The SiriusT3 is the next generation

physchem measurement system from Sirius

Analytical. It replaces our world renowned

and market leading GLpKa system, which

over the last decade has been established as

the industry standard for pKa, logP/D and

solubility determinations.

With a new modular and ultra-miniaturised

hardware platform, we have developed a

new system which meets the needs of the

pharmaceutical industry from early

discovery all the way through to

late stage formulation

and development.

Autoloader module, which has aworktable with four48-position vial trays. The traysare standard footprint, andeasily integrated with existingrobotics systems. It has a roboticarm which automatically picksup and move vials to the sampleposition of the Titrator module.At the front of the Autoloader module is abuilt-in ultrasonic bath, which can be automatically used toaid the dissolution of poorly soluble compounds.

Dispenser module, which housesthe precision micro dispensersfor adding water, solvents andacid/base titrants from thereagent bottles stored in thedrawer underneath. Theinstrument can have up to 6different cosolvents loadedsimultaneously (Methanol, Ethanol,DMSO, Acetonitrile, Dioxane, DMF, THF andothers). Also contained within this module is a UV/Visspectrometer and light source, which is connected to a fibreoptic dip probe. This provides the same high quality UV/Visspectroscopic functionality as our well established D-PASsystem for the GLpKa.

Titrator module, which features amoving arm with the assayprobes attached – pH electrode,UV dip probe, overhead stirrer,temperature sensor andcapillaries for reagent addition.All of these probes have beenminiaturised to fit into 1mlsolution volumes. There is a row ofbuffer and wash positions used forautomatically calibrating and cleaning the probes, includinga flowing water wash which cleans the electrode with freshwater after each experiment. At the front of the Titratormodule is the sample position, which is temperaturecontrolled with a Peltier device (fully controlled bycomputer), and an additional turbidity sensing device. Thisis the position where the titration for pKa, logP/D orsolubility is performed.

Low Throughput OptionSiriusT3 Dispenser Module + SiriusT3 Titrator Module

High Throughput OptionSiriusT3 Dispenser Module + SiriusT3 Titrator Module +SiriusT3 Autoloader Module

Both options are available with and without the UVspectroscopy system.

The highest throughput assays on the SiriusT3 canmeasure pKas in under 4 minutes, using just 3µL of 10mMDMSO stock solution. No other system can offer richlydetailed measured pKa data at this speed and small samplerequirement.

SiriusT3 measures multiple physchemproperties on a single hardware platform:

pKas, logPs, logDs, Kinetic Solubility and Equilibrium(thermodynamic) Solubility.

The UV system allows us to also measure pKas by a UVmethod, and also determine Molar Extinction co-efficients.The flexibility of the platform allows scope for new andcustomized assays to be easily implemented.

The SiriusT3 allows “goldstandard” physchem measurements to

be performed on sub milligram quantities ofmaterial. This revolution allows the best quality

data to be obtained even on discovery compoundswhere sample quantity is a limiting factor.

By obtaining high quality information at an earlierstage in the discovery and development process,

decisions about chemistry changes andformulation routes can be made early, savingmoney, and potential problem areas in other

assays can be indentified andresolved with ease.
