Automation Control System (ACS) RADSTIM Manual

This manual describes briefly the installation and use of the Automation Control System (ACS) software in conjunction with RADSTIM hardware. ACS Software and the ACS Junction Box allow communication and control of:

-RADSTIM Stimulators-Radnoti Fluidic Control Pump Modules (RADPUMP)

This application provides control of the RADSTIM stimulators and RADPUMP pumps on a Windows operating system. The application communicates to devices via a USB port which is connected to the ACS Junction Box. Up to 16 Radnoti compatible devices, pumps or stimulator modules can be connected to the ACS Junction Box.

Recommended Hardware Setup and Placement

The RADSTIM Stimulator head is designed to be mounted on the lab stand in proximity to your sample chamber. It is recommended it be placed well above the chamber to minimize the risk of fluid contact.

The RADSTIM power supply can be located on the lab bench clear of any potential fluid contact or spillage, or up on a shelf to the rear of your system.

The ACS Junction Box is also designed to be mounted to your lab stand. It is recommended to place towards the rear of the stand clear of potential fluid contact or potential spillage.

Note:As there are many possible uses of the RADSTIM stimulators key factors for placement of components to be observed is caution for potential fluid contamination and or exposure to spillage. Care should be taken at all time to avoid these issues.

RADSTIM INTEGRATION AND SYNCHRONIZATION TO DATA ACQUISITION

There are two ways to connect an external trigger.A/D Data acquisition output pulse to sync input on the Junction Box.

A/D Data acquisition output pulse to sync input on the Junction Box will have a 100ms to 500ms response time that can vary. However, this enables either Monitor Mode or Sync Pulse mode from the stimulator.

Sync pulse allows you to record a sync pulse that indicates the start of stimulation for that channel.

Monitor mode is a pulse for every stimulation pulse. This allows recording of start time (the first pulse) and frequency (the number of pulses over a given time period).

Example: Tissue Organ Bath Electric Field Stimulation (EFS) initiated contraction for force measurement. Monitor Output would show each stim pulse regardless of train, or single pulse repeat etc.

Note: Front BNC trigger has a minimum 50us pulse width and the ACS Junction Box tirigger has a minimum 1ms pulse width.

Direct Trigger of RADSTIM bypassing Junction Box

A/D Data acquisition output pulse directly to the Multi Function BNC on the front panel of the RADSTIM Stimulator(s) using a BNC cable. For multiple RADSTIM’s the signal can be routed from the single output of the Data Acquisition signal output BNC using BNC cables and BNC “T” connectors. “T” Daisy chain the signal to stim 2,3,4., etc. The final stimulator in the series will not require a T as it is the end of the line.

This bypasses the ACS Junction Box and has a 10ms response time to triggering a step. In addition, depending on your acquisition package, you can also set up an automated annotation assigned to a key stroke that triggers the start as well as annotates at the start of a stimulation.

Automation Control System (ACS) Software Installation

Software Installation

The application communicates with the hardware via USB port.

The drivers need to be installed first.

Do not connect the ACS Junction Box while installing software.

Run the program CDM v2.12.12 WHQL Certified.exe on the flash drive.

After installation of drivers, the JB can be connected to a USB port.

Next, install the ACS program by executing the setup.exe on the flash drive.

Now connect the pumps or stimulators(devices) to the ACS Junction Box and power them on.

DO NOT CONNECT OR DISCONNECT RADSTIM’S TO POWER SUPPLY WITH THE POWER SUPPLY ON. THIS MAY RESULT IN DAMAGE.

Run the setup.exe file to install the ACS program

Once the program has been installed run the AutomationControlSystem.exe file to launch the program

Upon startup, the ACS program searches for the ACS Junction Box and devices.

The ACS Junction Box status is shown in the upper left corner of the ACS window. If the ACS Junction Box was not found, try disconnecting and reconnecting the ACS Junction Box from the USB port. Wait a while for Windows to enumerate and then under Files select Refresh. This will search for the ACS Junction Box and devices again.

In the Status pane on the left side, the number of devices will be displayed.

First select the stimulator in Bank 1.

The stimulator will appear in the drop down menu by way of the devices serial number.

It is recommended that you rename the stimulator based on your experimental setup for ease of recollection.

Under the File menu select “Alias/Calibrate Pumps” or Ctrl+P as a shortcut.

In the pop up window type in the desired name for the device.

For example “Stimulator 1” etc.

Close the window when done.

Under the file menu select the “Refresh xStick” option or Ctrl+R as a shortcut.

The screen will refresh and “none” will reappear in the stimulator drop down box.

Click on the stimulator drop down box and your newly named device will appear.

Select your device.

Repeat the above steps for each of the stimulators that you are going to use.

Once all devices have been named, you can begin to build your stimulation protocol.

Click on “Add Step”.

This will add a step function window to the Bank.

You can name steps based on your particular experimental design. For example in this case we have used “Voltage Response”

Once the step has been named, you can begin inputting the stimulator values for the step.

In creating many steps you can copy (Ctrl+C) the step and paste (Ctrl+V) also.

Step 1 “Voltage Response”

There are 4 tabs in the step function window that we have named “Voltage Response”

Note: The “Pump” selection is for another application that is not covered in this manual.

The General Tab

Step Name (which we have already done)

Mode: Select which mode you are going to use. In this case you will be using the “Stimulate” mode.

Time:

This is the duration of the step from start to finish. The time setting in this tab overrides time parameters set in the “Stim(2)” tab.

Care must be taken to be sure that this time setting will accommodate the full duration of the parameters from “Stim(2)” tab specifically in the train duration which we will cover later.

The time setting in this tab should cover the time allocated to the step. The program will not progress to the next step until this time value has lapsed.

The Stim(1) Tab

Mode

Using the drop down menu for “Mode” you have the option of selecting “Single” or “Continuous”

Single: This setting instructs the stimulator to send a single pulse.

Continuous: This setting instructs the stimulator to send continuous pulses.

Freq/Period

The Freq/Period is the time from the rise of one pulse to the rise of the next pulse. It is one cycle of the pulse. For example, with a setting of 30Hz the cycle will complete 30 times in 1 second.

Options for Freq/Period are Hz, ms and sec.Hz is the most common usage for frequency.

Duration

The duration is the time that the pulse is at the set voltage.

Duration cannot exceed 50% of the frequency cycle (Duty Cycle)

Voltage

The RADSTIM operates in either voltage (V or mv) or Current (ma).

In Voltage mode the unit puts out the desired voltage and current will regulate based on the load.

For reference’s of high voltage with low amperage, which is common in many publications where voltage is called out at 100v or more, the amperage is usually limited to milliamp range.

The RADSTIM’s maximum voltage is either 32V Monophasic or 64V Biphasic, the amperage is 2A (2000ma).

In order to accomplish such protocol requirements, we use the following equation.

The power P in watts (W) is equal to the current I in amps (A), times the voltage V in volts (V): P(W) = I(A) × V(V)

Thus, watts are equal to amps times volts:(W) = (A) x (V)

For example:Your publication calls for 100V stimulation. Assuming the max output of the stimulator used is 600ma for amprage. The wattage would be caclulated as follows.

100V X 0.6A = 60W

Using the RADSTIM equal power (W) would be achieved by the following settings:

30V X 2A = 60W (The amperage will auto regulate up to the max output of the RADSTIM)

If the “Output” drop down selection is ma(Current), then the voltage will auto regulate up to the max output of the RADSTIM.

Using the RADSTIM, maximum power is 64W

32V X 2A = 64W

Interpreting previously published stimulator settings:

There are many factors that impact voltage or current settings in EFS. Electrode materials and fabrication, wire type, proximity of leads to sample, sample mass, bathing solutions etc., not to mention physiological response variances. Simply duplicating previously published settings may not yield optimum results.

In all cases some preliminary testing will have to take place to determine best voltage settings, current settings and frequency settings in order to yield maximal responses from your samples.

Voltage response curves, current response curves and frequency response curves are recommended.

Output

The output drop down will enable you to select either “Monophasic” or “Biphasic”.

You will notice that when you change the output to “Biphasic” the level will double in value.

Monophasic will make the pulse only + voltage while Biphasic will make the pulse cycle from + to 0 to - voltage.

Sync Pulse

Outputs a TTL active high pulse at the start of output

Monitor outputs a TTL level signal representing the RADSTIM output. In Biphasic mode, minus outputs are represented by a positive pulse.

The Trigger input BNC on the front panel of the stimulator head is a TTL active high trigger. Minimum pulse width is 50us and has a delay of about 10ms before outputs become active.

The Stim(2) Tab

Train:

Once the “Train” drop down menu has been clicked on you will see options for “Train Rate” and “Train Duration”

Train Rate is the start of a burst to start of next burst.

Repeat Rate is redundant to Train Rate. However Repeat Rate overrides Train Rate. It is recommended to turn Repeat “Off” in the drop down next to “Train Off/On”

Train Duration must be less than Train Rate.

The fastest response time to a trigger is achieved by connecting the Data acquisition output pulse directly to the multifunction BNC on the front panel of the stimulator head. For multiple RADSTIMS the signal can be routed from the single output of the Data Acquisition signal output BNC using BNC cables and BNC “T” connectors. “T” Daisy chain the signal to stim 2,3,4., Stim 4 will not require a T as it is the end of the line.

Alternately, using the sync input on the ACS Junction Box will have a 100ms to 500ms response time that can vary. However, this enables either Monitor Mode or Sync Pulse mode from the multifunction BNC on the stimulator head, depending on which is selected on the Stim(1) tab. This will allow you to record (if open data acquisition channels are available) a sync pulse that indicates the start of stimulation for that channel, or in monitor mode a sync pulse for every stimulation pulse allowing recording of start time (the first pulse) and frequency (the number of pulses over a given time period).

Example: Tissue Organ Bath EFS initiated contraction for force measurement. Monitor Output would show each stim pulse regardless of train, or single pulse repeat etc.

Note: Front BNC trigger has a minimum 50us pulse width and the ACS Junction Box trigger has a minimum 1ms pulse width.

The channel can then have a counter associated with it to convert to Hz

Alternately Sync output would simply send one pulse at the onset of a stimulation step.
