CULTIVO PMECs(Células endoteliales pulmonares microvasculares)
Experimento realizado con el fin de aislar células de un organismo en condiciones especificas que permitan su supervivencia y crecimiento, y donde se tenga control de su desarrollo.
Se utilizaron células endoteliales microvasculares de ratones para poder manipular de manera practica y realizar control de los procedimientos realizados.

FLUORESENCIA
Técnica para localización de proteínas dentro de una célula, y su posterior
observación por microscopía de fluoresencia. Un fluorocromo se define como la sustancia capaz de emitir luz a
cierta longitud de onda especifica.
• Caspasa 3: se usó fluoresencia para estudiar la actividad de la Caspasa 3, iniciadora de la via extrínseca de la apoptosis.
• Calpaina: Se utiliza un sustrato fluorescente que permite marcar la proteasa Calpain.

ELISATécnica usada para estudiar la unión de un anticuerpo frente a un antígeno en la muestra establecida, junto con la enzima que corresponde a la señal de la unión frente al complejo antígeno-anticuerpo.
Se utilizó para reconocer el DNA libre en la muestra causado por la fragmentación, y de esta manera determinar si hubo apoptosis causado
por la sepsis bacteriana.

TRANSFORMACIÓN Procedimiento por el cual las células de un cultivo se mezclan con un vector de DNA recombinante, donde posteriormente la célula incorporará el DNA plasmídico, y se reproducirá con esta variable.
Células endoteliales microvasculares pulmonares fueron infectadas con Adenovirus recombinante donde se observó el proceso de
transformacion con la transferencia del gen.

WESTERN BLOT-IMMUNOBLOT
Proceso para detectar una proteína particular la cual combina la resolución de la electroforesis en gel, especificidad de anticuerpos y sensibilidad de pruebas enzimáticas. Separa e identifica una proteína particular.
Se uso con el fin de observar la expresión de Calpain y p38 MAPK, usando anticuerpos
específicos.

Heat stress, as a physiological way, allows prevent apoptosis.
Calpain are very important to induce an apoptotic process.
The Calpastain, an endogenous inhibitor of Calpain, reduced acute lung injury.
