Career Profile

Dr. Waseem Ahmad (Faridi) is a permanent faculty; a Professor of Genetics and Molecular Biology in the Faculty of Life Science, Department of Zoology, Aligarh Muslim University, Aligarh (UP); has a wide experience of teaching and research nationally and internationally. He has also worked in Saudi Arabia and Asmara in various capacities. His work on Molecular Genotoxicity and Genomic Medicine is evidenced by many awards, grants and research papers in the peer reviewed journals of national and international repute. He has also acted as consultant to many research organizations and authored many books. Recently he authored a book on ‘Genetics and Genomics’ for Pearson Publication Incorporation.

ABSTRACT: MS/Thesis, Ph.D Dissertation & Post doctorate Research

Mosquitoes being vectors of many diseases and have great potential for human

exposure, the doctoral work taken therefore. The sentinel genetic potential was

harnessed with an aim to contribute in mosquito controlling programmes. The

seasonal variations of different genera of mosquitoes, epidemiology of certain

diseases, new mutants, and the problems with respect to some agricultural

pesticides were done. To begin with, immature stages of various genera was

determined. The population of Culex was found to be constantly higher than Aedes

and Anopheles. The work was extended to study the epidemiological studies and

incidence of malaria and other mosquito borne diseases at different seasons. The

disease index for malaria was significantly higher in younger group with a gender

bias.

The populations showed great genetic plasticity. Mutations affecting structural

modifications of various organs were isolated. Thus, hb, ra, ab, bg, bs and bi were

analyzed for inheritance pattern, frequency distribution and test statistics (crs).

Linkage group study was made with respect to lp4 gene. The origin of

gynandromorphism (sgm, gm) on the basis of bi-nucleated eggs bearing an m

factor and independently fertilized by male gametes of M and m genotype was

postulated.

In addition, a comprehensive assessment of potential hazards of chlorinated

phenoxy compounds (2,4-D, PCP, 2,4,5-T) was embarked upon. Principal objective

of the study was to assess the mutagenic activity using wide range of end point

parameters. Anti-mutagenic studies for repairing the chromosomal and DNA

damage by plant extract showed promising results. Comparative studies were

made with many mutagens. Electromagnetic radiations, including full solar eclipse

radiations were subjected to test for making sterile release program possible in

controlling the mosquito species. Some mutants discovered during the course of

study such as d and bb being lethal genes, showed a great potential for restricting

these vectors. In addition, predation behavior of Culex (Lutzia) raptor larvae on the

aquatic stages of Culex , Aedes and Anopheles showed a great promise in

biological control.

Environmental and Molecular Genotoxicity

Our group is engaged in active research on in-vivo and in-vitro genotoxicity of aquatic organisms. Histopathological studies on germinal epithelium, clastogenic effects on various cell types and biochemical lesions of DNA have invariably shown remarkable results in catfishes (Chem. Env. Res., 5, 267-281, 1996; Ind. J. Exp. Biol. 36, 304-307, 1998). The practical value of micronucleus assay aided by Computer Image Analysis (CIA) have shown better computing statistics and more objective assessment of micronuclie (Chromosoma, 110, 570-574, 2002). Studies have further focussed on electrophoretic reactivity as the fundamental cause of toxicity and stress of some xenobiotics on aquatic organisms (Chemosphere, 55, 257-265, 2004). Cytochemical estimation of protein and DNA revealed a significant variation of bands (Chem. Environ. Res., 5, 267-281, 1996). Pentachlorophenol - mediated cellular and changes in germinal epithelium and kidney cells of H. fossilis have shown ample evidence of apoptosis (Proceedings of Environ. Mutagen Society, Lucknow, 19-21 March, 2002, Proceedings of Environ. Mutagen Soc., 2010, J. Life Sciences, USA, accepted, 2011). Research is further extended to herbicides for apoptotic and mutagenic evaluation (Saudi J. Biol. Sci., 2012). Techniques such as comet assay, DNA diffusion method, MNT (CIA) are being incorporated. Inhibitory activity of some endogenous substances (neem and vitamins) viz-a-viz chemicals used, is in progress for the assessment of antimutagenicity (STOTEN, Science of Total Environment, Elsevier, 2005). Detection of DNA damage by alkaline single cell gell electrophrosis is the sensitive parameter used successfully against some chemicals (Ecotoxicology and Environmental Safety, Elsevier, 2005).The work is further extended in other test system, Rattus norvegicus and Mus musculas (Ecotoxicology and Environmental Safety, Elsevier, 2006, Saudi J. Biol. Sci., 2012 ) and research work is earnestly persued along these lines since 2010-14.

Other aspects like oxidative damage and cytoarchitural assesments (Environt. Toxicol., 2014), In silico studies (Biotech & Appl. Biochem.m 2014) have been investigated. Genotoxic studies expanded further with newer parameters and organisms with protective measures (Int. Sch. Res., 2014; Int. J. Cytol., 2013; Toxicol., 2013; Sci. Dilliman, 2012; Eco. & Env. Safety, 2006). More recently commet assay, mitochondrial membrane potential and other DNA studies have been further incorporated (2014). Genomic Medicine

Active researches in CEGMR earnestly started on Cancer Genomic, Stem cell, Neurogenetics and PGD. Many international researches were also admitted in the Center. As a result number of research employing molecular parameters have started yielding results. The cytogenetic profiles of Down’s syndrome by G banding presented a very detailed insight of comparative studies (Sp J Biochem Mol Biol, vol3, no1, 2008, J. Med. Sciences, US, China, 2010. Similarly genetic disorders and implication of chromosomal abnormalities have been analysed very elaborately giving incidence, pattern and influencing factors (JKAU: Med. Sci., vol 15, no 3, 2008; Proc. Env. Mut. Soc., 2010). All these studies are unique in relation to Saudi Arabia. FLT3 oncogene mutations in acute myeloid leukaemia has been analysed

successfully with the help of conformation sensitive gel electrophoresis (Int. J. Mol. Sci., 2008). Frequency of genetic disorder, G6PD Mediterranean mutations among a group of Saudi patients have given new insight about its pattern in Jeddah region (Int. J. Lab. Hem.,2008). Recently a study on Molecular Genetics of breast cancer using HER_2/neu gene amplification by FISH and IHC has been concluded( joint study with Faculty of Medicine, KAU, Jeddah, CEGMR, KFMRC, 2010). In addition, analysis on serum protein and enzyme levels in human subjects with osteoarthiritis have given interesting results (Med. Biochem., 30:15-24, 2011). In silico and in vivo studies show significance response in human brain tissue for certain enzymes (Biotech & Appl. Biochem., 2014)

Research Background

Exposed to the state of the art laboratories of Molecular cytogenetic techniques such as comparative genomic hybridization (CGH), microarray, immunehistochemistry (IHC) and PCR at Genomic Medicine Unit, GMU, Center of Excellence in Genomic Medicine Research, CEGMR, King Fahd Medical Research Center, KFMRC, King Abdulaziz University, Jeddah, Saudi Arabia. At the Centre For Advanced Studies on Animal Genetics and Breeding, NDRI (ICAR), KARNAL, different methods of Animal cell and tissue culture technology were learned including in vitro culture of mammalian embryos, culture of bovine gametes and neonatal rat skin of fibroblasts and maintenance and preservation of cell lines. Special attention was paid for training in DNA fingerprinting and isolation and characterization of DNA at the Biotechnology Division of the National Bureau of Animal Genetic Resources, Karnal, Haryana. Also initially obtained practical training on various aspects of genetics and cytogenetics at the Department of Zoology (AMU), Aligarh. The training and international exposure of various laboratories has made the author well versed with latest Biological, Medical and Molecular Cytogenetic techniques in the field of Genomic Medicine and Molecular Genotoxicity.

Visits to Labs Engaged in Mosquito Research

(During Doctoral Program) Visited Bangalore University, Department of Zoology where pioneer work on cytogenetics was in progress under the leadership of Prof. B. N. Chowdaiah in Culex pipiens fatigans. Interacted with the pioneer scientists like Subbarao S. K., Adak T., Krishnamurthy B. S. in Malaria Research Center (VCRC) – a joint venture of WHO and ICMR. The team produced outstanding work on cytoplasmic incompatibility. Also visited Punjab University, Chandigarh where Sudershan C. and his team was engaged in Cytogenetics and somatic cell chromosome studies on mosquitoes. Our own lab under the stewardship of late Prof. Adhami U. M., who obtained his doctorate from Notre Dame University, USA had done extensive work on yellow larva in Aedes aegypti. We were exposed to high quality research work (under the aegis of WHO Research Project), were engaged in Insecticide resistance and induced mutations in Aedes aegypti and Culex fatigans. There was an active coordination between our lab and other lab of reputes in America (Kitzmiller, J. G. Department of Zoology, University of Illinois, Craig G. B., Rai K. S., Adhami U. There was a brief association and thought of collaboration of controlling the dengue fever mosquitoes with Prof. Khalid Al Ghamdi, WHO Project Director and Prof. of Biology at KAU, Jeddah. Although offered as Advisor to WHO project, but could not collaborate as my extra ordinary leaves ended and I was to join my parental university.
