National Brain Tumor Society’s Feldman Founder’s Award

The National Brain Tumor Society 2013 Feldman Founder’s Award is presented to Dr. David Louis, for his outstanding contributions and invaluable impact on the field of brain tumor research.

Dr. Louis is Pathologist-in-Chief at Massachusetts General Hospital and Benjamin Castleman Professor of Pathology at Harvard Medical School. His work in the molecular genetic basis of gliomas has been fundamental in the advancement of brain tumor research. Dr. Louis’s laboratory was the first to demonstrate that molecular approaches could subdivide malignant gliomas biologically and could predict their response to therapies. This work has resulted in worldwide adoption of molecular testing for brain tumor patient management.

The Feldman Founder’s Award is a new merger of two separate and distinct honors that the National Brain Tumor Society has traditionally bestowed on the research community. The original National Brain Tumor Society Founder’s Award for Research Excellence was created in 2008 as part of our merger to recognize exceptional researchers or teams whose research holds the promise for clinical improvements. The Seth Harris Feldman Chair of Research supported a grant given each year to a researcher or team whose project has the potential to significantly advance the understanding of the field. This chair was awarded in remembrance of Seth Feldman, in honor of his courageous battle with glioblastoma multiforme.

Today’s Feldman Founder’s Award recognizes individuals or groups for a transcendent scientific discovery, an exceptional body of work, or leadership that had a significant and sustained impact on the brain tumor community consistent with National Brain Tumor Society’s mission to find better treatments, and ultimately a cure, for people living with a brain tumor. The recipient will receive a $50,000 award and will join the prestigious group of past National Brain Tumor Society Founder’s Award for Research Excellence winners including co-recipients, Dr. Anna Barker, then Deputy Director of the National Cancer Institute and Dr. Kenneth Kinzler, Professor of Oncology at The Johns Hopkins Kimmel Cancer Center, for their work on The Cancer Genome Atlas (TCGA) and CEO of American Association for Cancer Research, Dr. Marge Foti, PhD, MD (h.c.) for her years of leadership in the field, particularly in systems biology.

National Brain Tumor Society Programmatic Overview

Our strategic initiatives and funded programs aim to improve the understanding of brain tumors to transform research findings into new and more effective treatments. To achieve these goals, we foster collaboration and change within all facets of the drug discovery and development landscape and invest wisely in key initiatives poised to deliver results.

• Defeat GBM Research Collaborative – Our largest and boldest initiative to date, this strategic research initiative aims to double the five-year survival rate of glioblastoma multiforme (GBM) patients in just five years through an innovative funding model and novel scientific construct.

• Clinical Trial Endpoints Initiative – This initiative works with the US Food and Drug Administration (FDA) to recommend alternative (and additional) clinical trial endpoints to accelerate the pace of development for drugs which will provide clinical benefit(s) to brain tumor patients.

• Pediatric Research Grant Initiative – This program analyzes the development of a child’s brain to identify therapies which could be effective and minimize long-term side effects for pediatric brain tumor patients. See related grants on page 11.

• Oligodendroglioma Research Fund – This program aims to better understand the origins of growth for oligodendrogliomas, and apply these findings to the development of new treatments. See related grants on page 12.

• Advancing Research to Treatments (ART) – The goal of this program is to translate laboratory science into commercialized brain tumor treatments by bringing together researchers, biotechnology and pharmaceutical executives, venture capital (VC) investors, and brain tumor patients to address the challenges in drug development.

• Grand Challenge - A crowd-sourcing effort to predict the specific drugs and drug combinations that elicit the strongest therapeutic response in in-vivo drug testing of high grade gliomas, using systems biology methodologies.

In addition to the programs above, we continue to foster collaboration by hosting think tanks and conferences covering critical brain tumor topics, participating in key meetings taking place in the community, and mobilizing volunteer advocates across the country.

2013 National Brain Tumor Society Grant Recipients

The National Brain Tumor Society is proud to recognize our 2013 research grant recipients. Through our innovative funding initiatives created to maximize impact to patients, and rigorous and thoughtful grant process, we fund only the most imminently promising projects, funding researchers dedicated to approaches that will deliver compelling results.

PEDIATRIC RESEARCH

These three grants are awarded over a period of three years, beginning in 2011.

Robert Wechsler-Reya, PhDSanford-Burnham Medical Research InstituteGenetic targeting of cerebellar stem cells to study development and tumorigenesis

The most aggressive subtype of medulloblastoma occurs in tumors that have in-creased expression of the MYC oncogene. In this research, the Wechsler-Reya labo-ratory will build on previous research that suggests that stem cells can give rise to MYC-driven medulloblastoma. The researchers will create and study mouse models that allow targeting of genes to stem cells and then follow the course of normal stem cells and generate models of MYC-driven medulloblastoma. The goal is to discover potential new treatments based on an understanding of the tumor cells of origin.

Alexandra Joyner, PhDMemorial Sloan-Kettering Cancer CenterIdentification of genes and cell behaviors regulated by Shh/Gli2 signaling in the cell of origin of a major subtype of medulloblastoma using novel genetic tools in mouse

Medulloblastoma, the most common brain tumor in children, occurs in multiple molecular subtypes. A process called hedgehog signaling (named after the hedgehog gene Hh) is elevated in tumor subtypes that arise from cerebellar granule cell pro-genitors (GCPs). In this research, the Joyner laboratory will investigate how altering the level of signaling in GCPs in genetically engineered mice alter GCP behaviors. The goal is to discover novel signaling pathways that are important for normal cerebellum development.

Joseph Scafidi, DOChildren’s Research InstituteThe effects of molecularly targeted therapies on the developing neurogenic niches

Although recent treatment compounds have shown promise in decreasing the burden of pediatric brain tumors, the compounds also affect the developmental pathways of

a normal developing brain, and long-term effects are unknown. In this study, the Scafidi laboratory will assess the cellular and functional effects of compounds in rodents at various developmental ages, and whether any rehabilitation efforts after treatment can induce brain plasticity and address the deficits associated with treatment.

INNOVATION GRANT - PEDIATRIC

This grant was awarded in 2012 for a period of two years.

David H. Gutmann, MD, PhDWashington University School of MedicineKIAA1549:BRAF genetically-engineered mice for pediatric glioma-targeted therapeutic drug discovery and evaluation

One of the most common brain tumors in children is the pilocytic astrocytoma. This tumor is often caused by either genomic rearrangement of the BRAF gene (sporadic) or the inactivation of the NF1 tumor suppressor gene (inherited) that, when mutated, causes neurofibromatosis type 1. For this research project, the Gutmann laboratory will conduct experiments using advanced systems biology bioinformatics to identify potential biological treatment targets for both sporadic and inherited pilocytic astrocytoma.

OLIGODENDROGLIOMA RESEARCH FUND

These grants were awarded in 2013 for a period of two years.

David Louis, MDMassachusetts General Hospital (MGH) Mapping the Epigenomic Landscape of Oligodendroglioma to Elucidate Tumor Growth and Chemosensitivity

Dr. Louis and his team will seek to identify the elements that drive the growth of oligodendrogliomas by applying next-generation sequencing tools and experiments. Dr. Louis’ work is designed to lead to discoveries of specific molecular targets, which subsequent studies would examine for potential therapeutic approaches.

Anders Persson, PhDUniversity of California, San Francisco IDH1R132H-induced Transformation of Oligodendrocyte Progenitor Cells into Oligodendroglioma

Dr. Persson and his team will seek to create predictive disease models (in mice/human/xenograft tissue) to prove that a specific gene mutation of the common enzyme IDH1 creates a molecule, called oncometabolite 2-HG, that facilitates the transformation of normal oligodendrocyte cells (OPC), a common cell in the human brain, into malignant, oligodendroglioma cells. Dr. Persson’s team will work to validate that these OPCs are the “cell of origin”, and know what mutations drive them to develop into an oligo tumor, in order to develop targeted therapies to attack their pathways.

The Oligodendroglioma Research Fund is part of our Community Research Fund program – an initiative allowing contributors to pool donations to support research on a specific type of tumor. Donations large and small can be earmarked and combined to fund a two-year grant of $300,000, the minimum grant amount necessary to conduct significant research studies.
