Instructions to lab:

This assay detects all known sarbecoviruses sequences including those representing Human Strains SARS-COV-1 (2003 outbreak), SARS-COV-2 (2019-2020 outbreak associated with COVID-19), and all animal related sarbecoviruses including those found in Bats, Civets and Pangolins. Information from these tests can be used as a data point in a study or to confirm the presence or absence of an infectious agent, but not for diagnosis of disease; refer to the CDC guidelines for further details. These tests are intended to be used only for the purposes of respiratory virus surveillance and research. The recipient agrees to use the results in compliance with all applicable laws and regulations. Every effort has been made to assure the accuracy of the testing, but Charles River cannot provide any warranty regarding their accuracy. Please note that the following sample are not accepted: - Human Samples - Samples collected from research animals inoculated with sarbecoviruses or knowingly infected with sarbecoviruses - Research reagents, cell lines or other biologics that knowingly contain sarbecoviruses - Any live animals Sample Specifications and Shipping Instructions Samples that represent materials collected directly from research animals which are considered a higher susceptibility risk for Sarbecoviruses (e.g., nonhuman primates, ferrets, bats) will be processed in our BSL-2 laboratory. - Respiratory Track: Because sarbecoviruses are primarily respiratory viruses, the highest concentration of shedding virus is expected to be the respiratory tract. - Oral and nasal swabs: Swabs should be placed in a 5 mL collection tube. When collecting oral and nasal swabs, remove swab shafts before placing the swab tip into the tube to improve assay sensitivity. Never use calcium alginate swabs for PCR sample collection. Swabs can be shipped at room temperature or on ice packs. - Bronchial wash: 200-500 uL total of bronchial wash can be submitted in a 2.0 mL microcentrifuge. Bronchial wash should be shipped on ice packs or dry ice. - Lung tissue or respiratory tract tissue: Pre-excised 100-300 mg of tissue can be submitted in a 2.0 mL microcentrifuge tubes on ice packs or dry ice. Do not submit tissue samples larger than 300 mg (~300 uL). - Other Sample Types: - Fecal swabs or fecal pellets: Although it is primarily a respiratory virus, sarbecovirus has been detected in defecate of humans and, therefore, may be detected in feces of other species. - Nonhuman primates: Fecal material must be submitted as a swab (not whole feces) and placed in a 5 mL conical tube and shipped on ice packs. Never use calcium alginate swabs for PCR sample collection. - Smaller research animals: Place fecal pellets in a 2.0 mL microcentrifuge tube (up to 70% full) and ship at room temperature - Whole blood: Whole blood can be submitted, but virus is cleared first from the blood by the humoral immune response so the window of detection is smaller than respiratory samples. 200-500 uL of whole blood can be submitted in a non-heparin anticoagulant in a non-glass vial. - Environmental Testing: - Exhaust air dust samples from rodent IVC caging can be collected by submitting IVC-vendor-provided media, 2”X 3” filter material, plenum swabs or exhaust vent swabs. Swabs displaced from shafts can be placed in a 5 mL conical tube and submitted a room temperature. Place a copy of page 1 of this form in the appropriate shipping container with your samples. Please ship following International Air Transport Association (IATA) guidelines; visit www.criver.com/shippingmaterials to request free sample collection and shipping materials online. Submit your samples to Charles River:

Sample Type Options by Species The sample type options by species are shown in the table below. Only samples for precautionary screening of laboratory animals may be submitted. In addition, Sarbecovirus PCR can be added to any CLEAR panel for research biologics screening. Contact [email protected] prior to sample submission.
