RESUMEN
Sechium edule (chayote) es una especie neotropical con
amplia variacin biolgica en forma, color y sabor de
los frutos, los cuales han sido utilizados como fuente de
alimento y medicina. En esta investigacin se usaron frutos
de S. edule var. nigrum spinosumpara evaluar la actividad
antiproliferativa y citotxica en dos lneas tumorales (L-929
y HeLa); adems, de aislar y caracterizar los componentes demayor actividad a partir de un extracto alcohlico. El extracto
se fraccion por cromatografa en columna, los ensayos
biolgicos se realizaron con el extracto y las fracciones
principales evaluando seis concentraciones (0, 0.047, 0.23,
0.47, 1.18, 2.37 mgmL-1). De cuatro fracciones aisladas, tres
tuvieron actividad antiproliferativa y citotxica de tipo dosis
dependiente. El anlisis por resonancia magntica nuclear de
hidrgeno y cromatografa de gases acoplada a espectrometra
de masas, revel como componentes mayoritarios de la
fraccin ms activa a steres de tipo metlico de cido
hexadecanoico, etlico del cido hexadecanoico, metlico del
cido 10-octadecenoico, metlico del cido octadecanoico,octlico del cido octadecenoico y metlico del cido 9-oxo-
nonanoico, ampliando con ello la gama de especies vegetales
con posibilidad de detener la proliferacin de clulas
cancergenas.

ABSTRACT
Sechium edule (chayote) is a Neotropic species with wid
biologic variation of form, color, and fruits flavor fruits
which has been utilized as source of food and medicine. In
this research, fruits of S. edule var. nigrum spinosum wer
used in order to assess antiproliferative and cytotoxic activit
in two tumoral lines (L-929 and HeLa), besides for isolatin
and characterizing the components of most importanactivity starting from an alcoholic extract. e extract wa
fractionated by column chromatography; the bioassays wer
carried out using the extract and the main fractions assessin
six concentrations (0, 0.047, 0.23, 0.47, 1.18, 2.37 mg mL-1)
ree out of four isolated fractions had antiproliferative an
cytotoxic dose-dependent activity. e analysis by hydrogen
nuclear magnetic resonance and gas chromatography
coupled to mass spectrometry, revealed esters of: methyl o
hexadecanoic acid, ethyl of hexadecanoic acid, methyl o
10-octadecenoic acid, methyl of octadecenoic acid, octyl o
octadecenoic acid, and methyl of 9-oxononanoic methyl aci
as the principal components of the most active fraction, thuextending the range of vegetal species with the possibility o
blocking proliferation of carcinogenic cells.

INTRODUCCIN
Los productos naturales provenientes deplantas son valorados por el hombre comofuente de principios activos para usomedicinal (Farnsworth et al., 1992) debido a susconstituyentes qumicos, generalmente metabolitossecundarios (Bye et al., 1992), que en la naturalezafuncionan como defensa qumica contra herbvoros.La complejidad estructural de estos metabolitospuede determinar su actividad biolgica; por loque representan un recurso para el desarrollo denuevos frmacos (Hostettmann et al., 1995). La
bsqueda de medicamentos naturales surge por elincremento en la frecuencia de padecimientos comoel cncer. ste se ha extendidos en el panoramaepidemiolgico mundial desde finales del siglo XX,con un registro de 22 millones de casos. En Mxico,el cncer ocupa desde 1990 el segundo lugar comocausa de muerte (INEGI, 2007); causando en el ao2001, la prdida de 33.5 aos potenciales de vida(RHNM, 2001). En trminos de tratamiento delcncer, existen serias limitaciones debido a la falta deselectividad de los agentes quimioteraputicos y a laresistencia desarrollada por algunas clulas cancerosasa dichos productos (Setzer y Setzer, 2003); por ello,los compuestos obtenidos mediante la extraccinfitoqumica, cobran relevancia como nuevos recursospara tratamientos alternativos y la farmacutica(Mans et al., 2000).
Sechium edule (Cucurbitaceae), comnmentellamado chayote, es una especie neotropical(Newstrom, 1991), estudiada desde el punto de vistataxonmico, ecolgico, nutrimental, agronmico,fisiolgico, qumico y gentico (Lira-Saade yCaballero, 2002; Modgil et al., 2004; Cadena-iguez,2005). El chayote se emplea en la medicina tradicionalcomo diurtico, cardiovascular, antiinflamatorio,
contra calcificaciones renales y arteriosclerosis(Jensen y Lai, 1986). Contiene alcaloides nofenlicos, saponinas, esteroles, triterpenos (Salama etal.,1986) y flavonoides glicosilados (Siciliano et al.,2004). Se le atribuyen propiedades antiinflamatorias(Salama et al., 1987); antihipertensivas (Gordon etal., 2000); antimicrobianas (Ordoez et al., 2003)y antioxidantes (Ordoez et al., 2006), validadascon estudios farmacolgicos. Una caractersticaimportante de S. edule, es su variacin biolgica,observada principalmente en la forma, color y sabor

In terms of cancer treatment, therare serious limitations due to the lack of selectivityof chemotherapeutic agents and to the resistancdeveloped by some cancerous cells to such product(Setzer and Setzer, 2003); therefore, the compoundobtained by means of phytochemical extractionbecome important as new resources for alternativtreatments and pharmaceutics (Mans et al.,2000).
Sechium edule (Cucurbitaceae), usually calledchayote, is a Neotropic species (Newstrom, 1991)studied from the taxonomic, ecological, nutritionalagronomic, physiological, chemical, and genetic poinof view (Lira-Saade and Caballero, 2002; Modgil eal.,2004; Cadena-Iiguez, 2005). Chayote is used intraditional medicine as diuretic, cardiovascular, antiinflammatory cure, against renal calcification, andarteriosclerosis (Jensen and Lai, 1986). It containnonphenolic alkaloids, saponins, sterols, triterpene(Salama et al., 1986), and glycoside flavonoid(Siciliano et al.,2004). Anti-inflammatory (Salamet al., 1987), antihypertensive (Gordon et al2000), antimicrobial (Ordoez et al., 2003), andantioxidant properties (Ordoez et al., 2006) arattributed to chayote, validated by pharmacologicastudies. An important characteristic of S. edule is it
biologic variation, mainly observed in form, colorand flavor of the fruits, the latter attributed to thvariation in tetracyclic triterpene concentrationsuch as cucurbitacins (Cadena-Iiguez et al., 2007a)Starting from the aforementioned and based on thfact that these metabolites have potential activity aantineoplasic agents(Anonymous, 1996), besides thsearch of new uses for this species, Cadena-Iigue(2005) and Cadena-Iiguez et al.(2006) assessedalcoholic fruit extracts of eight varieties in thtumoral lines P-388 and L-929 (monocytic leukemi

de los frutos, atribuido ste ltimo a la variacin
en la concentracin de triterpenos tetraciclicos,tales como las cucurbitacinas (Cadena-iguez etal., 2007a). Partiendo de lo anterior y con base enque estos metabolitos tienen actividad potencialcomo agente antineoplsico (Annimo, 1996),adems de la bsqueda de nuevos usos para estaespecie, Cadena-Iiguez (2005) y Cadena-Iiguezet al. (2006), evaluaron los extractos alcohlicos defrutos de ocho variedades en las lneas tumoralesP-388 y L-929 (leucemia monoctica y fibrosarcomade pulmn respectivamente), de los cuales S. edulevar. nigrum spinosum, mostr la mayor actividadantiproliferativa, sin embargo, no se identificaron losmetabolitos contenidos en el extracto responsables dedicha accin. Con base en lo anterior, en el presentetrabajo se planteo la hiptesis bajo un enfoquebioprospectivo que S. edule var. nigrum spinosumtiene actividad antineoplsica por contener en sucomposicin bioqumica metabolitos del grupo delos triterpenos tetraciclicos; por tal motivo, se evaluun extracto alcohlico de frutos de dicha variedad,con el fin de determinar su actividad en dos lneastumorales, adems de identificar los principalesmetabolitos responsables de la misma, de tal formaque lo anterior permita enriquecer el valor de uso de
este recurso fitogentico Mesoamericano como unalimento funcional.
MATERIALESYMTODOS
Material vegetal, extraccin y aislamiento
Se utilizaron frutos de Sechium edule (Jacq.) Sw., var.nigrum
spinosum, procedentes del Banco Nacional de Germoplasma de
S. edule en Mxico. Esta variedad se desarrolla bajo condiciones
de cultivo, en altitudes que oscilan entre los 800 a 2500 m, con
requerimientos de alta humedad ambiental (80-85 %), suelos
cidos a ligeramente cidos (4.5 a 6.5 de pH), bien drenados y ricosen humus (Flores, 1989; Cadena, et al., 2001), con precipitacin
media anual de 1500 a 2000 mm., fotoperiodo de hasta 12 h luz
para inicio de floracin y temperaturas de 20 a 25 C; es muy
susceptible a la sequa, heladas que generalmente ocasionan
muerte de la planta (Cadena et al., 2001). El ejemplar herborizado
se deposit en el Herbario Ortorio del Colegio de Postgraduados,
Montecillos, Texcoco, Mxico, con la accesin No. 300-2005.
A diferencia del resto de las variedades tanto cultivadas
como en condiciones de traspatio, n. spinosum, presenta en la
composicin de sus frutos hasta diez veces ms contenido de

MATERIALSANDMETHODS
Vegetal Material, Extraction and Isolation
Sechium edule (Jacq.)Sw., var.nigrum spinosumfrom the Banc
Nacional de Germoplasma de S. edulein Mxico was used. i
variety is developed under crop conditions at altitudes betwee
800 and 2500 m, with requirements of high environmenta
humidity (80-85 %), acid and slightly acid soils (4.5 to 6.5 pH)well-drained and rich in humus (Flores, 1989; Cadena et al
2001). With annual mean precipitation of 1500 to 2000 mm
photoperiod of up to 12 h light for the beginning of flowering
and temperatures of 20 to 25 C, it is highly susceptible to drough
and frosts that generally cause plant mortality (Cadena et al
2001). e herbaceous specimen was deposited in the Herbari
Ortorio of the Colegio de Postgraduados, Montecillo, Texcoco
Mexico, with accession No.300-2005.
Unlike the rest of the varieties, cultivated as well as unde
backyard conditions, n. spinosumhas in its fruit composition u
to ten times as much tetracyclic triterpene content as the varietie
of yellow fruits like a. levis and a. dulcis; and presumably thtype of metabolites with respect to the varieties of green fruit
like nigrum levis, virens levis, nigrum xalapensis show highe
antiproliferative activity in the preliminary assays (Cadena
Iiguez, 2005; Cadena-Iiguez et al., 2005). e fruits were use
when reaching horticultural maturity at 182 days after anthes
(Aunget al.,1996; Cadena-Iiguez et al.,2007b), chopped i
small pieces, dried for three days at 50 C up to 10% humidity
and macerated for standardizing at 2 mm particle size (no.1
mesh). Subsequently, a discontinuous extraction was mad

triterpenos tetraciclicos que las variedades de frutos amarillos,
tales como albus minor, a. levis y a. dulcis, y presumiblementeel tipo de metabolitos respecto a las variedades de fruto color
verde como nigrum levis, virens levis , nigrum xalapensis, muestran
mayor actividad antiproliferativa en los ensayos preliminares
(Cadena-Iiguez, 2005: Cadena-Iiguez et al., 2005). Los frutos
se usaron cuando alcanzaron su madurez hortcola a los 182
das despus de antesis (Aung et al., 1996; Cadena-Iiguez et
al., 2007b), cortados en trozos pequeos, secados por tres das
a 50 C, hasta 10 % de humedad y macerados para estandarizar
a tamao de partcula de 2mm (malla No. 10). Posteriormente,
se realiz una extraccin discontinua al sumergir el macerado
en metanol (99.8 %, ACS certificado, Merck) por 48 horas a
temperatura ambiente (20 C2). El extracto alcohlico se filtr
con papel en 12 ocasiones, renovando el disolvente, hasta que
el macerado no produjo filtrados con color; posteriormente,
se evapor el disolvente a presin reducida (Rotavapor Bchi,
R-114, Suiza) a 45 C, hasta obtener un extracto crudo sin
disolvente orgnico (Kuklinski, 2000), el cual fue evaluado en un
primer bioensayo. Posteriormente, a partir de 20 g de extracto, se
realiz el fraccionamiento por cromatografa en columna (CC),
empacada con gel de slice 60 (Merck, D. F. Mxico), con fase
mvil de hexano-acetato de etilo (10:0, 9:1, 8:2, 7:3, 1:1) y acetato
de etilo-metanol (10:0, 9:1, 8:2). Se recolectaron fracciones de
100 mL cada una; stas se monitorearon por cromatografa en
capa fina, empleando como adsorbente gel de slice (60 GF254
,
Merck, Mxico) con fase mvil acetato de etilo-metanol (8:2).Las cromatoplacas se observaron con luz UV a 254 y 365 nm
(Cole-Parmer 9818, Chicago) y se revelaron con un agente
cromognico de anisaldehdo-cido actico glacial-metanol-cido
sulfrico (0.05, 1.0, 8.5 y 0.5 mL, respectivamente) calentando
a 110 C por 5 min, lo que permiti reunir las fracciones por
su identidad cromatogrfica. La seleccin de las fracciones
para la segunda fase de ensayos biolgicos, se realiz con base
en la coloracin de las marcas rojo violeta y caf marrn de la
cromatoplaca relacionadas con compuestos triterpnicos (Che et
al., 1985; Afifi et al., 1999).
Evaluacin de la actividad antiproliferativa
La evaluacin de la accin antineoplsica se realiz a travs
de la medicin de la actividad antiproliferativa y el ndice de
citotoxicidad. La primera se define biolgicamente, como la
reduccin en nmero de la multiplicacin celular, mientras que
la citotoxicidad se refiere a la cualidad de ser txico a clulas
provocando su destruccin (National Cancer Institute, 2009).
Los bioensayos se realizaron empleando las lneas celulares
tumorales L-929 (fibrosarcoma de pulmn de ratn) y HeLa
(cncer crvico-uterino humano). Las lneas se mantuvieron

thin-layer chromatography, employing silica gel as absorben

, Merck, Mexico) with ethyl acetate-methanol (8:2
as mobile phase. Chromatoplateswere observed under UV ligh
at 254 and 365 nm (Cole-Parmer 9818, Chicago) and reveale
using a chromogenic agent of acetic anisaldehyde-acetic acid
methanol-sulfuric acid (0.05, 1.0, 8.5, and 0.5 mL, respectively
heating to 110 C for 5 min, which allowed joining the fraction
by their chromatographic identity. e selection of fractions fo
the second phase of bioassays was made, based on the coloring o
violet red and coffee brown marks of the chromatoplate related
to triterpenoid compounds (Che et al.,1985; Afifi et al.,1999)
Assessment of Antiproliferative Activity
Evaluation of antineoplasic action was carried out througmeasuring of antiproliferative activity and the cytotoxicity index
e former is biologically defined as reduction in number o
cell multiplication, whereas cytotoxicity refers to the qualit
of being toxic for cells provoking their destruction (Nationa
Cancer Institute, 2009). Bioassays were conducted employin
the tumoral cell lines L-929 (rat lung fibrosarcome) and HeL
(human uterine cervical cancer). e lines were kept as crop
in monolayerin IMDM (Iscoves Modified Dulbecos Medium
Invitrogen, U.S.A.) supplemented with 10 % deactivated SFB
(bovine fetal serum) (Gibco BRL, U.S.A.) at 37 C at 5 % CO
atmosphere and 95 % humidity in incubator (scientific erm
Form, Marietta, U.S.A.), at density of 1106
cells mL-1
. alcoholic extract, as well as the fractions (71.2 mg of each) wer
solubilized in 40 mL absolute ethanol and 960 mL phosphat
buffer solution (PBS: sodium chloride (8 g), potassium chlorid
(0.2 g), monobasic potassium phosphate (0.2 g), dibasic sodium
phosphate (2.16 g), 7.2 7.4 pH (SIGMA, CHEM, St. Louis
U.S.A.). e mixture was centrifuged at 500 g during 5 mi
(Hermle-Labortechnik, Germany). Supernatants were filtered i
order to obtain a stock of 14.24 mg per every 600 mL of PBS
this was diluted to concentrations of 0, 0.047, 0.23, 0.47, 1.18

como cultivos en monocapa, en IMDM (Iscoves Modificed
Dulbecos Mdium, Invitrogen, USA) suplementado con 10 %de SFB (suero fetal bovino) desactivado (Gibco BRL, USA), a
37 C en una atmsfera al 5 % de CO2y 95 % de humedad
en incubadora (ermo Forma cientfica, Marietta, USA), a una
densidad de 1106 clulasmL-1. Tanto el extracto alcohlico
como las fracciones (71.2 mg de cada una) se solubilizaron en 40
mL de etanol absoluto y 960mL de solucin amortiguadora de
fosfatos (PBS: cloruro de sodio (8 g), cloruro de potasio (0.2 g), fosfato de
potasio monobsico (0.2 g), fosfato de sodio dibsico (2.16 g), pH 7.2-
7.4 (SIGMA, CHEM, St. Louis, USA). La mezcla se centrifug
a 500 g durante 5 min (Hermle-Labortechnik, Alemania). Los
sobrenadantes se filtraron para obtener un stock de 14.24 mg
por cada 600mL de PBS; ste se diluy a concentraciones de 0,
0.047, 0.23, 0.47, 1.18, 2.37 mgmL-1. Las lneas L-929 y HeLa
se resembraron en placas de 96 pozos a una densidad de 2104
clulasmL-1de medio de cultivo, adicionando el extracto o las
fracciones en las concentraciones indicadas y se co-cultivaron por
72 h. Posteriormente, las clulas adheridas al pozo se fijaron con
glutaraldehido al 1 % durante una hora, despus se adicion la
solucin de cristal violeta (Sigma, St. Louis Missouri, USA) para
colorear el ncleo celular (Kueng et al., 1989), como mtodo
indirecto para cuantificar el nmero celular (Gillies et al., 1986);
las placas se agitaron por 10 min (Labnet International Inc.
Woodbridge, USA) y se retir el exceso de colorante con agua
destilada. En seguida se agregaron 50mLpozo-1de cido actico
al 10 %, agitndolo durante 20 min para solubilizar el colorante.La densidad ptica de las clulas fue evaluada a 570 nm en un
espectrofotmetro de placas (Spectra Tecan Image, Austria). Los
bioensayos se realizaron en dos ocasiones, con tres repeticiones.
Evaluacin de la citotoxicidad
Los sobrenadantes de los ensayos de proliferacin celular
se emplearon para evaluar la presencia de la enzima lactato
deshidrogenasa (LDH), encontrada en el citoplasma celular
(Tejin et al., 2006). La enzima LDH es empleada para
cuantificar el dao a la membrana celular mediante la liberacin
del lactato de deshidrogensa en el sobrenadante (Fischer etal., 2002). La actividad de LDH es medido por una prueba
enzimtica para lo cual, primero el NAD+es reducido a NADH/
H+ por tanto el LDH catalizado es convertido a lactato de
piruvato. En la segunda etapa de catlisis se transfieren H/H+
y se convierte en sal de tetrazolio que es reducida a formazan
que puede ser identificada colorimtricamente (Abe y Matsuki
et al., 2000). Los sobrenadantes se co-cultivaron a temperatura
ambiente (222 C) por 30 min con el kit de deteccin de
LDH (RocheMR, Mxico). Para determinar el porcentaje de
citotoxicidad se requiere de un control positivo el cual permite

Cytotoxicity Assessment
Supernatants of cell proliferation assays were utilized t
evaluate the presence of lactate dehydrogenase enzyme (LDH
found in cell cytoplasm (Tejin et al.,2006). e LDH enzym
is utilized to quantify damage to cell membrane by release o
dehydrogenase lactate in the supernatant (Fischer et al.,2002
LDH activity is measured by enzymatic test for which first NAD
is reduced to NADH/H+, therefore the catalyzed LDH is turne
to pyruvate lactate.In the second phase of catalysis, H/H+ ar
transferred and turned into tetrazolium salt, which is reduceto formazan which may be identified colorimetrically (Ab
and Matsuki et al., 2000). e supernatants were co-cultivate
at environmental temperature (222 C) for 30 min with th

cytotoxicity percentage a positive control is required, whic
allows obtaining absolute cell toxicity; to this purpose, 30 mi
before concluding culturingtime, a Triton X-100 solution at 2 %
(Sigma,USA) with culture medium, in relation 1:1, was added
the culturing time having passed, cytotoxicity was assessed, usin
the Cytotoxicity Detection (LHD) kit (Roche, Diagnostics
Germany).e tests were read at optical density of 490 nm on
a plate reader(Image, Tecan-Spectra, Austria). ree controls peassay were established: one in depth with 200 mL well-1of 10 %
SFB medium, the negative one with 100m L medium in 100m
of cells; and the positive one with 100 mL of cells and 100m
of Triton X-100 (Sigma, USA) at 2 % culture medium. In orde
to determine cytotoxicity percentage absorbance readings wer
utilized; the difference between the value of negative contro
and the value obtained experimentally was divided between
the difference of positive and negative control. e resultin

obtener la toxicidad celular absoluta; para ello, 30 min antes de
concluir el tiempo de cultivo se adicion una solucin de TritnX-100 al 2 % (Sigma, USA) con medio de cultivo, en relacin
1:1; transcurrido el tiempo de cultivo se evalu la citotoxicidad
con el kit Cytotoxicity Detection (LDH) (Roche, Diagnostics,
Germany). Las muestras fueron ledas a una densidad ptica de
490 nm en un lector de placas (Image, TECAN-SPECTRA,
Austria). Por cada ensayo se establecieron tres controles: el
de fondo con 200 mLpozo-1 de medio al 10 % de SFB;
el negativo con 100 mL de medio en 100 mL de clulas; y
el positivo con 100 mL de clulas y 100 mL de Triton X-100
(Sigma, USA) al 2 % de medio de cultivo. Para determinar
el porcentaje de citotoxicidad, se emplearon las lecturas de
absorbancia; la diferencia entre el valor del control negativo y
el valor experimental obtenido se dividi entre la diferencia del
control positivo y el negativo. El cociente resultante se multiplic
por 100. Para eliminar el efecto de la absorbancia del medio de
cultivo, se rest el valor promedio del control de fondo a los
valores experimentales, el control positivo y el negativo (Mora-
Garca et al., 2006).
Identificacin de las fracciones
activas y anlisis estadstico
Las fracciones activas fueron sometidas al anlisis de
resonancia magntica nuclear de hidrgeno (1H NMR) a 200
MHz y a cromatografa de gases acoplada a espectrometra demasas (CG-MS) usando un espectrmetro JMS-AX 505HA
(JEOL) acoplado al cromatgrafo GC Hewlett Packard 5890
series II, equipado con una columna capilar (25 m 20 mm i.
d.0.33mm de espesor), con fenil-metil polisiloxano como fase
estacionaria. La columna se mantuvo a 100 C por un min y luego
se calent a 260 C a 10 C por min. La temperatura del inyector
fue de 250 C y se oper en modo Split en una relacin 70:1. El
anlisis de las masas separadas se hizo por impacto electrnico
de 69.9 eV con la fuente de ionizacin a 230 C. El anlisis
estadstico fue realizado considerando los valores de absorbancia
(570 nm) obtenidos en los ensayos de proliferacin, como forma
indirecta de cuantificar el nmero celular de cada cultivo. Dichosvalores fueron sometidos a un anlisis de varianza, con un nivel
de significancia de a0.05,a0.01 y prueba de Tukey. Se emple
el software SAS, versin 9.0 (SAS Institute Inc., USA), teniendo
como variables al extracto, las fracciones y las concentraciones.
RESULTADOSYDISCUSIN
La primera fase de bioensayos se realiz conel extracto crudo y se observ que las actividadesantiproliferativa y citotxica fueron de tipo dosisdependiente. La inhibicin de la proliferacin

Identification of Active Fractions and Statistical Analysis

RESULTSANDDISCUSSION
e first phase of bioassays was carried out usinraw extract, and it was observed that antiproliferativand cytotoxic activities were of dose-dependentype. Inhibition of cell proliferation was highein L-929 than in HeLa starting from intermediatdoses, recording inhibiting mean concentration(IC
50) of 1.54 and 1.82 mg mL-1, respectively
in other words, the amount of extract (inhibitinagent), needed to inhibit the biological process ocell proliferation to 50 %. Related to cytotoxic meanconcentration (CC
50) or effective maximum mean
concentration (EC50
), values of 2.22 and 2.68 mg
mL-1
for L-929 and HeLa, respectively (Figur1), and in both cases, dose-response curves werrecorded. It has been demonstrated that raw extractof species like Syzygium samarangense (Blume) andPolyalthia longifolia (Sonn.) w., are cytotoxic oncancer lines of human colon (SW-480 and SW-620with IC

dosis intermedias, registrando una concentracininhibitoria media (IC50

Aun cuando las concentracionesevaluadas del extracto crudo de S. edule var.nigrumspinosumfueron mayores a las referidas, los resultadospermiten establecer en principio la potencialidaddel extracto crudo como agente antineoplsico. A
este respecto, Wu et al. (1998) demostraron que laprotena sechiumina de S. edule, inhibi la sntesisprotica en clulas HeLa y concluyeron que sta

concentrations of S. edulevar. nigrum spinosumwerhigher than those referred to, the results in principlallow establishing the potential of raw extract aantineoplasic agent. Regarding this, Wu et al.(1998proved that sechiumine protein of S. eduleinhibitedproteinaceous synthesis in HeLa cells and concludethat it may be used in the preparation ofimmunotoxinwith chemotherapeutic potential. Cadena-Iiguez eal.(2007b) reported antiproliferative activity of rawextracts of eight fruits of S. edulevarieties in P-38and L-929. erefore, it is expected that S. edulvar. nigrum spinosum alcoholic extract may containmetabolites, able to impede the carcinogenesiprocess, fractioning of raw extract being needed.
Based on CC50
, four main fractions were obtainedused to direct the second phase of bioassays. e firsfraction 5-7 (215 mg) was of liquid oily consistenceyellow colored, obtained from hexane-ethyl acetatsystem (9:1). e second fraction 18-20 (35.4 mgwas sticky, of coffee-brown color, from ethyl hexaneethyl acetate system (8:2); fraction 35 (18 mg) osticky consistence showed light yellow color, waobtained from ethyl acetate-methanol (9:1), andfraction 39 (24 mg) was from ethyl acetate -methanosystem (8.2). e bioassay results of antiproliferativ

puede emplearse en la preparacin de inmunotoxinas
con potencial quimioteraputico. Cadena-Iiguez etal. (2007b) reportaron la actividad antiproliferativade extractos crudos de frutos de ocho variedadesde S. edule en P-388 y L-929. Por ello, se esperaque el extracto alcohlico de S. edule var. nigrumspinosum, contenga metabolitos capaces de inhibirel proceso de carcinognesis, haciendo necesario elfraccionamiento del extracto crudo.
Con base en la CC50, se obtuvieron cuatrofracciones principales con las cuales se dirigi lasegunda fase de ensayos biolgicos. La primerafraccin 5-7 (215 mg), fue de consistencia lquidaoleosa color amarillo obtenida en sistema hexano-acetato de etilo (9:1). La segunda fraccin 18-20(35.4 mg) de consistencia pegajosa, color caf marrna partir del sistema hexano-acetato de etilo (8:2), lafraccin 35 (18 mg), de consistencia pegajosa, coloramarillo claro obtenida con el sistema acetato deetilo-metanol (9:1), y la fraccin 39 (24 mg) delsistema acetato de etilo-metanol (8:2). Los resultadosde los bioensayos de la actividad antiproliferativa,mostraron un efecto inhibitorio de tipo dosisdependiente para las fracciones 5-7, 18-20 y 39, conlas cuales se sigui el estudio biodirigido. La fraccin35 no mostr actividad alguna y fue eliminada. La
fraccin 5-7 tuvo el mayor efecto antiproliferativoen ambas lneas, desde dosis intermedias (0.23y 0.47 mgmL-1) para L-929 y desde dosis bajas(0.047 mgmL-1) para HeLa, lo cual permite sugerirespecificidad sobre esta ltima. Respecto a la fraccin18-20, el mayor efecto se dio en la concentracin msalta (2.37 mgmL-1) sobre L-929 y en HeLa en dosisintermedias (0.23, 0.47 y 1.18 mgmL-1) (Figura 2).
Los valores medios de esta fraccin fueronsignificativamente diferentes en HeLa, lo quepermite sugerir mayor especificidad para la misma,segn la dosis. La fraccin 39 mostr efecto
significativo en la dosis ms alta (2.37 mgmL-
1) enambas lneas. El Cuadro 1muestra la comparacinde medias de los valores de absorbancia, indicandodiferencias significativas entre las concentraciones;sin embargo, los valores ms altos en todos los casoscorrespondieron biolgicamente a los valores demenor actividad antiproliferativa, ya que a menorabsorbancia del sobrenadante de clulas tumorales,menor proliferacin de stas.
El anlisis de varianza (ANOVA), mostrdiferencia significativa a una a=0.05 y altamentesignificativa con a=0.01 para las fuentes de

Fraction 39 showed considerableffect in the highest dose (2.37 mg mL-1) on bothlines. e Table 1 presents comparison of means oabsorbency values, indicating significant differenceamong the concentrations; the highest values in althe cases, however, corresponded, biologically, to thvalues of lower antiproliferative activity, since thlower the absorbency of supernatant of tumoral cellsthe lower their proliferation.
e variance analysis (ANOVA) showed significandifference toa=0.05 and highly significant differencwith respect toa=0.01 for sources of variation (linefraction and concentration) as well as for interactionline-fraction and fraction-concentration, from whicit can be deduced that the response of antiproliferativ
activity on tumoral lines is strongly affected by thfraction and its concentration.
Since fraction 5-7 was the one of highesantiproliferative effect on both lines, the studywas directed to the structural elucidation of itcomponents. e 1H MNR spectrum demonstratedthat this fraction refers to a mixture of compoundwhose structural base is the aliphatic chainSubsequent chromatographic fractioning permittedisolation and identification of the principacomponents, which were: methyl ester of 9-oxononanoic acid, methyl ester of hexadecanoic acid
ethyl ester of hexadecanoic acid, methyl ester o10-octadecenoic acid, methyl ester of octadecanoiacid, and octyl ester of octadecenoic acid (Table 2)At separating these compounds from the mixturby CG (Figure 3), these structures were confirmedthrough fragmentation pattern analysis of theirmasspectra.

Dado que la fraccin 5-7 fue la de mayor efecto
antiproliferativo en ambas lneas, el estudio se dirigia la elucidacin estructural de sus componentes.El espectro 1H MNR mostr que dicha fraccinest referida a una mezcla de compuestos cuya baseestructural es la cadena aliftica. El fraccionamientocromatogrfico posterior permiti el aislamientoe identificacin de los componentes mayoritarios,los cuales fueron: esteres del tipo metlico del cido9-oxo-nonanoico, metlico del cido hexadecanoico,etlico del cido hexadecanoco, metlico del cido10-octadecenoico, metlico del cido octadecanoicoy octlico del cido octadecenoico (Cuadro 2).Estos compuestos al separarse de la mezcla por CG(Figura 3), por medio del anlisis del patrn defragmentacin de sus espectros de masas confirmarondichas estructuras.
Los constituyentes de la fraccin 5-7correspondieron a esteres de cidos grasos; se sabeque altas concentraciones de estos metabolitoscausan muerte celular por apoptosis, y cuando lasconcentraciones se incrementan, la muerte celularocurre por necrosis debido a inhibicin de sntesisde ADN (Kharroubi et al., 2004; Benzaria et al.,2007; Takeara et al., 2007). fatty acids, mainly unsaturated ones, are toxic fovarious human tumoral cell lines, such as prostatcancer (Pandalai et al., 1996), breast and bladde(Davies et al., 1999), lung cancer (Schonberg andSkorpen, 1997), and leukemia (Siddiqui et al., 2001Lima et al.,2002; Pompeia et al.,2002). Related tsaturate fatty acids, toxicity for tumoral cells has beenreported (Ostrander et a.,2001; Mu et al., 2001; DSousa Andrade et al., 2005). Ohashi et al. (2003demonstrated that C18 fatty acids at 10 mg mL-
concentration inhibited cell proliferation by 99.4 %of rat lung cancer cell lines. Recently, Yoo et al.(2007recorded that palmitic (Z)-9-octadecenoic andoctadecenoic fatty acids have apoptotic propertiein colon tumoral lines, due to damage in DNA andcaspasa-3 activation.
Conventionally, antineoplasic activity of mancucurbitaceae has been attributed to the presence ocucurbitacins (Dantas et al.,2006; Cadena-Iiguez eal.,2007b; Li et al., 2007; Chen et al.,2008; Wanet al., 2008). autores han comprobado que algunos cidos grasos,principalmente los insaturados, son txicos paravarias lneas celulares tumorales humanas comocncer de prstata (Pandalai et al., 1996), pecho yvejiga (Davies et al., 1999), pulmn (Schonberg ySkorpen, 1997) y leucemia (Siddiqui et al., 2001;Lima et al., 2002; Pompeia et al., 2002). Conrelacin a los cidos grasos saturados, se ha reportadotoxicidad para clulas tumorales (Ostrander et al.,2001; Mu et al., 2001; De Sousa Andrade et al.,2005). Ohashi et al. (2003) demostraron que loscidos grasos C18 a una concentracin de 10 gmL-1inhibieron la proliferacin celular en un 99.4 % delneas celulares cancergenas de rata. RecientementeYoo et al. (2007), registraron que los cidos grasospalmtico (Z)-9-octadecenoico y octadecenoicotienen propiedades apoptticas en lneas tumoralesde colon debido a dao en el AND y activacin dela caspasa-3.
De manera convencional la actividadantineoplsica de muchas cucurbitceas, ha sidoatribuida a la presencia de cucurbitacinas (Dantas etal., 2006; Cadena-Iiguez et al., 2007b; Li et al., 2007;Chen et al., 2008; Wang et al., 2008). Sin embargo,

importance as high-valley crop within the MiddlAmerican zone of Mxico (Cadena et al., 2001Bancomext, 2004). Chayote being an importanfood resource in rural zones, under backyardconditions, and considering the promising resultdescribed in previous paragraphs, its re-valuationas a functional food with higher benefits than thstrictly nutritious ones, may be suggested. Eventhough in the present study there is no referencto the continuance of antiproliferative activity aftecooking the fruits, which is the common way of theiintake (Lira-Saade, 1996), recent laboratory result(unpublished) indicate that the metabolites involvedin these properties do not lose their antiproliferativand cytotoxic effectiveness in vitro.

con los resultados del presente estudio se demuestra
que tambin los esteres de cidos grasos saturados defrutos de S. edule, tienen dicha actividad; estoamplala gama de fuentes de principios qumicos para supotencial uso como agentes antineoplsicos, loscuales en principio permiten sugerir nuevas lneas deinvestigacin. Es relevante apuntar que Mxico es elsegundo exportador mundial de chayote y que nigrumspinosum es la segunda variedad en importanciaeconmica como cultivo en zonas de valles altosdentro de la zona Mesoamericana de Mxico(Cadena et al., 2001; Bancomext, 2004), lo cualaunado a que es un recurso alimentario importanteen reas rurales bajo condiciones de traspatio y alos resultados prometedores descritos en prrafosanteriores, se puede sugerir su revalorizacin comoun alimento funcional con mayores beneficios que elestrictamente nutritivo. Aun cuando en el presentetrabajo no se hace referencia a la permanencia de laactividad antiproliferativa despus de la coccin delos frutos, que es la forma comn de su ingesta (Lira-Saade, 1996), resultados recientes de laboratorio (nopublicados) indican que los metabolitos involucradosen estas propiedades, no pierden su efectividadantiproliferativa y citotxica in vitro.
CONCLUSIONES
El extracto alcohlico de frutos de Sechiumedule var. nigrum spinosum, present actividadantiproliferativa y citotxica de tipo dosisdependiente sobre las lneas celulares L-929 y HeLa,mostrando mayor especificidad para sta ltima endosis altas. De las cuatro fracciones principales delextracto, tres mostraron actividad antiproliferativaen ambas lneas, siendo la 5-7 la de mayor actividady especificidad sobre HeLa desde dosis bajas.Los componentes qumicos de la fraccin 5-7
correspondieron al grupo de esteres de cidos grasossaturados. Estadsticamente, cinco de siete fuentesde variacin involucradas en el presente estudiofueron altamente significativas, atribuible a que larespuesta de la actividad inhibitoria sobre las lneastumorales est fuertemente influida por la fraccinqumica y la concentracin de sta. El contenidode cidos grasos saturados en los frutos comestiblesde S. edule var. nigrum spinosum, ampla la gamade metabolitos y fuentes de especies vegetales conactividad antineoplsica y su revalorizacin como unalimento funcional.
