ご挨拶
頭部の放射線暴露の神経機能への影響に対して問題意識を持つ放射線医療従事者と、放
射線の作用を応用して神経回路を研究している神経科学者が参画し、血管内皮細胞・グリ
ア細胞・神経前駆細胞へのダメージによる機能障害のみならず、シナプス機能そのものに
もたらす放射線作用の解明をも目指す放射線神経生物学という新たな研究領域の創出を目
指し、本年1月に国際放射線神経生物学学会が組織されました。
その直後の3月11日に東日本大震災に見舞われ、福島第1原発事故により、大量の放
射性物質が外部に飛散しました。事故がいつ収束するか未だ不確定であり、外部に漏出し
た放射性物質が我々の健康にどのような影響を与えるのか、多くの国民が不安に感じてい
る状況です。このようなときこそ、放射線が健康に与える影響を客観的に評価する指標と
なる基礎的なデータを集めることが重要であると思います。今大会において、放射線神経
生物学の最前線で活躍する研究者が一同に介して、最新の成果を発表し、実りある討論を
行うことを期待しております。ふるってご参加ください。

Dear Colleagues,
The International Society for Radiation Neurobiology (ISRN) wishes to invite you to Maebashi, the
capital of Gunma Prefecture, Japan, to participate in its Second Annual Meeting, which will be held
on 3 December 2011.
On 11 March this year, we experienced the Tohoku earthquake and Tsunami. The Fukushima
Daiichi nuclear disaster followed, and we still don't know when it will be overcome or to what extent
it will affect our health and life. At the meeting, recent advances in the forefront of radiation
neurobiology will be presented, which, we hope, will help promote understanding of the effect of
radiation on brain development and function.
Maebashi is located at the feet of Mts. Akagi and Haruna, and the famous hot spas of Kusatsu and
Ikaho are in the neighborhood.
We are looking forward to welcoming you to the meeting and having fruitful discussions together.

PL Recent progress of NBS1-mediated responses to radiation damage

Nijmegen breakage syndrome (NBS) is characterized by high sensitivity to ionizing radiation (IR) and
microcephaly. NBS1, which is mutated in NBS, contains a conserved MRE11-binding domain and an
ATM-binding domain at the C-terminus. NBS1 initiates homologous recombination repair (HRR) with
MRE11 and recruits ATM kinase to sites of DNA damage, where it activates the DNA damage checkpoint.
Therefore, the roles of NBS1 are associated with the presence of two binding domains. We recently
identified other two conserved domains at the C-terminus, where NBS1 binds to RNF20 for chromatin
remodeling and to RAD18 for regulation of damage tolerance. RNF20) A well-characterized early histone
modification that is triggered by DNA damage is the phosphorylation of the histone H2AX. However, it is
likely that another histone modification could be involved in this pathway because H2AX-/- cells still display a
normal localization of RAD51, a key protein HRR, to sites of DNA damage. We showed that RNF20, E3
ubiquitin ligase of histone H2B, is localized to DSBs independently of H2AX, and is required for the
recruitment of the chromatin remodeling factor SNF2h. Depletion of RNF20 or SNF2h compromises HRR
and enhanced sensitivity to radiation. RAD18) Translesion DNA synthesis is critical for DNA damage
tolerance. While ubiquitin ligase enzyme complex RAD6/RAD18 is known to mono-ubiquitinate PCNA, how
they are regulated by DNA damage is not understood. We showed that NBS1 binds to RAD18 after UV
irradiation and mediates the recruitment of RAD18 to sites of DNA damage. Thus, in addition to IR-response,
NBS1 plays a key role in damage tolerance.

S1-1 成体脳および培養神経細胞における放射線照射の急性効果

一般に、成熟した非分裂細胞は分裂中の非神経細胞や未成熟神経細胞に比べて放射線照射に対す
る感受性が低いと考えられているが、発達途上の神経細胞は最終分裂終了後もシナプス形成期まで
は、比較的高い放射線感受性を示すことが明らかとなっており(Shirai et al. Neurosci. Lett. 399:57,
2006)、放射線療法の副作用の機序を考える上で重要と思われる。放射線エネルギーは核酸や蛋白
質など生体高分子化合物に作用するので、最終分裂を終え成熟した神経細胞の機能にも影響するこ
とが考えられるが、多くの蛋白質機能や遺伝子発現に支えられているシナプス機能や学習行動への
急性効果に関してはほとんど知見がない。そこで我々は放射線を頭部への照射のもたらす影響につ
いて行動レベルで解析を行なった。その結果、恐怖条件づけの学習や記憶における効果は必ずしも
明確にみられなかったものの、条件づけの直後に放射線照射を受けた群で文脈依存的な恐怖反応
(フリージング)が低下する傾向があった。ラット海馬の初代培養神経細胞では、放射線照射によ
りシナプス蛋白の減少と樹状突起スパイン形態の変化が示されており(Okamoto et al. Rad. Res
172:718, 2009)、このようなスパイン構造の微小変化が放射線照射の学習行動におよぼす急性効果
の基礎となっている可能性がある。

S1-2 小児脳腫瘍における頭部放射線療法後の脳障害克服を目指して

小児の脳腫瘍は、小児における悪性腫瘍の約1/3を占めるが、この 20年間で生存率は飛躍的に
向上し、現在では、その 70%以上の児が生存する。頭部放射線療法は脳腫瘍に対する最も有効な治
療法の一つであるが、その後に起こる認知障害は深刻な問題であり、特に 3歳以下の小児において
は、その合併症は顕著である。また、放射線療法による認知障害は時間とともに増悪し、少量の頭
部放射線療法でも起こり得る。脳腫瘍の小児の生活を改善させるために、この放射線療法後の神経
学的合併症を軽減させることは極めて重要であり、急務なことであるが、現在のところその予防法
はなく、研究報告もわずかである。
私達は、幼若マウス/ラットを用い小児頭部放射線障害モデルを作製し、後障害の予防法、軽減
法開発に向け研究してきた。これまでに、頭部放射線照射により、海馬歯状回と脳室下帯が選択的
に受傷すること、未熟脳では成熟脳より受傷の程度が強いこと、海馬歯状回においての受傷は成獣
になっても回復しないこと、各種炎症マーカーは低酸素虚血による受傷時より早くピークを迎える
ことなどがわかってきた。また、予防/軽減法に関しては、apoptosis-inducing factorの抑制、低
体温療法、積極的運動、神経幹細胞の海馬への移植などの可能性を見出している。
本シンポジウムでは、モデルの作製、これまでにわかってきた放射線療法の未熟正常脳への影響、
予防/軽減法に関して発表する。

ASPM はヒトの遺伝性小頭症の原因遺伝子である。我々は、がん細胞を含む多くの細胞で、ASPM 遺
伝子の転写(mRNA)が放射線によって一過性に減少することを見出した。マウスの胎仔脳の nestin
陽性細胞(神経幹細胞)では、亜致死線量の放射線が Aspmの mRNAと蛋白質の発現を著しく抑制し
た。今回、Cre-loxP 組換えシステムによる条件的 Aspm ノックアウトマウスを作製した。Aspm+/-
ヘテロマウス同士の交配からは、Aspm 遺伝子型+/+, +/-, -/-の個体はほぼメンデル遺伝の法則に
従って生まれた。12 週齢の成体マウスにおいて、異なる遺伝子型の間に体重差はないが、Aspm-/-
の個体にのみ、脳で -15%、睾丸で -80%という顕著な重量の減少が見られた。HE染色による病理学
的所見で脳に著しい変化は検出されなかったが、精子形成の障害が明かであった。次に Cre を
‘Nestin 遺伝子プロモータ’下に発現する TgNes-Cre マウスを用い、胎生期の中枢神経特異的に
Aspm 遺伝子を破壊したところ、Aspm 欠損マウスと同様に脳重量の減少が見られたのに対し、精巣
は野生型と同じく正常であった。これら Aspm欠損マウスは、遺伝性小頭症の中で最も一般的な「V
型ヒト家族性小頭症(MCPH5)」のモデル動物として今後、同疾患の病態解明と治療に、また放射線
による ASPM発現抑制の機構解明に貢献するであろう。

Radiation exposure dose in space accounts for 0.2 ~ 10, mSv per day. Long-term manned space mission
inevitably receive space radiation exposure and microgravity might give a synergistic effect on those space
radiation effect. We previously suggested that low dose radiation as well as microgravity may cause in
intracellular oxidative stress and result in different types of cell death (Majima et al., Biological Sciences in
Space, 2009). A human SK-N-SH cell line is a slow growing neuron like cells, of which p53 status is normal.
We send the cells to International Space Station (ISS), at Japan Experimental Module (JEM), namely“Kibo”by
STS131(19A) in April 2010, and cells were cultured at 37˚C with 1G or µG condition for 14 and 28 days at
Cell Biology Experiment Facility (CBEF) at JEM. Consequently, the cells were fixed or frozen and stored at
-80˚C, then the cells were returned by STS/132 (ULF4). The samples were transported back to Kagoshima
University. Now, gene expression analysis has been undertaken using the sample. Mitochondrial impairment
has been also examined using the same samples. The following results were obtained from analysis of
rethawed up cells. 1) ISS short and long-term cultured cells grew up faster than the control cells. And, no
difference was observed between µG and 1G in any case. 2) ISS cultured cells generated more intracellular
ROS and increased the expression of HSP proteins and antioxidant enzymes compared with control cells. The
results suggest that space environment give oxidative stress in cells during the stay in space.

S2-1 Gamma knife thalamotomy for the treatment of tremors

Purpose: We performed Gamma knife thalamotomy on 18 cases of tremor dominant Parkinson disease and
essential tremor. The outcomes and the location of effective lesions in the thalamus were analyzed.
Materials and Methods: Eighteen patients ( 9 Parkinson patients and 9 essential tremor) ranged in age from 66
to 81 years old (mean age: 75.6) were treated. Gamma knife thalamotomy was performed aided by thin slice
MRI, Surgiplan and space ATLAS. The target point was selected in the radiologically identified VLp nucleus
of human thalamus (Hirai and Jones, 1989) for control of tremor. The target point was decided on the basis of
measurement data of optimal locations of 12 coagulative lesions in the thalamus treated previously using MRI
guided electrophysiologically controlled thalamotomy. A single isocenter exposure with the 4 mm collimator
helmet was used to make the lesions. The maximum dose was 130-140 Gy. The follow up period was 36-85
months (mean 63 months).
Results: Symptoms of the patients improved gradually. One year later, tremor were improved by 50-90% in
tremor scores in all patients and become stable. The overall results are satisfactory, being 85% successful
without any noticeable complications. The target area of effective lesion was located in 40-50 % of the
thalamic length.
Conclusion: Gamma knife thalamotomy can be safely applied to patients with tremor. Lesions located in
optimal areas in radiologically identified thalamic nuclei provide relief from tremor in selected cases.

S2-2 側頭葉てんかんに対するガンマナイフ治療
-最近の知見と新たな治療パラメターを用いての独自の治療戦略-

[目的] MTLE (Mesial Temporal Lobe Epilepsy) に対するガンマナイフ治療は臨床上効果が認められ、
欧米ではいくつかの大規模臨床研究がなされている最中である。今回、われわれ独自の治療戦略に
基づき、当科にてMTLE 5 症例治療を経験したのでその有用性について報告する。
[方法] Marseille グループでの retrospective study より、重要構造物を target 内に入れ照射体積 7cc で
収め、辺縁線量 22-24Gy として治療している。同時に各部位への十分な照射が必要なことより、な
るべく内部線量分布を高いレベルで均一化させるべく、80%領域を target 内で多く占拠させるよう
工夫している。
[結果] 5症例のうち3症例が Engel class 1、1症例が class 2(まだ1年の経過)、1例が class 4 であっ
た。効果発現までには最低でも 12 か月を要した。経過中画像上 severe な radiation necrosis は認めら
れていない。また全症例で重篤な合併症は認めていない。
[結論] 難治例に対して、開頭手術を必要とせず治癒へ導ける可能性のある治療であることは間違い
ない。今後症例をさらに重ね、基礎的研究による治癒機序解明も併せて、新たな治療戦略から
common indication となるべき治療法へと upgrade させていきたい。

S2-3 脳磁図を利用したてんかんの診断と治療

脳磁図は脳内における電気生理学的変化を、頭皮からの磁場の変化としてとらえるものであり.
てんかんの診療においては、てんかんの焦点を CT、MRIなどの画像診断の一点として把握すること
が可能である.したがっててんかんの部位診断においては脳波に勝る診断法と言える。
当科では特に難治性のてんかん症例に対して脳磁図を利用した診断と治療を施行しているが、今
回は真性てんかんとしての内側側頭葉硬化症と、各種病変に由来する症候性てんかんの中で海綿状
血管奇形、結節性硬化症、視床下部過誤腫、グリオーマついて検討した.内側側頭葉硬化症、視床
下部過誤腫では発作焦点は病変内側に発生し,かつ周囲脳組織に伝搬して二次性焦点を形成してい
た.一方転移性脳腫瘍、海綿状血管腫においては発作焦点は病変の外部にあって,転移では周辺浮
腫に中に,海綿状血管腫では周囲のヘモジデリン蓄積部位に明瞭に認められた.結節性硬化症、グ
リオーマにあっては発作焦点は腫瘍の内側、外側に散在していた.いずれの病変についても辺縁線
量 18−20Gy の病変自体のガンマナイフ照射にてけいれん発作の一定のコントロールが得られる
が,海綿状血管腫においては病変周囲の発作焦点がヘモジデリン沈着にともない不規則に散在する
ため,病変の処理のみでの発作コントロールはしばしば困難であった.病変の種類によって,発作
焦点の分布、集中度、伝搬形式が異なるのでそれぞれに対応する治療法が必要であり,発作の完全
な消失を意図する場合,辺縁線量として 20Gy あるいはそれ以上を、発作焦点に確実に定位的照射
する必要がある。

S2-4 Gamma knife callosotomy in children with drug-resistant epilepsy

Objective: We evaluated the efficacy of gamma knife callosotomy for two patients with intractable epilepsy
associated with tuber sclerosis.
Material and methods: Case 1: a 14-year-old female had a history of complex-partial seizure and drop attack
from the age of 2 and half years. At the age of 9-years, the patient suffered behavior disturbance. At 14-years
old the patient received gamma knife radiosurgery (GKS) for the anterior half of the corpus callosum with a
maximum dose of 140 Gy. After the procedure, drop attacks decreased 25%, urinary incontinence
disappeared, and behavior disturbance was improved. 8 to 14 months after GKS, brain edema occurred
without symptoms. The patient received a second GKS for the posterior third of the corpus callosum with a
maximum dose of 140 Gy 23 months after the first GKS. 5 months after second GKS, the seizures had
almost disappeared. Case 2: a 5-year-old male. The patient suffered complex seizure from the age of 4
months. He also suffered drop attack from 3 years old. GKS was performed on the anterior one third of the
corpus callosum with a maximum dose of 140 Gy. This patient showed no improvement with brain edema at
the frontal lobe between 6 to 9 months after GKS. The patient received a second GKS for the posterior one
third of the corpus callosum with a maximum dose of 140 Gy 19 months after the first GKS.
Conclusion: Our experiences revealed that the gamma knife callosotomy can be effective for intractable
seizure.

S2-5 三叉神経痛の放射線外科治療

三叉神経痛は片側の三叉神経領域に生じる電撃性の顔面痛であり、抗痙攣薬(カルバマゼピン)が
有効であり、病状に応じてさらに神経ブロックや各種の外科的治療(神経節凝固、グリセリン注入、
血管減圧術など)が行われる。より低侵襲の治療として、放射線外科治療が用いられている。しか
しながら放射線による治療効果の機序は定かでない。高線量治療(80-90Gy)のターゲット、至適
線量、治療体積など術者(施設)により治療方法が異なっている。いずれの報告でも治療効果(痛
みの消失)と合併症頻度(しびれ、知覚障害など)が相関するとされている。放射線外科治療の歴
史をふりかえりながら、最近の我々の経験をふまえた治療効果から治療機序について考察する。

LS PET Molecular Imaging for Innovation in Drug Development and Preemptive Medicine

In vivo molecular imaging has become a key technology for medical innovation, i.e., integrated live
science, patho-physiological science, molecular evidence-based diagnosis, innovative drug development, and
preemptive medicine. Rapid progress in life science brought the stage up to the living objects or subjects,
even functioning human beings. The invention of a variety of imaging techniques including Positron
Emission Tomography (PET) accelerated the paradigm shift in life science from patho-physiological science in
disease model animals to that realized in patients. When the biomarkers for early detection of signs toward
diseases and also those for surrogate end-point were established and their changes could be followed by
molecular imaging, it might be quite beneficial for preemptive medicine and evaluation of therapeutic outcome.
Recently, beta-amyloid imaging and pancreatic beta-cell mass imaging have been highlighted for prediction of
Alzheimer’s disease and diabetes mellitus, respectively. Such types of valuable biomarkers related to the
cause or influencing factors are also really important to develop the new drugs. In this sense, mutual
collaboration between the research consortia in biomarker exploration and in molecular imaging would be
really important. In this context, in 2005, Japanese Government initiated the Molecular Imaging Research
Program under MEXT. Our Center in RIKEN was selected as the key hub center for development of
All-Japan research network to further promote mutual international and multi-disciplinary collaboration on in
vivo molecular imaging. To realize the molecular probing concept, we have so far developed a variety of
novel chemical methods for labelling the low and higher molecular weight compounds with due positron
emitters, such as C-11, F-18, Ga-68, and Cu-64, and the number of different types of the molecular probes to
be used in PET study is more than 175 so far developed in our Center and the repertoire being increasing every
day. Direct application of this molecular probing concept is pharmacokinetics and efficient DDS
development. We also combined molecular imaging techniques with functional imaging techniques on the
preemptive medicine, one example is the application of the combination to molecular/neural mechanisms of
fatigue. The concept, outline of our activities, pharmacokinetics studies, and preemptive medicine studies
with efficient application of molecular imaging will be presented.

P01 Control of spontaneous firings by Ca2+-activated ion channels triggered by the Ca2+ release
via ryanodine receptors in inhibitory neurons in the rat prepositus hypoglossi nucleus

In our previous study on the rat prepositus hypoglossi nucleus (PHN) that is a brainstem structure involved in
gaze control, we found that some neurons showed spontaneous outward currents (SOCs). In the present
study, we investigated 1) ionic mechanisms of SOCs, 2) neuronal types exhibiting SOCs frequently, and 3) the
effect of Ca2+
-activated conductances on neuronal firings, using whole-cell recordings in rat slice preparations.
Pharmacological analyses revealed that SOCs were induced by the activations of small K+ conductance
(SK)-type Ca2+
-activated K+ (KCa) channels and ryanodine receptors. The analysis of firing patterns and the
study using transgenic rats, in which inhibitory neurons are labeled fluorescently, revealed that most PHN
neurons exhibiting SOCs frequently were inhibitory. Spontaneous firings of inhibitory PHN neurons were
enhanced by the block of SK-type KCa channels. Spontaneous firings of inhibitory PHN neurons were
enhanced by the block of SK-type KCa channels. However, their firings were reduced by the block of
ryanodine receptors, suggesting that the Ca2+
release via ryanodine receptors activates not only SK-type KCa
channels but also ion channels involved in membrane depolarization. This was supported by the reduction of
the firing frequency by the block of Ca2+
-activated nonselective cation (CAN) channels. These results
suggest that the Ca2+
release via ryanodine receptors activate SK-type KCa and CAN channels, and the
activation of the channels regulate spontaneous firings in a push-pull manner.

P02 The role of excitatory action of GABA in adult GnRH neurons

The GnRH neurosecretory system constitutes the final common pathway in the neuroendocrine control of
reproduction. The hypothalamus contains a relatively small number of GnRH neurons and these are diffusely
scattered throughout the hypothalamus. Hence the mechanisms enabling GnRH neurons to synchronize their
activity to generate the discrete episodes of GnRH secretion remain unknown. GABA has long been
implicated as one of the major players in the regulation of GnRH secretions. Using a transgenic rat expressing
EGFP under the control of the GnRH promoter, we examined the direct action of GABA. GABA increased
[Ca2+
]i in adult female GnRH neurons at each estrous cycle stage. The percentages of neurons with increased
[Ca2+
]i were 90% in proestrus, 59% in estrus, 84% in diestrus I, and 89% in diestrus II. The percentage was
lower in males than in females. GABA responses were blocked by bicuculline. Removal of extracellular Ca2+
completely suppressed the GABA action, and bumetanide attenuated the response. These results indicate that
GABA depolarizes GnRH neurons by activating GABAA receptors, thereby activating voltage-gated Ca2+
channels and facilitating Ca2+
influx. In addition, the response to GABA is modulated according to the estrous
cycle stage, diurnal rhythm, and sex. Although GABA is typically an inhibitory neurotransmitter in the adult
mature central nervous system, mature GnRH neurons show an unusual characteristic of being excited by
GABA. To examine more detailed characterization of these actions in vivo, we have currently generated
transgenic mice, which would enable controlled modulation of the excitatory action of GABA on GnRH
neurons.

P03 Inhibitory synaptic plasticity in the mouse lateral amygdala

The lateral nucleus of the amygdala (LA) receives and integrates cortical and thalamic inputs to establish fear
memory. Although it is well-known that the timing of these two inputs is critical for formation of the fear
conditioning, how this circuitry processes temporal information is unknown. Since inhibitory circuits in LA is
known to plays an important role in the regulation of fear memory and its extinction, analysis of inhibitory
circuits in LA will provide us important information to resolve the issue. A voltage-sensitive imaging system
that enables monitoring of electrical activity at multiple sites is a powerful tool to analyze how the amygdala
integrates inputs temporally and spatially. Using a voltage-sensitive dye, Di-ANEPPS, we have analyzed
optical signals in response to stimulation of the external capsule (EC). Long-lasting hyperpolarization (LLH)
that spread throughout LA was observed when the stimulating electrode was placed at a proper site on the EC
close to the upper part of LA in the mouse coronal slice preparation. LLH has two components mediated by
GABAA and GABAB receptors. GABAB receptor mediated the late component of LLH (the late LLH) lasted
about 800 ms, and reach the maximal responses around at 300-400 ms after the EC stimulation. The late LLH
may be involved in the time window related to establish fear memory and its extinction.

P04 軸索伸長を促すポジティブフィードバック機構

これまでに見つかっている温度センサー・TRPチャネルは成体(adult)で温度・痛み感覚を惹起
している。では、発生過程において、いつから発現を開始するのであろうか?この素朴な疑問がこ
の発表内容のプロジェクトを立ち上げた動機であった。
TRPV2は 1999年に 52℃以上の侵害熱刺激を感知する熱センサーとしてクローニングされた。と
ころが、胎生期マウスを用い、脊髄及び DRGにおけるこのチャネルの発現時期を調べると、なんと、
未成熟な神経細胞の出現にあわせて胎生期において既に TRPV2発現は開始していた。子宮内の胎仔
が 52℃以上の侵害熱刺激に遭遇する機会はないため、熱刺激以外の TRPV2リガンドが存在し、発達
期には熱センサーとは全く異なる役割を持つと考えた。そして、解析を進めていくと、胎仔期の
TRPV2は脊髄運動神経・DRG感覚神経が、末梢(皮膚・筋肉など)に向けて非常に長い軸索を伸長
している時に細胞膜にかかる膜伸展刺激で活性化し、軸索伸長を促進させていることを突き止めた
(Shibasaki et al., J. Neurosci. 2010)。伸長中の軸索自身が自分をより伸ばそうと、膜伸展刺
激を用いたポジティブフィードバックを行っていることを明らかにした。現在、TRPV2CKOマウスの
表現型解析や in ovoエレクトロポーレーションを応用したメカノセンサー機能解析と共に、これ
らの知見を応用した軸索再生の実用化研究を進めている。

P05 ドレブリンノックアウトマウスを用いた成体新生ニューロンの解析

海馬歯状回及び、脳室下帯は成体脳でニューロン新生が確認されている脳部位である。海馬歯状
回での新生ニューロンは顆粒細胞層の神経回路に組み込まれ、学習や記憶などに重要な役割を持っ
ていると考えられている。脳室下帯で作られた新生ニューロンは吻側移動経路(RMS)を通り、嗅
球へ移動し、そこで顆粒細胞へ分化して嗅覚の情報処理にはたらく。これまでの研究で、脳室下帯
の新生ニューロンには、発達期の脳に多く存在するアクチン結合蛋白ドレブリン Eが発現すること
がわかっている。本研究では新生ニューロンにおけるドレブリン Eの機能的な役割を知るため、ド
レブリンノックアウト(DXKO)マウスを使用し、海馬歯状回及び、脳室下帯、RMS における新生ニュ
ーロン数を調べた。深麻酔後、灌流固定を行い、固定脳を凍結後、前顎断で凍結切片を作成し、抗
ダブルコルチン(DCX)抗体で新生ニューロンを標識し、免疫陽性シグナルを染色した。陽性細胞
数を比較したところ、DXKOマウスの RMSで、有意に少なかった。海馬歯状回や脳室下帯での陽性細
胞数に差がないことから、ドレブリン Eは新生ニューロンの分裂、増殖や生存、には関与していな
いと考えられる。また、海馬歯状回の各切断部位での差も見られないことから、新生ニューロンの
分化にも関与していないと考えられる。よって、RMS において、DCX 陽性細胞数の減少が見られた
ことは、ドレブリン Eが新生ニューロンの移動に関係している可能性を示唆する。

P06 The dorsoventral boundary of the germinal zone is a specialized niche for the generation of
cortical oligodendrocytes during a restricted temporal window

Oligodendrocyte precursor cells (OPCs) appear in the late embryonic brain, mature to become
oligodendrocytes (OLs) and form myelin in the postnatal brain. Recently, it has been proposed that early-born
OPCs derived from the ventral forebrain are eradicated postnatally and that late-born OLs predominate in the
cortex of the adult mouse brain. However, the types of intrinsic and extrinsic factors that specify the ability of
self-renewing multipotent neural stem cells in the ventricular zone/sub-ventricular zone (VZ/SVZ) of the
embryonic brain to generate cortical OL-lineage cells remain unknown. Using an inducible Cre-loxP system to
permanently label Nestin- and Olig2-lineage cells, we identified that cortical OL-lineage cells start
differentiating from neural stem cells within a restricted temporal window just prior to E16.5 through P10. We
then showed, by means of electroporation of a Cre expression plasmid into the VZ/SVZ of E15.5 reporter
mouse brains, that neural precursor cells in the dorsal VZ/SVZ are inhibited by Wnt signaling from
contributing to cortical OLs in the adult brain. In contrast, neural precursor cells present in the dorsoventral
boundary VZ/SVZ produce a significant amount of OLs in the adult cortex. Our results suggest that neural
stem cells at this boundary are uniquely specialized to produce myelin-forming OLs in the cortex.

It has been suggested that neurons, glia, and capillaries are organized into a neurovascular unit, which is
involved in the regulation of cerebral blood flow. Molecules that have crucial functions in both nervous and
vascular systems have attracted keen attention recently, and the name "angioneurins" has been proposed. The
most striking example of angioneurins is vascular endothelial growth factor A (VEGF), which was originally
identified as a key regulator of angiogenesis and has only recently been found to have important functions in
the nervous system. In this study, we compared VEGF expression in the vasculature in the brain with that in
the aorta and the vasculature in the kidney in mice. In larger vessels containing smooth muscle cells, VEGF
was expressed by smooth muscle cells covering the lining of endothelial cells, both in and outside the brain. In
cerebral capillaries lacking smooth muscle cells, endothelial cells were closely covered by VEGF-expressing
foot processes of astrocytes, whereas capillaries were surrounded by VEGF-expressing processes of podocytes
in the renal glomeruli. We also found that cultured cerebral capillary endothelial cells do not express VEGF,
whereas cultured cortical astrocytes do express VEGF. Specific expression of VEGF in the astrocytic foot
processes covering capillaries in the neurovascular unit suggests that VEGF plays important roles in the unit.
Elucidation of these roles may contribute not only to understanding of physiology of the neurovascular unit but
also to developing the therapeutic strategy for neurodegenerative diseases.

P08 Disruption of mGluR-mediated signaling at cerebellar parallel fiber-Purkinje cell synapses in
staggerer mutant mice

The staggerer mutant mouse is a classical natural strain with deletion mutation of a transcription factor,
Retinoid-related Orphan Receptor α (RORα), which is abundantly expressed in Purkinje cells (PCs) of the
cerebellum. Homozygous staggerer (sg/sg ) mice show inherent ataxia and serve as an important extreme
mouse model of the hereditary spinocerebellar ataxia type 1 (SCA1), since RORα dysfunction is strongly
correlated with SCA1 pathology. However, synaptic abnormalities at parallel fibre (PF)-PC synapses have not
been examined in detail electrophysiologically in SCA1-related sg/sg mice. In this study, we report that fast
PF-PC synaptic transmission is still reasonably preserved in sg/sg mice, in spite of reduction in the number of
PF-PC synapses. In contrast, sg/sg mice lack metabotropic glutamate receptor (mGluR)-mediated slow
synaptic responses completely. Neither intense PF stimulation nor exogenous mGluR agonist, DHPG, could
induce mGluR-mediated slow currents. Western blot analysis of sg/sg cerebella revealed greatly reduced
expression of mGluR1 and TRPC3, both of which underlie mGluR-mediated slow currents in PCs.
Immunohistochemical data demonstrated mislocalization of mGluR1 and absence of TRPC3 in sg/sg PCs.
Moreover, we found that mGluR-mediated retrograde synaptic modulation of PF-PC EPSCs by
endocannabinoid is also abolished in sg/sg mice. These results suggest that disruption of mGluR signaling is
one of the major defects at PF-PC synapses in sg/sg mice and may underlie SCA1 pathology.

Cerebellar Purkinje cells (PCs) express a large amount of the isoform of protein kinase C (PKCγ) and a modest
level of PKCα. The PKCγ is involved in the pruning of climbing fiber (CF) synapses from developing PCs,
and PKCα plays a critical role in long-term depression (LTD) at parallel fiber (PF)-PC synapses. Moreover, the
PKC signaling in PCs negatively modulates the nonselective transient receptor potential cation channel type 3
(TRPC3), the opening of which elicits slow EPSCs at PF-PC synapses. Autosomal dominant spinocerebellar
ataxia type 14 (SCA14) is caused by mutations in PKCγ. To clarify the pathology of this disorder, mutant
(S119P) PKCγ tagged with GFP was lentivirally expressed in developing and mature mouse PCs in vivo, and
the effects were assessed 3 weeks after the injection. Mutant PKCγ-GFP aggregated in PCs without signs of
degeneration. Electrophysiology results showed impaired pruning of CF synapses from developing PCs,
failure of LTD expression, and increases in slow EPSC amplitude. We also found that mutant PKCγ
colocalized with wild-type PKCγ, which suggests that mutant PKCγ acts in a dominant-negative manner on
wild-type PKCγ. In contrast, PKCα did not colocalize with mutant PKCγ. The membrane residence time of
PKCα after depolarization-induced translocation, however, was significantly decreased when it was present
with the mutant PKCγ construct. These results suggest that mutant PKCγ in PCs of SCA14 patients could
differentially impair the membrane translocation kinetics of wild-type γ and α PKCs, which would disrupt
synapse pruning, synaptic plasticity, and synaptic transmission.

P10 Isolation and characterization of CD44-positive cells: candidate cells for astrocyte
precursors in the developing mouse cerebellum

Neural stem cells are generally considered to be committed to becoming precursor cells before terminally
differentiating into either neurons or glial cells during neural development. Neuronal or oligodendrocyte
precursor cells have been isolated and studied extensively, but the development of astrocytes remains largely
unknown. In this study, we report isolation of APC candidates from the developing mouse cerebellum. CD44-
strongly-positive cells were isolated from mouse cerebellum on postnatal day 3 using fluorescence-activated
cell sorting, and characterized in vitro. The isolated cells expressed brain-lipid binding protein (BLBP), a
marker for immature astrocyte lineage cells, but neither glial fibrillary acidic protein (GFAP) nor S100-,
markers for mature astrocytes. When the cells were cultured in the absence of any signaling molecule, many
cells died by apoptosis. The surviving cells gradually expressed GFAP, indicating that differentiation into
mature astrocytes is the default program for these cells. These cells showed neither of the two cardinal
characteristics of neural stem cells; the ability to self-renew and the ability to produce neurons, astrocytes, and
oligodendrocytes. Leukemia inhibitory factor greatly promoted astrocytic differentiation of CD44-positive
cells. In the presence of fibroblast growth factor-2 (FGF-2), many cells incorporated BrdU, suggesting that
FGF-2 is a potent mitogen for these cells. Many of these cells, however, died by apoptosis, suggesting FGF-2
alone is insufficient for their survival. In the presence of both FGF-2 and bone morphogenetic protein-4
(BMP4), the cells maintained their survival and proliferation. BMP4 inhibited activation of caspase-3 and
greatly promoted survival, suggesting a novel role for BMP4 in the control of development of astrocytes in
cerebellum. Characterization of these cells may be useful for understanding the mechanism for generation of
various astrocytes during cerebellar development.

P11 発生期小脳における CD44発現細胞の分布

1)目的
中枢神経系は、神経幹細胞に由来するニューロン、アストロサイト、オリゴデンドロサイトによ
って構成されている。我々は FACSによって単離した CD44陽性細胞がアストロサイト前駆細胞の性
質を示すことを報告した (Cai et al., 2011)。 本研究では、CD44 がアストロサイト前駆細胞の
マーカーとなり得るか否かを検討するため、発生期小脳における CD44の発現を解析した。
2)方法
ICRマウス小脳の凍結切片を作製、in situ hybridization法と免疫蛍光染色法を用いて、in vivo
での発生期小脳における CD44発現を検討した。この際、各種細胞マーカーとの二重染色及び BrdU
取り込みによる分裂能の検討も行った。
3)結果と考察
生後0日目(P0)、P3では多くの細胞が CD44陽性であったが、P7ではハーグマングリアと白質に
存在する細胞に強い発現が観察された。これらは GLAST(神経幹細胞とアストロサイト系譜のマー
カー)を発現していた。P14 以降ではバーグマングリアの GFAP(成熟アストロサイトのマーカー)
発現が強くなる一方で CD44発現は弱くなるのに対して、白質に存在する細胞の CD44発現は維持さ
れた。これらは GFAP強陽性の成熟線維性アストロサイトと SOX2陽性の神経幹細胞と考えられるが、
発生が進むにつれて白質内の CD44 陽性細胞中の SOX2 陽性神経幹細胞の割合は減少した。また P0
から P10では CD44発現細胞の一部が BrdU陽性であったが、P14以降では CD44発現細胞は BrdUを
取り込まず、細胞周期を脱していることが示唆された。

細胞質型蛋白質チロシンホスファターゼ Shp2は、増殖因子の下流において Ras/MAPKカスケードの
活性化に重要な役割を果たすことが示されている。またヒト Shp2の遺伝子異常は、精神発達遅滞、
先天性心疾患、骨格異常などを特徴とするNoonan症候群の原因となることが明らかとされている。
Shp2は組織普遍的に発現しており、脳では Shp2が発生期の神経分化の制御に重要な役割を果たす。
一方、Shp2 は成熟脳にも強く発現するが、Shp2 の全身性遺伝子破壊(KO)マウスは胎生致死とな
るため、成熟脳における Shp2の機能は十分に検討されていない。そこで我々は、Cre-LoxPシステ
ムを利用して成熟前脳神経細胞特異的な Shp2 コンディショナル KO(cKO)マウスを作製し、成熟
脳の機能制御への Shp2の関与について検討を行った。Shp2 cKOマウスは正常に生まれ、脳の構造
にも異常は認められなかった。一方、行動解析の結果、Shp2 cKOマウスは、強制水泳テスト、明暗
箱テスト、オープンフィールドテストなどの行動テストにおいて異常が認められたが、それらはい
ずれも Shp2 cKOマウスが高い活動性(多動)を示すことに起因しており、実際に、Shp2 cKOマウ
スはホームケージでも多動を示すことが明らかとなった。現在、Shp2 cKO マウスの多動について、
その分子メカニズムの解析を進めている。

brain circuit development and autism-related behavior in a mouse model

Autism is a pervasive disorder of brain development characterized by impaired social
interaction and communication. The potential molecular risk factors are poorly characterized in
animal models. We previously identified dex3, a rare splice variant of CADPS2 (also termed
CAPS2), a protein essential for enhanced release of brain-derived neurotrophic factor (BDNF)
from neurons, which is missing an exon critical for axonal transport in in vitro neuronal
cultures. CADPS2-dex3 is overrepresented in patients with autism. Here, we generated
Cadps2-dex3 mice and demonstrated a severe impairment of in vivo axonal transport and
BDNF release. Because of this specific deficit, circuit connectivity, measured by spine and
interneuron density, was globally diminished. The collective impact of reduced axonal BDNF
release during development was a striking and selective repertoire of deficits in social- and
anxiety-related behaviors. Together, these findings represent the first mouse model of a
molecular mechanism linking BDNF-mediated coordination of brain development to
autism-related behaviors and patient genotype.

P14 X線耐性獲得細胞における重粒子線感受性

[目的] X線耐性獲得細胞における重粒子線感受性を明らかにすることを目的とした。
[方法] 1日 2 Gyの X線照射(総線量 2,278 Gy)を続けても生存・増殖し続けたヒト舌扁平上皮が
ん SAS-R細胞とその親株 SAS細胞を用いた。X線照射装置(TITAN-225S, Shimadzu)にて X線(200 kV,
14.6 mA) または群馬大学重粒子線照射装置にて 6cm-SOBP 炭素線(290 MeV/u, ~50 keV/μm)を照
射した。放射線感受性は高密度生存アッセイ法で調べた。また、がん幹細胞マーカーCD44 および
CD326 の蛍光染色によりフローサイトメトリーでがん幹細胞様集団と非がん幹細胞様集団を分画し
て、X線と炭素線照射後の存在比を比較した。
[結果] X線の 20%生存率線量は SAS細胞で 7.5 Gyに対して、SAS-R細胞で 12 Gyと 1.6倍抵抗性を
示した。炭素線の 20%生存率線量は両細胞でほぼ同じで、SAS細胞で 3.0 Gy、SAS-R細胞で 3.2 Gy
であった。この結果から求めた RBEは SAS細胞で 2.5、SAS-R細胞で 3.8を示した。CD44および CD326
ダブルポジティブの割合は非照射時において SAS細胞に比べて SAS-R細胞で高く、等生存率線量の
炭素線に比べて X線で高かった。
[結語] 重粒子線は X線耐性獲得がん細胞に対しても有効であることが示唆された。

P15 線虫の全身運動に対する重イオンマイクロビーム照射の影響

研究目的:放射線被ばくの生体影響を考える際には、発がんリスクと共に運動機能や高次神経機能に対
する影響を知ることも重要である。発表者らは、モデル生物線虫を用いて運動機能に対する放射線影響
を調べ、全身照射によって全身運動(移動)が一時的に低下することを明らかにした。しかし、この結果の
みからでは、放射線による運動低下が、全身の組織への影響によるものか一部の細胞への影響によるも
のかを判別できなかった。そこで、本研究では、線虫の体のごく一部を狙ってマイクロビームを照射し、全
身運動に対する放射線影響が照射部位によって異なるか否かを調べることを目的とした。
方法:原子力機構のマイクロビーム細胞局部照射装置を用いて、線虫(成虫)の頭部、中央部、尾部の細
胞群を狙って直径 20 m径の炭素イオンマイクロビームを照射した。各個体 1箇所のみを狙い、各部位
への照射実験を独立して行った。照射直後に線虫を寒天平板上に移して運動を撮影し、動画をもとに 10
秒間の頭部屈曲回数(全身運動の評価指標)を計数した。
結果:頭部照射群、中央部照射群、尾部照射群の全身運動は、いずれも非照射群に対して有意に低下し
た。低下の度合いには照射群間で差が無かった(予備的な結果)。以上から、体の一部だけでも照射され
れば、運動が低下することが明らかになった。今後は、更にマイクロビーム照射実験を重ね、照射部位と
放射線影響との関係を詳細に調べていく。

P16 X-ray irradiation induces acute depolymerization of axonal and dendritic microfilaments in
cultured neuron

Purpose: Only a little has been reported about the effect of irradiation on neuronal cells. In axon and
dendrite, cytoskeletons including microtubules, microfilaments and neurofilaments form the backbone and
scaffold of neurons that coexist in the cytoplasm. The cytoskeleton proteins play a pivotal role in maintaining
the appropriate dendritic spine morphology and providing synaptic function. In this study, we irradiated
cultured neurons with x-ray and observed the structural changes of axon and dendrite.
Materials and methods: Primary hippocampal neurons were prepared from hippocampus of Wistar rats on
embryonic day 18 by Banker's methods. Neurons were incubated for 21 days and then irradiated with 30 to
90 Gy of X-ray. After irradiation, they were fixed immediately. The morphology of axon and dendrite were
examined by scanning electron microscopy (SEM). Neurons were stained with filamentous actin (F-actin),
and were observed by confocal laser scanning microscope.
Results: At 30 Gy, structural changes in axons and dendrites could be observed by SEM. In the region
which several axons and dendrites were bundled together, irradiated neurons showed the loss of connectivity
between axons and dendrites compared with the non-irradiated control. In un-bundled axons and dendrites,
the increase of tortuosity and decrease of diameter were seen following irradiation. The fluorescence
intensity of F-actin in the axons and dendrites was markedly reduced in the 30 Gy irradiated neurons.
Conclusion: These data suggest that x-irradiation evokes depolymerization of microfilament and
immediately changes the structure of axons and dendrites.

P17 The effects of X-irradiation on proliferation and differentiation of mouse ES-cell-derived
neural stem cells

Normal brain formation requires regionally and temporally appropriate proliferation and differentiation of
neural stem cells (NSCs) into neurons and glia. Exposure of the fetal brain to ionizing radiation cause
congenital brain abnormalities, which is thought to result from defects in neurogenesis during the developing
stage. Here, we investigated the effects of X-irradiation on proliferating homogenous NSCs, which were
prepared from mouse embryonic stem cells by the Neural Stem Sphere method. Cells cultured in the
proliferation medium, containing fibroblast growth factor-2 (FGF-2) were irradiated with X-rays and analyzed
with growth curve measurements, flow cytometry, real-time RT-PCR, and immuno-fluorescence. The cells
irradiated at a dose of > 5 Gy did not show an increase in number, but a gradual decrease was observed. The
gradual decrease was explained by the loss of proliferation capability and also apoptosis. In contrast, the cells
irradiated at a dose of 1 Gy stopped proliferation temporarily but recovered the proliferation capability.
Moreover, sub-cultured at 96 hrs after irradiation, the cells proliferated logarithmically in the proliferation
medium, and successfully differentiated into neurons and astrocytes in the differentiation medium. These
results clearly demonstrate that exposure of the fetal brain to radiation at relatively high doses leads to
congenital morphological and functional brain abnormalities. In addition, because brain development is a very
stringently coordinated process from a clonological and regional viewpoint, these results suggest that the delay
in cellular proliferation of NSCs after irradiation at relatively low doses may cause congenital brain
abnormalities.

P18 神経膠芽腫細胞における X線照射により誘導された遊走能の分子機構の解明

放射線治療は高精度放射線治療技術の発達により、腫瘍の局所制御は向上したものの、浸潤制御
は未だ満足のいく成果は得られていない。放射線照射によって腫瘍細胞の遊走能が亢進するという
報告はあるが、その分子機構についてはよくわかっていなかった。そこで、悪性度の高い神経膠芽
腫細胞における X線誘導遊走能の分子機構を解明することを本研究の目的とした。
ヒト神経膠芽腫細胞に対し、X 線照射(200 kVp, 14.6 mA)を行った。X 線照射 24 時間後に
wound-healing assay法を用いて解析した結果、細胞の遊走能は線量依存的に亢進した。照射後 24
時間以内に培養液中に放出されるグルタミン酸量を測定した結果、線量依存的に増加した。そこで、
グルタミン酸受容体の 1 つである α-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA)
型グルタミン酸受容体の拮抗薬を添加し、1時間後に X線照射したところ、遊走能の亢進が抑制さ
れた。
以上の結果から、X 線照射による神経膠芽腫細胞の遊走能の亢進にはグルタミン酸が関与してい
ることが示唆され、AMPA型グルタミン酸受容体拮抗薬は放射線治療において有用な遊走阻害剤とな
る可能性が示唆された。

P19 炭素線照射による脳の生物学的反応における組織学的評価

脳の放射線治療による生物学的反応は、照射後数週間以内に生じる急性期反応、照射後数ヶ月以
内に生じる早期遅発性反応、照射後 6ヶ月以降に生じる晩期遅発性反応の 3つに分類されることが
わかっている。これまでに、ガンマ線やX線によるこれら反応の組織学的検討は多くされてきたが、
重粒子線による脳の生物学的反応はよくわかっていない。そこで、我々は重粒子線照射による脳の
生物学的反応を調べることを本研究の目的とした。
タングステン製マルチリーフコリメータを利用し,右脳の Bregmaより+2から-8 mm(照射野 10 mm)
を照射した。照射深度は,拡大ブラッグピーク 5 mm-SOBP(290 keV/μm)を用いて,視床下部や脳
幹を避けて頭頂部より 5 mmまで照射した。
今回は照射後 1ヶ月の脳の生物学的反応について組織学的に評価した。GFAP抗体で染色した結果、
10 Gyおよび 30 Gy照射した脳では、同一個体の非照射脳よりも、明らかな反応性アスロトサイト
の増勢が認められた。また、同部位においてグルタミン酸の発現増加が認められた。このことから、
照射による脳の炎症反応にはグルタミン酸が関与していることが示唆された。

P20 中枢神経原発悪性リンパ腫の放射線治療成績

【目的】当院における中枢神経原発悪性リンパ腫の放射線治療成績について検討した.
【対象と方法】1990年から 2009年までに,群馬大学医学部附属病院にて放射線治療が施行された
中枢神経原発悪性リンパ腫 64名を対象とした.46名が病理組織学的に診断され,その他は臨床的
に診断された.年齢は 24-81歳(中央値 64歳),性別は男:女=41:23,治療前 PSは PS0: 1: 2: 3:
4= 3: 19: 12: 10: 20であった.放射線治療は,原則,全脳照射 40Gy/20回行った後,局所に絞り
10Gy/5回を照射した.なお,放射線治療前に高用量メソトレキセート(MTX)療法を 2コース以上
施行した 16症例では全脳照射を 30Gy/15回に減量した.
【結果】観察期間中央値は 19.4ヶ月(0.7〜115.5ヶ月)で,全患者の生存期間中央値は 32.5ヶ月
であった.PS0〜2の患者の生存期間中央値は 38.1ヶ月で,PS3〜4の 8.3ヶ月と比較して有意に
予後良好であった(p< 0.0001). また高用量 MTX療法を導入した 2001年以降では,MTXを施行し
た患者 19名の生存期間中央値は 38.1ヶ月で,照射単独(18名)の 21.1ヶ月と比較して有意に予
後良好であった(p=0.02).
【結語】他の部位の悪性リンパ腫に比べ,中枢神経原発悪性リンパ腫の放射線治療成績は依然不良
であり,治療法の改善が望まれる.

P21 WHO grade 3 gliomaにおける組織亜型と予後に関する解析

【目的】WHO grade 3 gliomaは組織亜型によって,予後や治療効果に差があることが報告されてい
るが,いまだ十分な検討は少ない.
【方法】1984 年から 2001 年に当院で放射線治療が施行された grade 3 Glioma の 68 例(男性 31
例,女性 37例)を対象に,組織亜型や臨床因子(年齢・性別・線量・摘出度)について予後との相関
を検討した.
【結果】組織亜型は anaplastic astrocytoma (AA)41例,anaplastic oligodendroglioma (AO)16
例および anaplastic oligoastrocytoma (AOA) 11 例であった.線量の中央値は 60 Gy であった.
全症例の 5年全生存率は 34%で,生存期間中央値は 32ヶ月であった.組織亜型別の 5年全生存率は
AAで 21%,AOAで 38% AOで 80%であり,AAに比べ AOおよび AOAは有意に予後良好であった(P=0.01,
P<0.01).摘出度は有意な予後因子であったが(P<0.01),線量や性別は予後因子とはならなかった.
年齢については,50歳以上と 50歳未満の群では,50歳未満の群が予後良好であったが,有意差は
認めなかった(P=0.09).多変量解析では組織亜型が最も強い予後因子であり,次いで摘出度であ
った.
【結語】WHO grade 3 gliomaの放射線治療は組織亜型についで摘出度が強い予後因子であった.
