Glycosylation is one of the most common post-translational modifications of proteins. Almost all proteins are glycosylated. Glycosylation influences changes to folding, targeting, ligand recognition, binding, stability and immunity. The function of the protein is adjusted by glycosylation to suit the respective purpose. Glycans play an important role in cancer, immunology, infectious disease and regenerative medicine. For instance, it is well known that glycoproteins are useful cancer markers. Approximately half of cancer markers utilized in current clinical practice are glycan markers. It is also well known that viruses target glycoproteins on cell membrane receptors.

Glycotechnica’s glycan analysis system is a very useful tool for developing biomarkers, conducting quality control of pharma-ceutical proteins and characterization of stem cells. We aim to improve the future of medical treatment by providing this unique tool to research institutes all over the world.

GlycostationTM2200 is an ultra-high speed and highly sensitive optical microarray

scanner utilizing the printciple of evanescent-field fluorescence excitation

to analyze an entire microarray slide in only two minutes.

LecChipTM is arrayed with 45 different lectins which bind to specific glycans. It is possible to measure up to 7 samples in only one chip.

Major differences between GlycoLiteTM2200 and GlycostationTM2200 are:

Lectin Microarray: LecChip™

Lectins are well known as glycan binding proteins and can be classified into several categories, for instance: fucose-binders, sialic acid-binders, asialo-form-binders, high mannose-binders and O-glycans binders. We developed a slide glass-like lectin microarray on which 45 different lectins are immobilized in triplicate (LecChipTM). These lectins were selected from a number of candidates, based on the results of the hector-by-hecto project performed at Advanced Institute of Industrial Science and Technology (AIST) in Japan.

The basic idea of our technology is to use these lectin to decipher epitope structures of glycans. However, as is well known, the binding between lectins and glycans is weak so that the washing process easily breaks the bond resulting in loss of the specific binding information. In order to overcome this issue, we developed a dedicated scanner using the principle of evanescent-field fluorescence excitation (GlycoStationTM Reader1200/2200, GlycoLiteTM2200).

In this way, our technology makes it possible to detect even the weakest interactions between lectins and glycans in buffer solutions without any washing process. Glycotechnica is pleased to introduce its novel system GlycoStationTM for quick and accurate glycan structure profiling and analyses. This is a Joint development with AIST.

A Short-Cut to Diagnostic and Therapeutic Developments

1. DISCOVERY OF BIOMARKERS IN CANCEROUS TISSUES.

Using very small sections of paraffin-embedded tissue, differential glycan profiling has identified potential biomarkers in colon cancer, bile duct cancer, liver cancer, esophageal cancer, breast cancer and uterine cancer.
