SINTESIS '3-KETO ESTERSEBAGAI SENYAWA ANTARA

Telah berhasil disintesis senyawa dietil p-metoksi-

sinamoil malonat dan senyawa etil 2-(p-metoksisinamoil)-

3-okso-butanoat, dari bahan dasar etil p-metoksisinamat

yang merupakan senyawa bahan alam.

Etil p-metoksisinamat diekstrak dari serbuk rhizoma

kencur ( Kaempferia galanga Linn ) dengan metoda perko-

lasi etanol teknis. Etil p-metoksisinamat merupakan kris-

tal putih dengan TL = 48,50C - 49,0°C diperoleh sebanyak

9,4%. Senyawa ini terlebih dahulu dihidrolisis dengan

KOH dalam etanol menjadi asam p-metoksisinamat ( 11 = 171°C

- 172°C ) diperoleh dengan rendemen 83,8%. Asam p-metoksi-

namat ini direaksikan pada 40°C - 500C dengan tionilkhlo-

rida menghasilkan p-metoksisinamoilkhlorida sebagai se-

nyawa antara.

Senyawa dietil p-metoksisinamoil malonat merupakan

kristal j ar m kuning dengan TL = 83, 5°C - 84; 0°C diper

oleb dari reakss p-metoksisinamoilkhlorida dengan anion

enolat dietil nalonat, reaksi ini berhasil dengan ren

demen 68,41%. Senyawa ini dihidrolisis dengan KOH dalam

etanol menghasilkan 86,4 % asam p-metoksisinamoil malonat,

yang merupakan kristal jarum kuning muda dengan TL = 174

I760C.

Etil 2-(p-metoksi sinamoil)-3-okso-butanoat adalah

merupakan hasil reaksi dari p-metoksisinamoilkhlorida

dengan anion anolat etil aseto asetat ester. Senyawa ini

75,5 diperoleh dengan rendemen 32%. Anion enolat etil

aseto asetat ester dibuat dengan jalan mereaksikan logam

magnesium dengan etanol kering dan etil aseto asetat

ester dibawah eter kering.

A B S T R A C T

A natural ethyl p-metoxycinnamate, had been used for

the synthesis of diethyl p-metoxycinnamoyl malonat and

ethyl 2-(p-metoxycinnamoyl)-3-oxo-butanoat.

Ethyl p-metoxycinnamate was obtained, from rhyzomes

of Kaempferia galanga Linn by percolation with ethanol,

as a white crystalline compound m.p. 48,5°C - 49,0°C in

9,4% yield. The raw material ethyl p-metoxycinnamate was

hydrolized with ethanolic potassium hydroxide to yield

p-metoxicinnamic acid, m.p. 171°C - 172°C, and the acid

thus obtained was treated with thionylchloride at 40°C-

50°C to give p-metoxycinnamoylchloride as an intermediate

product. Treatment of p-metoxycinnmgylchloride with eno-

lic anion of diethyl malonate yielded a yellow needle

crystalline diethyl p-metoxycinnamoyl malonat, m.p. 8315°C -

84,0°C , in 68,4% yield. This malonate derivate was hyd-

rolized with ethanolic potassium hydroxide to yield p-me-

toxycinnamoyl malonic acid, as pale yellow needle, m.p.

174°C - 176°C, in 86,4% yield.

Treatment of p-metoxycinnamoylchloride with magnesium

ethyl aceto acetic ester yielded ethyl 2-(p-metox_ycinna-

moyl)-3-oxo-butanoat, as pale yellow needle, m.p. 174,5°C

- 175,5°C , in 32% yield. The magnesium ethyl aceto ace-

tic ester which was used for this reaction, was obtained

from reaction between magnesium, dry ethanol and ethyl

aceto acetic ester in dry eter.
