LIQUID CRISTALS

• Technology capable to promote high stability to the formulations,high skin feeling and boost up penetration of active compounds;

• Liquid cristals overcome the concept of classic emulsion in which awater phase and an oily one are mixed together;

• Liquid cristals are “small islands” in which water and oil mixtogether thanks to particular emulsifiers: as like in a cream boxthere are milions of microscopic droplets of active principles;

CENTELLA ASIATICA(vegetal stem cells)

• Glucosides extracted from this plant show a vigorous action in the inhibiting ofjaluronidase, enzyme involved in jaluronic acid destruction;

• Association with Glucosamine is particularly inteersting because represents thebrick of jaluronic acid and promote its biosynthesys; it is possible to reducephysiologic jaluronic acid degradation and so inducing an “endo-filler” effect;

• Centella asiatica is widely employed in dermatology as epithelia restorationagent, scar promoting and blood vessels protector;

• An innovative biotechnological technique is capable to isolate Centella asiaticastem cells highly enriched in active molecules. This technique allows to maintainplant phyto-complex with an high active principles content (up to 50% ofglucosides on the plant weight) (Patent, IRB);

• Both glucosamine and its derivative N-acetyl glucosamine are amino-monosaccharides thatserve key biochemical functions on their own and as substrate precursors for the biosynthesisof polymers such as glycosaminoglycans (e.g., hyaluronic acid) and for the production ofproteoglycans. Glucosamine has an excellent safety profile and has been shown to providebenefits in several clinical disorders. Glucosamine compounds have been reported to haveseveral beneficial effects on the skin or skin cells. Because of its stimulation of hyaluronic acidsynthesis, glucosamine has been shown to accelerate wound healing, improve skin hydration, anddecrease wrinkles. In addition, as an inhibitor of tyrosinase activation, it inhibits melaninproduction and is useful in treatment of disorders of hyperpigmentation. Mechanistically,glucosamine also has both anti-inflammatory and chondroprotective effects. Clinical trials haveshown benefit in using oral glucosamine supplementation to improve symptoms and slow theprogression of osteoarthritis in humans. Glucosamine has also been used to prevent andtreat osteoarthritis in animals. Based on other observations, glucosamine has been suggestedfor additional clinical uses, including treatment of inflammatory bowel disease, migraineheadaches, and viral infections. The current clinical uses for topical and oral glucosaminecompounds and the mechanistic rationale for these uses are reviewed here.

Epub 2009 Aug 28.Reduction in the appearance of facial hyperpigmentation after use of moisturizers with a combination oftopical niacinamide and N-acetyl glucosamine: results of a randomized, double-blind, vehicle-controlledtrial.Kimball AB, Kaczvinsky JR, Li J, Robinson LR, Matts PJ, Berge CA, Miyamoto K, Bissett DL.Harvard Medical School, Boston, MA 02114, USA. [email protected]: Topical niacinamide and N-acetyl glucosamine (NAG) each individually inhibit epidermalpigmentation in cell culture. In small clinical studies, niacinamide-containing and NAG-containing formulationsreduced the appearance of hyperpigmentation. OBJECTIVES: To assess the effect of a combination ofniacinamide and NAG in a topical moisturizing formulation on irregular facial pigmentation, including specificdetection of changes in colour features associated with melanin. METHODS: This was a 10-week, double-blind,vehicle-controlled, full-face, parallel-group clinical study conducted in women aged 40-60 years. After a 2-week washout period, subjects used a daily regimen of either a morning sun protection factor (SPF) 15sunscreen moisturizing lotion and evening moisturizing cream each containing 4% niacinamide + 2% NAG (testformulation; n = 101) or the SPF 15 lotion and cream vehicles (vehicle control; n = 101). Product-inducedchanges in apparent pigmentation were assessed by capturing digital photographic images of the womenafter 0, 4, 6 and 8 weeks of product use and evaluating the images by algorithm-based computer imageanalysis for coloured spot area fraction, by expert visual grading, and by chromophore-specific imageanalysis based on noncontact SIAscopy for melanin spot area fraction and melanin chromophore evenness.RESULTS: By all four measures, the niacinamide + NAG formulation regimen was significantly (P < 0.05) moreeffective than the vehicle control formulation regimen in reducing the detectable area of facial spots and theappearance of pigmentation. CONCLUSIONS: A formulation containing the combination of niacinamide +NAG reduced the appearance of irregular pigmentation including hypermelaninization, providing an effectbeyond that achieved with SPF 15 sunscreen.

Arch Biochem Biophys. 2011 Apr 15;508(2):164-70. Epub 2011 Jan 4.The grape antioxidant resveratrol for skin disorders: Promise, prospects, and challenges.Ndiaye M, Philippe C, Mukhtar H, Ahmad N.SourceDepartment of Dermatology, University of Wisconsin, Madison, WI 53706, USA.AbstractResveratrol, a phytoalexin antioxidant found in red grapes, has been shown to have bothchemopreventive and therapeutic effects against many diseases and disorders, including thoseof the skin. Studies have shown protective effects of resveratrol against ultraviolet radiation-mediated oxidative stress and cutaneous damages including skin cancer. Because many of theskin conditions stem from ultraviolet radiation and oxidative stress, this antioxidant appears tohave promise and prospects against a wide range of cutaneous disorders including skin agingand skin cancers. However, there are a few roadblocks in the way of this promising agentregarding its translation from the bench to the bedside. This review discusses the promise andprospects of resveratrol in the management of skin disorders and the associated challenges.

Arch Dermatol Res. 2010 Mar;302(2):71-83. Epub 2009 Nov 7.Skin photoprotection by natural polyphenols: anti-inflammatory, antioxidant and DNA repairmechanisms.Nichols JA, Katiyar SK.SourceDepartment of Dermatology, University of Alabama at Birmingham, 1670 University Boulevard,Volker Hall 557, PO Box 202, Birmingham, AL 35294, USA.AbstractEpidemiological, clinical and laboratory studies have implicated solar ultraviolet (UV) radiationin various skin diseases including, premature aging of the skin and melanoma and non-melanoma skin cancers. Chronic UV radiation exposure-induced skin diseases or skin disordersare caused by the excessive induction of inflammation, oxidative stress and DNA damage, etc.The use of chemopreventive agents, such as plant polyphenols, to inhibit these events in UV-exposed skin is gaining attention. Chemoprevention refers to the use of agents that can inhibit,reverse or retard the process of these harmful events in the UV-exposed skin. A wide variety ofpolyphenols or phytochemicals, most of which are dietary supplements, have been reported topossess substantial skin photoprotective effects. This review article summarizes thephotoprotective effects of some selected polyphenols, such as green tea polyphenols, grapeseed proanthocyanidins, resveratrol, silymarin and genistein, on UV-induced skin inflammation,oxidative stress and DNA damage, etc., with a focus on mechanisms underlying thephotoprotective effects of these polyphenols. The laboratory studies conducted in animalmodels suggest that these polyphenols have the ability to protect the skin from the adverseeffects of UV radiation, including the risk of skin cancers. It is suggested that polyphenols mayfavorably supplement sunscreens protection, and may be useful for skin diseases associatedwith solar UV radiation-induced inflammation, oxidative stress and DNA damage.

J Drugs Dermatol. 2010 Dec;9(12):1523-6.Dose-dependent antioxidant function of resveratrol demonstrated via modulation of reactive oxygenspecies in normal human skin fibroblasts in vitro.Jagdeo J, Adams L, Lev-Tov H, Sieminska J, Michl J, Brody N.SourceDepartment of Dermatology, SUNY Downstate, Brooklyn, NY 11203, USA. [email protected] study of free radicals is particularly relevant in the context of human skin carcinogenesis and photoagingbecause of their ability to induce DNA mutations and damaging lipid peroxidation byproducts. Therefore, itis important to identify and evaluate agents with the ability to modulate intracellular free radicals.Significant interest exists in evaluating the chemotherapeutic and anti-oxidant properties of resveratrol(trans-3,4',5-trihydroxystilbene). Resveratrol is a phytoalexin, a naturally occurring compound derived fromthe skin of grapes and other plants. Resveratrol was selected for evaluation because of demonstratedchemopreventive and chemotherapeutic properties in a murine skin cancer model and other human cancermodels through a variety of mechanisms. However, the intracellular anti-oxidant properties of resveratrolon free radicals in human skin cells in vitro is not well characterized. The purpose of this research is toinvestigate the ability of resveratrol to modulate the hydrogen peroxide-induced upregulation of reactiveoxygen species (ROS) free radicals in normal human skin fibroblast cells in vitro. Hydrogen peroxide is a wellknown generator of free radicals that occurs during endogenous and UV-induced oxidation processes in thehuman skin and was used to upregulate ROS in normal human skin fibroblast cells. Using a flow cytometry-based assay, the results demonstrate highly significant (P<0.001) dose-dependent reduction of intracellularhydrogen peroxide-upregulated ROS by resveratrol at 0.01%, 0.001% and 0.0001% concentrations in humanskin fibroblasts in vitro.

Biofactors. 2010 Sep;36(5):342-9.Effect of resveratrol on endothelial cell function: Molecular mechanisms.Schmitt CA, Heiss EH, Dirsch VM.SourceWolfson Institute for Biomedical Research, University College London, United Kingdom.AbstractThe polyphenolic natural product resveratrol (RV), best known for its occurrence in grape skinand red wine, is considered a candidate drug for prevention and treatment of cardiovasculardiseases. This review aims to summarize the molecular effects of RV on endothelial cells,which line the inner walls of blood vessels and play a key role in the development of thosediseases. We describe how RV enhances endothelial nitric oxide production, improvesendothelial redox balance and inhibits endothelial activation in response to pro-inflammatoryand metabolic insults. Furthermore, we summarize effects of RV on endothelial senescence,apoptosis, endothelin-1 release, and endothelial progenitor cell function. As many of RV'sactions seem to be mediated by SIRT₁, different mechanistic possibilities how RV may lead toSIRT₁ activation are discussed.

Br J Dermatol. 2008 Mar;158(3):472-7. Epub 2007 Dec 6.Repair of photoaged dermal matrix by topical application of a cosmetic 'antiageing' product.Watson RE, Long SP, Bowden JJ, Bastrilles JY, Barton SP, Griffiths CE.SourceDermatological Sciences Research Group, The University of Manchester, Manchester M13 9PT, [email protected]:Photoaged skin is characterized by coarse and fine wrinkles. The mechanism of wrinkle formation appears to involvechanges to components of the dermal extracellular matrix. Topical treatment with all-trans retinoic acid (RA) can repairphotoaged dermal matrix; this is regarded as the 'gold standard' against which repair agents are judged. To date, little isknown regarding the ability of over-the-counter 'antiageing' products to repair photoaged skin.OBJECTIVES:We used a modified occluded patch test to ascertain whether topical applications of cosmetic 'antiageing' products are ableto repair photoaged human skin.METHODS:Commercially available test products [basic moisturizer, 'antiageing' cream containing different active complex levels (6%active: lipopentapeptide, white lupin peptides, antioxidants, retinyl palmitate; 2% active: lipopentapeptide, white lupinpeptides, antioxidants)] were applied under occlusion for 12 days prior to biopsy and histological assessment in photoagedvolunteers (n=9). RA was used as a positive control.RESULTS:In agreement with previous studies, the patch-test study revealed that RA produced significant fibrillin-1 deposition in thepapillary dermis (P<0.01) but had little effect on procollagen I or matrix metalloproteinase-1 expression. The 6% total activecomplex formulation, however, increased the deposition of fibrillin-1 and procollagen I (P<0.01, P<0.05, respectively).CONCLUSIONS:This study indicates that in an in vivo 12-day patch test an over-the-counter cosmetic product can induce changes inphotoaged dermal extracellular matrix, which are indicative of repair.
