theyhost.Nematodesplayanimportant rolein thefunctioning ofepigeanecosystems butwhether the sameistrue for subterranean ecosystems remains unknown. In a Forum article entitledNematodes in caves: A historical perspective on their occurrence, distribution andecologicalrelevance(pp.627-644),DuPreezetal.conductedanin-depthreviewonallreports relatedtocave-dwellingnematodes. Aliterature survey of41 scientificworksfromoverthelast138years revealed295uniquetaxa reportedfrom78differentcavesystems.The authors discussthe historical trends incavenematology,peculiarfindingsfrom important studies, and an ecological classification system. Lastly, the trophicdistributionofthe reportedtaxa ispresented,whilstnematodesfromother(non-cave)subterraneanenvironmentsarealsoconsidered.

Potatois thefourthmostimportantfoodcropinthe worldafter corn,rice andwheat.It isessential tocontroland identify cyst nematode species, such asG. pallida, G. rostochiensis andH. schachtii. Next GenerationSequencing(NGS)has extendedthenumberofgenomic loci thatcannowbeusedformolecular identification

and,inparticular,NGShasincreasedopportunitiesforeasiermicrosatelliteisolation.Inapaper entitled Development and validation of real time PCR assays based on novelmolecularmarkersforthesimultaneousdetectionandidentificationofGlobodera pallida,G.rostochiensis andHeterodera schachtii (pp.789-804)Gamel etal.screenedhundredsofmicrosatelliteprimercombinations inG.pallida,G.rostochiensisandH.schachtiiandcombined the most interesting ones into a single real time PCR assay. The newlydesignedprimersandprobes enabledthedetectionofall targetpopulations testedandwithnocross reactionforcloselyrelatednon-targetspecies (55populations tested).Theauthors determined thelimit of detectionas one juvenile forG. rostochiensisandG.pallida andfive juveniles forH. schachtii. For monitoringpotato cyst nematodes, thisanalytical tool would extend thenumber of cyst investigatedas five juveniles can bedetectedamong50cystsina sample.Inaddition,thismultiplexassaydetects DNAofthe

threetargetedspeciesintemplate DNAobtaineddirectly from floatmaterial after nematodeextractionfromsoil.

Transmissionelectronmicroscopy is essentialforstudyingtissues andcells ofnematodes,andrapidfixationusing cryomethods is indispensable for high resolution and advanced approaches, includingimmunocytochemistry, tomography and 3D reconstruction, quantitative assessments of cytoplasmic

elements, and correlative light and electron microscopy. In a paper entitled Self-PressurisedRapidFreezing(SPRF):aneasy-to-useandlow-costalternativecryo--fixationmethod for nematodes (pp. 871-881), Claeys et al. evaluated SPRF based on acomparativeanalysis oftheultrastructure ofspermatozoa ofthe nematodesAcrobelescomplexus and Caenorhabditis elegans. Sealed copper tubes, packed with activenematodesinwater,wereplungedintonitrogenslush, a semi-solidformofnitrogen.Thewater inside thecapillary copper tubeexpandsuponcoolingdue tothe formationof hexagonal ice, thereby generating highpressurenecessary for cryo-fixationof thesample.Forspermcells cryo-fixedby SPRF, the preservationoftheultrastructurewascomparabletothatachievedwithhighpressurefreezing.SPRFfixationdidnotdestroyantigenicity,basedontheresults oftheimmunolocalisationofthemajorspermprotein.The authors conclude that SPRF is a low-cost alternative cryo-fixation method for

nematodes.

Plant-parasiticnematodes ofthe familyHeteroderidaearedividedintotwomajorgroups: cyst(femalebodyturns intoa hard-walledsacor cyst)andcystoid (femalebodydoes notturnintoahard-walled cyst) nematodes. Whereas the cyst nematodes have been studiedextensively, the cystoid nematodes have received less attention. In Molecularcharacterisation and phylogenetic relationshipsofcystoid nematodesof the familyHeteroderidae (Nematoda: Tylenchida) (pp. 1065-1081) Subbotin et al. collectednematodepopulationsofspecies belongingtosomeofthecystoid nematodegenerafromdifferent locations intheUSA,Vietnam,GermanyandRussia.Thepopulationsof cystoidnematodesrepresentedsevenvalidspecies, twounidentifiedspecies ofAtalodera,sixunidentifiedspecies ofCryphodera,andthreeputative newspecies ofRhizonemella. Phylogenetic relationships within the family Heteroderidae werereconstructed basedontheD2-D3 of 28S rRNA, ITS rRNA andmitochondrial COIgenesequences andtheauthorsdiscuss problemsoftaxonomyandphylogeographyofcystoidnematodes.

MoleculartaxonomyandDNAbarcodingprovidea powerful tool forthe identificationofnematodes butsomesequence data appear to be incorrect, with faults ranging from sequence errors over misassemblies to

mislabelled,unlabelled,andmisidentifiedsequences.In a paperentitledThepitfallsofmolecular species identification: a case study within the genus Pratylenchus(Nematoda: Pratylenchidae) (pp.1179-1199) Janssenet al. showthat publishedITSsequences ofPratylenchus goodeyi areactually sequences from distantly free-livingbacterivorous ‘cephalobids’.This incorrect labellingresultedin acascadeoferroneousinterpretations, as shownby the reports of ‘P. goodeyi’ onbananain Chinaand oncotton in India. This clearly illustrates the risk of mislabelled sequences in publicdatabases. Theauthors discuss othermislabelledPratylenchus cases todemonstratethatthis isnotanisolatedcase.Assequence-basedidentificationisgrowingrapidlythehighlightedproblemmayyetincreaseanda stronglinkbetweenmorphologyandDNAsequences will be of crucial importance in order to prevent, or at least minimise,sequence-basedmisidentifications.

ObituaryDieterSturhan(1936-2017)

Dr Dieter Sturhan, nematologist at the InstituteofNematology and VertebrateResearch of the former Federal Biological Research Center for Agriculture andForestry(BBA)passedawayinMünster,Germany,on29November,2017.

Dieter Sturhanwas born in1936 inMeerbeck,Schaumburg-Lippe,Germany.Hestudiedzoology, botany andgeography at theUniversities of Kiel, ErlangenandMunich.Hereceivedhisdoctoratein1962fromProf.Dr.med.H.J.StammerattheZoological Institute oftheUniversityofErlangen-Nurembergwithathesis entitled“Onthesystematics,biologyandecologyofLongidorinae (Nematoda,Dorylaimoidea)withspecial referencetoLongidorusmaximus”.The practicalpartofhis doctoralthesis wasperformedbyDieterSturhanfrom1958-1960attheBavarianStateInstituteforPlantProductionandPlantProtectioninMunich.

Fromhisyouthonwards,Dieter Sturhanwas apassionatenaturalist.As a teenager, he spentmostofhis freetimebirdwatching. At theageof19, he publishedhis firstscientificpaper entitled"Observations oftheringouzel (Turdus torquatus) in the autumn of 1954 inMeerbeck at Stadthagen" in Beiträge zur NaturkundeNiedersachsens (Contributions to the Natural History of Lower Saxony), Issue 8, pages 60-61. Ten moreornithological publications followed before he published his first nematological work, "Appearance ofnematodesonstrawberriesinBavaria"in1960thejournalPflanzenschutz.

OnOctober1,1962,DieterSturhanbeganworkingasa researchassistantatthe BBAinMünster.Firstemployedona DFGresearchproject,he movedshortlyafterwards toapermanentpositionandwas appointedin1973toSenior Scientist and in 1978 to Scientific Director. His research focused on the taxonomy, ecology anddistributionofnematodes.MajorresearchinterestsincludedtheraceprobleminDitylenchus dipsaciaswell astaxonomicworkoneconomicallyimportantnematodetaxa,inparticularcystnematodes (Heterodera)androot-knotnematodes(Meloidogyne). DieterSturhanalsoworkedonnematodesas bioindicatorsforwaterquality,the distribution of entomopathogenic nematodes (Steinernema, Heterorhabditis) in Germany, and theimportanceofnematophagousfungiandbacteriaasantagonistsofplant-parasiticnematodes.

Histaxonomicstudies tookhimtothe Canaries,Madeira,Azores,Iran,DominicanRepublic,Nicaragua,Vietnam,Egypt andNewZealand.However, his biggest travel activitywas concentratedonGermany. His goal was todocumentthedistributionofplant-parasiticnematodes inGermany. Theplanwas tostudy at least three soilsamples per10 km² quadrant, oneeachfromarablefields, grasslandandforest.Many family vacations weresubordinatedtothishighergoal.The resultingdataformedthebasis oftheGermannematodecollection,whichwasfoundedbyDieterSturhanandtoday includes morethan5,000permanent slides (including800types)ofabout1,000speciesfromabout250generaofabout4,000samplingsites.

Starting in 1980 Dieter Sturhan accepted a teaching assignment at theUniversity of Münster on "Appliedzoology with special consideration of animal pestson cultivatedplants", which involved the supervisionofdiplomaanddoctoral students.Dieter Sturhandevelopedidentificationkeys forall plant-parasiticnematodesoccurringinGermanyandeveryyearhe conductedworkshopsonnematodeidentificationfortheemployeesofthe plant protection service. He acted as ‘final reference’ for nematode identification in any request bycolleagues of the official plant protection services. Dieter Sturhanwas amember of several national andinternational societies and from 1980-1987 he coordinated the working groupNematology of the GermanScientificSocietyforPlantProtectionandPlantHealth.

DieterSturhanpublishedhisresearchresultsin299scientificpapers,ofwhich79paperswereproducedafterhis retirement. His last papers appeared in 2016, "On the presence or absence of phasmids inmales ofHeteroderidae (Tylenchida)" in Nematology and a comprehensive review on "Nematodes of the orderTylenchida inGermany–thenon-phytoparasiticspecies"inSoil Organisms,co-authoredbyKarinHohbergfromtheSenckenbergMuseumofNaturalHistoryinGörlitz.

Intotal,DieterSturhanproposed1family,1subfamily,12generaand76speciesofplant-parasiticnematodes,including11 species inthe economically importantgenus Heterodera, sixspeciesinthegenusXiphinemaandfour species inthegenusLongidorus.Healsodescribeda nematophagousfungus (Hirsutella heteroderae=H.rhossiliensis)andperformed44synonymisations orassignments ofindividual species toothergenera.Atotalofsix nematode species were named after him (Hemicycliophora sturhani, Longidorus sturhani, Oriverutussturhani,Ogma sturhani,Axonchiumsturhani andOxydirus sturhani).In2001,DrDieterSturhanwas honouredasFellowoftheSocietyofNematologists.

Until his lastdays, DieterSturhanworkedontaxonomic issuesinclosecontactwithtaxonomic expertsfromaround the world. The latter will continue his work, being aware that with Dieter Sturhan an almostinexhaustiblesourceoftaxonomicexpertiseinthefieldofnematologyisnolongeravailable.

DieterSturhanwas aninternationallyrenownednematologistbut,above all,hewasa committedfellowcitizen,great colleagueanda wonderful family person.Wewill always havegoodmemories ofDieter Sturhan. Hisrelativeshaveourcompassion.

ICN2020:ContinuingatraditionwecanallsupportThe nematology congresses have always been universal in scope, gatheringscientists from countries spanning the tropics, subtropics, and temperateregionsonbothsidesofthe equator.Thediversevenues ofthepast-Canada,Netherlands, Guadeloupe, Canary Islands, Australia, South Africa – reflect atraditionofmakingthesecongressesareunionoftheentire nematology family.This kindofspecial ‘family’ traditionisreasonenoughto lookforwardtothenextcongress in2020.Buttheterrificopportunities affordedbythenextvenueandprogramarealsowhyall ofusshouldbe sure notonlytoattend,buttohelpinourvariouswaystosupportICN2020.
