All users must read the entire Operating Instructions before installing, operating ormaintaining the product.

Always keep the Operating Instructions at hand when operating the product.

Intended useFraction collector F9-C and Fraction collector F9-R are automated fraction collectorsintended for the collection of fractions from purification runs. Fraction collector F9-Cand Fraction collector F9-R are intended for research use only, and shall not be used inclinical procedures, or for diagnostic purposes.

PrerequisitesIn order to operate Fraction collector F9-C and Fraction collector F9-R safely and accord-ing to the intended purpose the following prerequisites must be met:

• You should understand the concepts of liquid chromatography.

• You should be familiar with the purification instrument and system and have readthe Operating Instructions.

• You must read and understand the Safety chapter of these Operating Instructions.

• The system must be installed according to the instructions in Chapter 4 Installation,on page 49.

General precautions

Flammable liquids

CAUTIONExplosion hazard during fractionation of flammable liquids. Donot use a covered fraction collector (Fraction collector F9-C) tofractionate flammable liquids. When using an open fraction collec-tor (Fraction collector F9-R), do not place the fraction collector ina closed compartment and make sure that the room ventilationmeets the local requirements.

CAUTIONFire Hazard. Do not fractionate flammable liquids using Fractioncollector F9-C. When running RPC methods, or other proceduresusing organic solvents, collect fractions through the outlet valveor Fraction collector F9-R.

CAUTIONExplosion hazard if flammable liquid leaks during cleaning ofthe flow path. When cleaning the flow path of Fraction collectorF9-C with a flammable liquid like ethanol, carefully inspect the flowpath, including the waste tubing, to make sure there will be noleakage.

Fraction collector F9-R

CAUTIONFractionation with flammable liquids using Fraction collectorF9-R. Before you start the run, make sure that the tubes in thefraction collector are of correct size according to fraction size set-tings.

CAUTIONMoving tubes with flammable liquids. Move tubes filled withflammable liquids carefully, to prevent spillages that may come incontact with an ignition source.

Personal protection

CAUTIONAlways use appropriate personal protective equipment during op-eration and maintenance of Fraction collector F9-C and Fractioncollector F9-R.

CAUTIONHazardous substances.When using hazardous chemical and bio-logical agents, take all suitable protective measures, such aswearing protective glasses and gloves resistant to the substancesused. Follow local and/or national regulations for safe operation,maintenance and decommissioning of the equipment.

Installing and moving thefraction collector

WARNINGUniNet cable. Only use UniNet cables delivered or approved by GE.

CAUTIONHeavy object. Use proper lifting equipment, or use two or morepersons when moving Fraction collector F9-C. All lifting and movingmust be performed in accordance with local regulations.

NOTICEVents on Fraction collector F9-C. To ensure adequate ventilation,keep papers and other objects away from the vents of the fractioncollector.

Operation

CAUTIONRisk of breaking test vials. Do not use excessive force to pressvials with incorrect dimensions into the fraction collector. Glassvials may break and cause injuries.

CAUTIONAvoid spillage and overflow. Make sure that the equipment isprepared according to the settings in the method to be run.

WARNINGMoving parts. Do not open the door of Fraction collector F9-Cwhile the fraction collector is active. If you need to access thefraction collector, press Pause on the chromatography instrument,and make sure that no parts are moving before opening the door.

CAUTIONMaximum weight on the top of Fraction collector F9-C. Do notplace more than 30 kg on top of Fraction collector F9-C.

Fraction collector F9-R

CAUTIONReplacement of the fraction collector bowl in Fraction collectorF9-R. Do not replace the bowl during a run.

CAUTIONReplacing tubes. Do not replace the tubes in the fraction collectorduring a run.

Maintenance

CAUTIONElectrical shock hazard. All repairs should be done by servicepersonnel authorized by GE Healthcare. Do not open any coversor replace parts unless specifically stated in the user documenta-tion.

CAUTIONDisconnect power. Always switch off power to Fraction collectorF9-C and Fraction collector F9-R, before cleaning any of its compo-nents, unless stated otherwise in the user documentation. This isdone by switching off the chromatographic instrument that pro-vides Fraction collector F9-C and Fraction collector F9-R withpower.

CAUTIONHazardous chemicals andbiological agents.Before maintenance,service and decommissioning, wash Fraction collector F9-C andFraction collector F9-R with a neutral solution to make sure thatany hazardous solvents and biological agents have been flushedout from the fraction collector.

CAUTIONAlways use appropriate personal protective equipment when de-commissioning the equipment.

CAUTIONCleaning Fraction collector F9-C and Fraction collector F9-Rbefore decommissioning.

• Wipe Fraction collector F9-C and Fraction collector F9-R witha damp tissue using a cleaning agent so that no hazardoussolvents or biological agents remain on the surface. Do notforget to clean the interior of Fraction collector F9-C.

• Perform a system CIP using a neutral solution. Make sure thatany hazardous solvents or biological agents are flushed outfrom the fraction collector.

About this sectionThe ÄKTA™ instrument supplies the Fraction collector F9-C and Fraction collector F9-Rwith power. This section describes how to perform an emergency shutdown of theFraction collector F9-C and Fraction collector F9-R by shutting down the ÄKTA instrument.This section also describes the results in the event of power failure or network interruption.

Emergency shutdownIn an emergency situation, stop the run by either pausing the run or switching off theinstrument as described below:

2.4 Recycling information

IntroductionThis section contains information about the decommisioning of Fraction collector F9-Cand Fraction collector F9-R.

DecontaminationThe product must be decontaminated before decommissioning. All local regulationsmust be followed with regard to scrapping of the equipment.

Disposal of the productWhen taking the product out of service, the different materials must be separated andrecycled according to national and local environmental regulations.

CAUTIONAlways use appropriate personal protective equipment when de-commissioning the equipment.

Disposal of electricalcomponents

Waste comprising electrical and electronic equipment must not be disposed of as un-sorted municipal waste and must be collected separately. Please contact an authorizedrepresentative of the manufacturer for information concerning the decommissioning ofequipment.

Thenumber in the symbol is the Environment-friendly Use Period (EFUP), which indicatesthe period during which the hazardous substances contained in electrical and elec-tronic products will not leak or mutate under normal operating conditions so that theuse of such electrical and electronic products will not result in any severe environmen-tal pollution, any bodily injury or damage to any assets. The unit of the period is “Year”.

In order to maintain the declared EFUP, the product shall be operated normally ac-cording to the instructions and environmental conditions as defined in the productmanual, and periodic maintenance schedules specified in Product Maintenance Pro-cedures shall be followed strictly.

Consumables or certain parts may have their own label with an EFUP value less thanthe product. Periodic replacement of those consumables or parts to maintain thedeclared EFUP shall be done in accordance with the Product Maintenance Procedures.

This product must not be disposed of as unsorted municipal waste, and must becollected separately and handled properly after decommissioning.

3.1.3 Cassettes, Cassette tray and racks

IntroductionFractions can be collected in deep well plates and in tubes of different sizes. A numberof cassettes, trays and racks for different tubes and deep well plates are available. Thecassettes are placed on a tray with six cassette positions.

The cassette type codes, tray type code and rack type codes are scanned by the typecode reader to determine the type of cassettes, trays and racks used. For more informa-tion on requirements that need to be fulfilled by the tubes and deep well plates to beused in Fraction collector F9-C, see the ÄKTA pure User manual.

Available cassettes, trays andracks

The following Cassettes and racks are available:

• Cassette 3 ml tubes (for 40 tubes)

• Cassette 5 ml tubes (for 40 tubes)

• Cassette 8 ml tubes (for 24 tubes)

• Cassette 15 ml tubes (for 15 tubes)

• Cassette 50 ml tubes (for 6 tubes)

Software configurationWhen the fraction collector has been installed, the System properties for the systemhas to be updated in UNICORN. The system will restart automatically when the configu-ration has been changed and the system can be reconnected.

From hereon, UNICORN refers to UNICORN 6.3 or other compatible versions of the soft-ware. The examples given in these Operating Instructions refer to UNICORN 6.3.

4.2.2 Connect tubing

IntroductionThis subsection contains information on how to connect tubing from ÄKTA pure toFraction collector F9-C and Fraction collector F9-R.

Fraction collector F9-CFraction collector F9-C is delivered with all internal tubing in place. The tubing betweenthe fraction collector and purification instrument need to be installed.

Follow the instructions in the table below to connect the tubing from the ÄKTA pure in-strument to the fraction collector.

Place the Tubing holder with the tubing over the Tube adjustment cavity (2)of the Delivery arm. Push the tubing down against the bottom of the Tubeadjustment cavity, and then fingertighten the Tubing holder nut. This ensuresthe correct length of the exposed tubing end (6).

4.4 Set the delay volume

IntroductionThe delay volume settings are used to make sure that the fractions collected duringfractionation, using the outlet valve or the fraction collector, correspond to the fractionsindicated in the chromatogram.

Set the delay volumeFollow the instructions below to set the delay volume between the UV monitor and theoutlet valve and between the UV monitor and the fraction collector. Refer to the purifca-tion instrument User manual for more information about delay volumes.

About this sectionThis section describes how to prepare and assemble Fraction collector F9-C before arun.

The fraction collector is connected to ÄKTA pure and controlled by UNICORN. Control ofthe fraction collector can be achieved automatically in a method run, or manually.

CAUTIONFire Hazard. Do not fractionate flammable liquids using Fractioncollector F9-C. When running RPC methods, or other proceduresusing organic solvents, collect fractions through the outlet valveor Fraction collector F9-R.

The tray and racks can tilt slightly when not fully inserted into the fractioncollector and may harm the fractionation arm. The tilt is due to the height dif-ference between the door and the floor of the fraction collector. The tendencyto tilt is affected by the placement andweight of the cassettes, tubes or bottles

Note:

Prepare the fraction collectorBefore starting to prepare Fraction collector F9-C, check the fractionation settings in themethod to be run. Perform the steps described below according to the settings in themethod.

• Insert the Cassette tray or a rack for tubes or bottles.

• Change the System Settings in UNICORN to set the fractionation mode and othersettings for fraction collection.

Prepare and insert the Cassettetray

Follow the instructions below to add cassettes to the Cassette tray and insert the trayinto the fraction collector.

Place the tubes and deep well plates in the cassettes. Make sure that thedeep well plates are rotated so that the well marked A1 is positioned abovethe A1 marking on the cassette.

• position the tray and slide it into the fractionation collector until itreaches the end

• make sure that the Tray catch snaps into closed position, as shown be-low.

Note:• The tray can tilt slightly when not fully inserted into the fraction collector

andmay harm the fractionation arm. The tilt is due to the height differencebetween the door and the floor of the fraction collector. The tendency totilt is affected by the placement and weight of the cassettes.

• A height exclusion bar ensures that the tubes or deep well plates arecorrectly positioned and cannot damage the Dispenser head.

Result: After the door has been closed, the fractionation arm scans the cas-sette type code of each cassette to identify the cassette types. If deep wellplates are used, the instrument also identifies the types of deep well plates.

Note:

If the tray is inserted with the front of the tray facing the wrong way it willnot be possible to close the door.

Prepare and insert a rack fortubes or bottles

Follow the instructions below to insert a rack for tubes or bottles into the fraction collector.

CAUTIONExplosion hazard if flammable liquid leaks during cleaning ofthe flow path. When cleaning the flow path of Fraction collectorF9-C with a flammable liquid like ethanol, carefully inspect the flowpath, including the waste tubing, to make sure there will be noleakage.

6.1 Clean Fraction collector F9-C

Maintenance intervalClean the Fraction collector when required, for example if liquid has been spilled in theFraction collector chamber. The internal tubing of the fraction collector may need to bereplaced for maintenance or for process purposes. Information on how and when toreplace the internal tubing can be found in the ÄKTA pure User manual.

Required materialThe following materials are required:

• Wash bottle

• Water or 20% ethanol

• Cloth

InstructionFollow the instruction below to clean the interior of the Fraction collector. The locationsof the components of the fraction collector are shown in Section 3.1.2 Fraction collectorF9-C illustrations, on page 32.

Perform a fraction collector wash:1

In System Control, select Manual:Execute Manual Instructions:Fractioncollection:Fraction collector wash. Click Execute.

NOTICE

• If no column valve is used, make sure to replaceany columns in the flow path with tubing before afraction collector wash is performed.

• Lower the flow for the fraction collector wash inthe instruction Fraction collector wash settingsif the system back pressure is elevated during thewash.

• Make sure not to exceed the pressure limits for anyof the modules that are part of the flow path.

Open the door of the fraction collector and remove the rack3

Wash the Cassette tray or rack and the Cassettes (if applicable), with waterand a mild cleaning agent.

Lift off the Waste funnel and wash it with water and a mild cleaning agent.5

Refit the Waste funnel.

Wipe off the interior of the fraction collector using a damp cloth. Wipe offstains using a mild cleaning agent or 20% ethanol.

Wipe off the Dispenser head and its diode windows (the Drop sync sensorand the Type code reader) using a wash bottle with water or 20% ethanoland a cloth.

Let the fraction collector dry completely before starting a run.8

Close the door of the fraction collector.9

Result: Automatic scanning is performed.

In the System Control module, press the End icon in the toolbar.10

Result: The Dispenser head moves to home position.
