All: Just a reminder that we will be euthanizing next week - Tues (Jan 19) and Thurs (Jan 21). We will be working on the WADDL floor and for those of you not familiar with the floor I can give you directions. We need to be at WADDL by 8:30 to allow Annie and her team to collect CSF right away and then move along to other tissues, etc.

- I suppose that means you’ll need to dart around 7:45AM

Charlie - would you reserve the Steffan truck?

Katie-Did you schedule a CT scan and if so, when will that be done relative to the euthanasia (immediately before, after, just limb???). We’ll have to move the animal if a CT scan is done from radiology to WADDL which will take extra time.

We will have liquid nitrogen, dry ice and sample tubes for those who need them, otherwise please plan to bring your own since lots of things will be going on simultaneously and everyone needs to focus on what they are doing.

All meeting at the bear center is probably the most reasonable thing to do and then we can take an extra car or two to the Hospital. Nina/Gaylynn - I’m guessing you’ll just meet us at WADDL?

Any questions, let me know.

Thank you Heiko,

Joy, are you happy for me to leave the darting with you? It sounds like everything else is covered. I am rather swamped at the moment with courses & research as I am heading out of town again at the end of the month.

Thank you,

Hi All, Charlie’s asked me to pop down to help with sample collection on Tuesday morning. I realize there is going to be a lot going on, but could someone please let me know when you’re ready to start sampling as I’ll only have a limited amount of time to help out. My number: . Thanks very much,

Here are the order and the priority of what needs to be done. Everyone, please read because there are things that each of you need to do.

1. As soon as the bear is delivered to necropsy, Heiko and Annie will work as quickly as possible to collect CSF. If they or anyone else needs blood, this would be the time to collect it.

2. Once that is done, the bear will be euthanized with the euthanasia solution that will bring. Nina: Can you make that injection?

3. As soon as Nina says the bear is dead, Heiko will remove the head (not easy) while others can start collecting superficial tissues like muscle, fat and skin. I presume Craig and Katie want the legs and front and hind quarters on one side of the bear left intact? So, while Heiko and Annie

are collecting CSF, quickly discuss which side of the bear is available for Chantal and et al. to collect fat, muscle and skin. Once those tissues are removed or even during if things are going slowly and the bear is positioned so you can get into the body cavity, you can cut into the body cavity and start removing samples of the organs listed below. Note below that there are a lot of tissues being sampled, so I’m sure help from those with knowledge of anatomy would be appreciated.

4. After all “research” tissues are collected, Nina or someone else will collect viscera samples for baseline histopathology.

5. I’ve asked to remove all front claws at the first joint of the phalange, unless Craig and Katie need the feed left intact for their studies---and that is fine if they are needed by C & K.

Finally, to all of you, please work as a team. If you are not doing something, please help someone else. This is a rare event, so let’s get as much useful tissue as possible. If you want to change something that I’ve listed above, please send a response to everyone with your thoughts. Thanks. Joanna, Omar and I were discussing samples we would like to collect on Tuesday and Thursday and a long list came up that I wanted to run by everyone. We discussed brain samples with Heiko already and it sounds like that will only be possible later, after they have been formalin fixed. Other samples were are looking to collect will include: Liver, fat, muscle, lung, heart, skin, spleen, pancreas, and kidney. From the kidney we would be looking to collect samples from both the medulla and the cortex. As we understand it we will have to collect samples post-mortem and just work as quickly as possible. We are also hoping to get multiple samples of each tissue, flash-freeze half of them and fix the other half in RNAlater. We have tubes to bring and will fill half of them with RNAlater, we will also bring dry ice for transport of flash-frozen samples. It sounds like there will already be liquid nitrogen there, but should we bring more are we set with what we have? Does this work for everyone, or are there concerns I have not thought of yet? If there are any tissues I have listed here that are not available that is okay, just let us know.

Thanks everyone and we will see you Tuesday morning.

I have the record from Tuesday and was going to bring it over after tomorrow's procedure to save a trip. Since the second procedure is delayed, I can drop it off tomorrow. I have the anesthesia monitoring notes to add to the records binder as well

Thanks. Heiko

p.s. Did you add a Euth record to the file in the center or do you have it with you? If not, I’m heading over to the center now.

Just a reminder that I am on clinical service and will not be present. I don’t think you need anything from me with this procedure, but if you do let me know.

Heiko
