INTRODUCTION
PANCREATITIS- It’s the inflammation of the pancreas.- It happens when digestive enzymes
produced in the pancreas start digesting the pancreas itself.
ALCOHOLIC PANCREATITIS- It’s the pancreatitis caused by heavy
alcohol use (and it’s one of the main causes.)
- The alcohol is metabolised by the pancreas via both oxidative and non-oxidative ways. It’s metabolites produce changes in the pancrea’s cells, which promote intracellular digestive enzyme activation, predisposing the gland to autodigestive injury.
INTRODUCTIONADH, alcohol dehydrogenase catalyzes the oxidation of alcohol, contributing to the elimination of ethanol from the blood.
ADH3, it’s a genotype of alcohol dehydrogenase (the other one is ADH2), it is susceptible to have polymorphisms (alelles ADH3*1 and ADH3*2), coding for different subunits of the enzyme which would have higher or lower ethanol activity.
INTRODUCTIONPOLYMORPHISMS, are genetic variations represented in different forms on the DNA sequence. They provide an allelic variation between individuals and the diversity of the same specie.
In a single individual the two alleles (different forms) of a particular single copy gene could differ by just one nucleotide and so the gene would be described as polymorphic.

Alcohol tolerance depends too much on the presence or not of enzymes in charge of the metabolism of alcohol and its regulation, so the alcohol consumption may lead or not to alcoholic pancreatitis.
The polymorphisms in this alcohol metabolizing enzymes (specially the alleles of the ADH) could predispose subjects who consume alcohol to develop alcoholic pancreatitis. Since the authors think that the ADH3 increase the alcohol metabolizing, it perfectly explains the development of the disease, because the enzymes produces a higher level of alcohol metabolites which lead to the illness.

Demonstrate the relation between alcoholic pancreatitis and the polymorphism of the ADH enzyme (ADH3*1), most of all, for alcohol metabolism in the North Indian
population.

La enzima usada detecta polimorfismos D y C en el intron 6.
GENOTYPING HECHO EN 3% DE GEL DE AGAROSA CON BROMURO DE ETIDIO.
MÉTODOSLa PCR y PCR RFLP; primero, clonan el fragmento de DNA que codifica para cada enzima y así posteriormente en el caso de la PCR RFLP se utilizan en las enzimas de restricción, para el establecimiento de polimorfismos (en el caso de esta investigación) de las enzimas que pueden predisponer al padecimiento de pancreatitis alcohólica.
Sirven para determinar el polimorfismo puesto que en estos puede variar la recurrencia de los puntos de corte de la enzima (aumentando o disminuyendo, siendo esta la causa del polimorfismo), así generando diferente número de fragmentos de restricción a los esperados y detectar la presencia o no del polimorfismo.
Esta detección de los fragmentos se hace posible mediante la electroforesis.
● La prevalencia del polimorfismo ADH3*1 fue considerablemente alta en el grupo de la pancreatitis alcohólica, comparado con su prevalencia en los otros grupos. (TCP, AC, HC). La frecuencia heterocigota y la de ADH3*2 fue similar en todos los grupos.
● Todos los pacientes eran homocigóticos para el gen ALDH2*1/ALDH2*1.
● La frecuencia de distribución para CYP2E1 fue estadísticamente similar en todos los grupos.
