Initial Bottle Setup Reagent Management Method

We recommend the initial setup shown in the images below. Setup must be validated by the laboratory prior to use, according to local or regional accreditation requirements.

Reagent types screen – Purity thresholds. Reagent types screen – Temperature thresholds.

Configure the bottles as shown. Note: Put 70% and 90% Ethanol in bottles 3 and 4 only for initial setup. When replacing the least pure Ethanol as part of ongoing operation, always replace with 100% Ethanol, even for bottles 3 and 4.

Tissue Size Recommendations – Xylene and Xylene-Free

Protocol Example Maximum Thickness Specimen Type (Example)

1hr 1.5 mm Endoscopies and needle biopsies of breast and prostate.

2hr <3 mm All biopsies up to 3 mm diameter: GI biopsies, renal, prostatic, hepatic and breast cores, punch biopsies of skin, small colonic polyps.

4hr 3 mm Small specimens of non-dense tissues (kidney, liver, bowel etc), excisional and incisional skin biopsies, skin ellipses.

6 – 8hr 15 x 10 x 4 mm All routine tissues up to maximum dimensions (excluding brain specimens).

12hr 20 x 10 x 5 mm All routine tissues up to maximum dimensions. Very thick fatty specimens may require a longer protocol.

Running Protocols

Leica PELORIS – Running a Protocol

Check reagent and wax levels are between “MAX” and “MIN 3”(3 baskets) or “MIN 2” (2 baskets).

Check system status• Ensure retort is clean• Ensure no expired

reagents.

1Conduct Pre-Run Checks

4 View the progress of each step (expanded view shown).

View instrument status.

Monitor the Progress

Protocol complete. Drain the retort then remove the baskets.

Tap Clean now to start a cleaning protocol.

5 Complete the Run

Enter the number of cassettes.

Place the baskets (1, 2 or 3) into the retort.

Running Protocols

Adding Cassettes to a Running Protocol

2 Return the cassette basket to the retort and tap OK on the Retort unlocked dialog.

Enter the total number of cassettes in the retort and tap OK.

When prompted remove the cassette basket from the retort and add the new cassettes.

1Pause the Protocol

Add New Cassettes and Restart the Protocol

On the Paused dialog tap Unlock retort for the appropriate retort.

Tap Yes to drain the retort before adding new cassettes. This prevents reagent overflow.

On the Status screen tap Pause for the retort that you want to add the new cassettes to.

Wait for the Paused dialog to appear and tap Resume.

Leica PELORIS Reagent Top off/Top up

Important! Ensure no protocol is running then remove the bottle from the instrument.

Top off/up the bottle when the reagent is below the minimum level. This is MIN 2 for a two basket setup or MIN 3 for a three basket setup.

The software will indicate that the bottle is missing.

2 Tighten the cap by hand.Fill the bottle to the

maximum level.

Fill the Bottle

Tap the reagent row on the Inserted bottle configuration dialog.

Then tap Done.

3 Update the Software

Return the bottle to the cabinet – push firmly into the rear connectors.

Tap Topped off/Topped up.

Tighten the cap by hand.Fill the bottle to the maximum level.

Return the bottle to the cabinet – push firmly into the rear connectors.

2 Unscrew the lid. Safely dispose of the used reagent.

Remove the bottle from the reagent cabinet.

Or when warned at protocol start (final threshold exceeded).

Replace reagent when bottles appear hatched on the Status screen.

Ensure no protocols are loaded or running.

Select Remote, a retort and the bottles to drain, then tap Drain from bottle(s).

Open the Remote fill/drain screen.

Follow on-screen instructions then wait until notified of completion.

Leica PELORIS Reagent Replacement – Remote Fill and Drain

Connect the remote fill/drain hose.

4 Insert into reagent container.

Ensure empty bottles are securely in position and the caps are not loose.

Monitor the fill progress – wait until notified of completion.

5 Select Remote, a retort, and the empty bottles, then tap Fill to bottle(s).

Follow on-screen instructions then confirm or change new reagent details when prompted

Clean the retort if the residue is incompatible with the next run.

Connect the remote fill/drain hose.

Insert the hose into a stable container with sufficient capacity to avoid overflow.

Ensure a clean retort is available.

*Warning: Never use wax products containing DMSO – they can compromise processing quality.

Select Wax waste and the wax chambers to drain, then tap Drain to waste.

Open the Remote fill/drain screen.

Follow on-screen instructions then wait until notified of completion.

Leica PELORIS – Wax Replacement

Clean the wax bath using a lint-free cloth.

4 Add wax directly to the bath; use ParablocksTM for easy handling and rapid melt.

Or use Paraplast® (molten or pellets). Ensure wax does not go down the air vent.

5 Tap the wax chamber state in the Reagent stations screen (Wax chambers).

Wax concentration and cycle, cassette and day counts are automatically reset.

Set the chamber state to Full (molten wax) or Not molten (Parablocks or pellets).

Connect wax waste line. Insert hose into a stable container with sufficient capacity to avoid overflow.

Ensure wax is molten then close the lid.

Or when warned at protocol start (final threshold ex-ceeded).

Replace wax when chambers appear hatched on the Status screen.

Scrape around the lids and seals on the retorts and wax baths with the supplied plastic scraper. Ensure no material falls into the wax bath air vent.

Clean liquid level sensors (LLS – 1, 2, 3) and air hole. Ensure no material goes into the air hole.

Check the stirrer and clean the retort walls.

Check reagent levels – top up/off if required.

Check wax levels – top up/off if required. Scrape and wipe the top of the Leica PELORIS.

Clean the touch-screen.

Transfer Log Files

Set the date to the last day of the selected period.

For a record of on-going performance, select Event details and a suitable period.

Tap Transfer files to copy files to the USB drive.

When logs have been transferred tap Add/Remove USB device(s) again.

4 When prompted, remove the USB drive.

5 Attach the USB drive to a networked computer.

Locate the files at X:\[instrument serial number] \ [transfer date and time] (X is location of USB drive).

Email incident report zip file (from step 2) or all event details files (step 3) to your local support center.

2 On the File transfer screen tap Transfer files to copy files to the USB drive. Continue at step 4.

For the record of a specific incident, select it on the Event log screen and tap Incident report.

Wait until notified to proceed, then plug the USB drive into the front USB port.

Open the File transfer screen (Control menu) and tap Add/Remove USB device(s).

After the computer registers the drive, tap USB Key.

Tap OK to confirm. Tap Clean now for both retorts to start cleaning protocols.

Drain the retorts when prompted then remove the cassettes. Close the retorts and tap OK.

On the Status screen tap Pause.

On the Paused dialog tap Abandon for the appropriate retort.

On the Protocol abandoned dialog tap OK.

Tap OK to confirm. Tap Clean now to start a cleaning protocol.

Drain the retort when prompted then remove the cassettes. Close the retort and tap OK.

On the Status screen tap Pause.

Tap Emergency stop. Note: this will stop both protocols immediately.

When to shut down:• Instrument is being moved or

incidentAt other times keep the instrument on, even if idle for extended periods. Tap OK to confirm. The

software will now safely shut down.

If conditions permit, also switch the power off at the mains socket and pull out the power cord.

Only use the emergency stop procedure when there is immediate danger or risk to safety. This procedure can damage the instrument and tissue being processed.

Wait for the screen to go blank then press the power button at the front of the instrument.

In an emergency, press the power button at the front of the instrument.

Tap the shutdown button on the Function bar.

Ensure the instrument is idle with no protocols or other operations in progress.

General Warnings and Cautions

Persons operating the Leica PELORIS must be fully aware of the following warnings, in order to mitigate possible tissue damage or loss.

Reagent ConfigurationWARNING

Always ensure that the reagents configured in the software are the actual reagents loaded on the instrument. A station containing different reagent could damage tissue samples.

Replacing ReagentsWARNING

Always change reagents when prompted.

Always update station details correctly – never update the details without replacing the reagent.

Failure to follow these directives can lead to tissue damage or loss.

WARNING

Do not alter the concentration of a used reagent unless you are able to verify the actual concentration. If the concentration is incorrect a reduction in tissue processing quality or damage to the tissue sample may result.

Protocol ValidationWARNING

Do not set new protocols as validated until they have passed the validation procedures for your laboratory. Only then should you edit the protocol to set it as valid, making it available to operators for clinical use (see 4.1.4 Protocol Validation). Use of nonvalidated protocols may result in tissue damage or loss.

Basket and Cassette LoadingWARNING

Always ensure the cassettes are correctly inserted into the baskets and that the baskets are correctly placed in the retorts. Incorrectly placed cassettes or baskets may lead to samples being damaged as some tissue may not be fully covered by reagent during processing (see 2.2.4 Cassette Baskets).

WARNING

Never place three baskets into a retort when the instrument is configured with a two-basket fill level. If this occurs, reagent will not cover the top basket and tissue samples will be damaged.

Cleaning ProtocolWARNING

Do not load unprocessed tissue samples into a retort prior to running a cleaning protocol. Formalin in the residue purged to the wax bath at the start of the cleaning run may damage tissue on subsequent runs.

If you inadvertently load unprocessed samples into a retort prior to running a cleaning protocol, remove the samples and attempt to load a processing protocol before loading the cleaning protocol. The purge before the cleaning run will be skipped.

WARNING

Remove all tissue from the retort before running a cleaning protocol as the dry step will damage the tissue.

WARNING

Do not use cleaning protocols for reprocessing as the dry step will damage the tissue.
