To meet today’s needs, VICTOR2 accepts all types of microtitrationplates with between 6 and 1536 wells. Clear and opaque (black orwhite) plates, filterplates and membranes, Petri dishes, slides, filters and Terasaki plates are all accepted.
Any non-standard configuration can be defined using the instru-ment’s Windows Workstation software. The plates can be loaded manually or using 20-plate free access stackers (40-plate stackers are also available).
You can also integrate VICTOR2 into your robotics system.To meet the future demands on your laboratory, 32-bit WindowsWorkstation software is designed to utilize the full capabilities ofWindows NT, Windows 95 and Windows 98.

Not only is VICTOR2 easy to use, it is also easy to learn to use.
It gives you the freedom to employ it’s different technologies,
without forcing you to spend time learning how to use different
instruments. Nor do you have to waste time telling the instrument
things it should know. Details of the plates, filters and labels you
use are all pre-installed on VICTOR2.
It’s so easy but an extremely powerful tool
To measure your assay with VICTOR2,you simply load the plate, select aprotocol, and start measurement.Although easy to use, VICTOR2 doesnot restrict your freedom. It is a flexi-ble system providing access to count-less advanced features, all of whichare made available through the usersoftware.
Allowing everyone more time for science
There is a VICTOR2 configuration package to support every type of laboratory. Where manual loading is too slow, you can choose 20-plate or 40-plate stacker towers to allow automatic loading and continuouscounting.
The instrument’s HELP function is part of the versatileWindows Workstation package. You can click on pictorial“hot spots” to obtain more information about any feature ofthe counter. There is also an extremely comprehensivesearch function that allows you rapid access to specificinformation.
Factory set protocols cover labels, plates and filters, andthere is a PROTOCOL EXPLORER for quick access and editing of protocols. With ready-prepared protocols there is no need for manual tuning. However there are limitlessopportunities to create your own application-specific protocols.
After measuring of your plate, VICTOR2 automatically trans-fers results to appropriate application software such asMicrosoft Excel or MultiCalc®. Or you can manage bothassay design and results analysis using the instrument’soptional WorkOut™ data analysis software.

Time-resolved fluorometry (TRF) is based on the unique fluorescence properties of lanthanide chelates. Because theirfluorescence is as much as 200,000 times longer than conventional fluorophores, measurement starts only after allnon-specific fluorescence has died down. And because the Stokes shifts are so wide, measurement takes place atclearly separated wavelengths. The great specificity of each lanthanide’s emission spectrum also means you can measure several different labels as part of the same TRF assay.

Calcium response following addition of ATP to cultured cells:Chinese hamster ovary cells were challenged with the purinergic agonist ATP, introduced using the VICTOR2 injector. Dual wavelengthmeasurement alternating between 340 and 380 nm every 0.5 s provides a ratiometric index of intracellular Ca2+ activity, enabling realcomparison between different experiments, cell types etc. The fast filter changer capability of the VICTOR2 enables the use of severaldyes monitoring different cellular functions for each experiment.
A limitation of cell-based fluorescent assays in microplates today is the necessity of using single wavelength lightsources. In many cases much more information could be gathered through the use of ratiometric indicators or indeedmultiple probes in one experiment. For assay development, or secondary screening the information content of the assayis as valuable as absolute speed. All VICTOR2 counters are capable of pharmacological measurements on end-point typeinflux experiments such as NMDA receptors or voltage gated Ca2+ channels. Injector-equipped VICTOR2 models will dosub second resolution single wavelength dye pharmacology for GPCRs. VICTOR2 does multi wavelength and/or ratio-metric Ca2+ analysis with sub second time resolution and integral addition of effectors.
Improved below-reading optics in the VICTOR2
A new feature of the VICTOR2 is the option of a direct light guide for the fluorescence excitation. This is especially useful for fluorescence assays reading microplates from below. On the VICTOR2 you can now do assays needing extra sensitivity for this mode of epi-fluorescence such as the Aurora Biosciences ß-lactamase reporter gene assay system. Note: Improved below reading is now standard item in all VICTOR2s.
