2003In recognition of distinguished toxicologists and students, SOT presents several prestigious awards each year. Award recipients receive a plaque and a generous stipend, are listed in the annual Membership Directory, posted on the SOT Web site and are honored at aspecial Awards Presentation at the SOT Annual Meeting.

AchievementThe Awards Committee of theSociety of Toxicology is honored tohave unanimously selected Dr. LoisLehman-McKeeman as the recipientof the 2003 Achievement Award forsignificant contributions to the field oftoxicology.

Dr. Lehman-McKeeman received herundergraduate degree in Toxicologyfrom the Philadelphia College ofP h a rmacy and Science andsubsequently a Ph.D. from the

University of Kansas Medical Center in 1986. Followingher graduate wo rk she joined Procter and Gambl eCompany as a Biochemical Toxicologist, rising through theranks to Principal Research Scientist. In 2001, Dr.L e h m a n - M c Keeman joined Bri s t o l - M yers SquibbCompany where she is presently Research Fellow inInvestigative Toxicology.

The importance of Dr. Lehman-McKeeman's scientificcontributions have been internationally recognized forbeing directed towards investigating mechanisms bywhich chemicals cause toxicity and cancer in animals andthe use of this information for evaluating human risk. Dr.Lehman-McKeeman has published extensively in andcontributed significantly to the understanding of molecularmechanisms and the species specificity of α2u-globulin-mediated renal carcinogenesis. Her research in this areawas critical for elucidating the mechanism by whichchemicals cause this protein to accumulate in the male ratkidney and establishing the lack of human relevance ofthis syndrome. She has also expanded her researchefforts to pursue biochemical and molecular mechanismsof rodent liver carcinogenesis and cell-specific lung toxicityand carcinogenesis. In many cases the scientificinformation she generated and published has guidedregulatory policy in a highly politicized setting.

Dr. Lehman-McKeeman has also actively participated inservice-related activity for SOT. She was first a memberand then chairperson of the Continuing EducationCommittee, Director of the Placement Committee, andelected to serve as member of Council. She was also anAssociate Editor of Toxicological Sciences until recentlywhen she was named as Editor of the journ a l .Dr. Lehman-McKeeman has also participated on a numberof advisory groups in which she was always a keycontributor.

Dr. Lehman-McKeeman continues to make significantc o n t ri butions to mechanistic toxicology usingtechnological advances in both chemistry and molecularbiology and is well deserving of this Award.

Identification of Butyry l c h o l i n e s t e rase Adducts afterInhibition with Isomalathion Using Mass Spectrometry:Difference in Mechanism Between (1R) – and (1S) –Stereoisomers. J. Doorn, M. Schall, D. Gage, T. Talley, C.Thompson, and R. Richardson. TAAP 176, 73–80, 2001.

In the wo rk by Doorn et al. (2001), the authors explore therole of adduct fo rmation in the inhibition of cholinestera s e sby malathion, a major component of the toxicity of thiso r g a n o p h o s p h rus pesticide. The chemical activation ofmalathion produces potent inhibitors of cholinestera s e s,while thermal or photochemical isomerization of thepesticide can produce racemic isomalathion. By masss p e c t r o m e t ry, the authors find that isomer-specificc h o l i n e s t e rase adducts account for isomalathion inhibitionof the enzyme. F u rt h e rm o r e, they demonstrate a diffe r e n c ein the mechanism of inactivation between specific isomers. The differences in the adducts appears to contri bute to theaging process of this biochemical lesion, where the enzymecan no longer be reactiva t e d . The wo rk is elegantly doneand directly addresses a toxic mechanism at the molecular leve l .

A PBPΚ Modeling-Based Approach to Account fo rInteractions in the Health Risk Assessment of ChemicalM i x t u r e s. S. Haddad, M. B e l i veau, R. Ta r d i f, and K. Krishnan. ToxSci 63, 125–131, 2001.

Haddad et al. (2001) presented a novel risk assessmentmethodology for chemical mixtures that accounts for theconsequences of pharmacokinetic interactions amongcomponents. The authors used physiologically-basedpharmacokinetic (PBPK) interaction models to simulatethe change in tissue dose of chemicals during mixedex p o s u r e s. The change in tissue dose of mixturecomponents was used along with their "tissue dose-response" relationship to characterize the ensuing risk.Using this approach, an interaction-based hazard indexwas developed for mixtures of systemic tox i c a n t swhereas an interaction-based response addition wasperformed for mixtures of carcinogens. The applicabilityof these approaches was demonstrated by characterizingthe change in tissue dose of toxic moiety of mixturecomponents according to interaction mechanism andexposure concentrations. For various mixtures of volatileorganic chemicals, such information was then used tocharacterize health risk resulting from mixed exposures.

The methodological approaches developed in this articlefacilitate, for the first time, the consideration of the impactof multichemical pharmacokinetic interactions at aquantitative level in mixture risk assessments.

Dr. Pitot's research interests have been directed towardsunderstanding the multistage process of chemicalcarcinogenesis using morphological and biochemicalapproaches. He published his first two papers in 1954 andhas since published over 500 high-quality and frequently-cited scientific papers. He is well known for hisidentification of the va rious stages inhepatocarcinogenesis that have provided models for bothg e n o t oxic and nongenotoxic carcinogens. H i scharacterization of the heterogeneous nature of hepaticfoci and methods for quantifying the number and size ofthese precursor lesions allowed the quantitativeassessment of initiating and promoting potential ofdifferent chemicals. This work has had significant impacton regulatory agencies and public policy to benefit moreaccurate cancer risk assessment. He continues to makemajor contributions by incorporating molecular biology andthe development and use of transgenic animals as tools inhis investigations.

In addition to Dr. Pitot's outstanding research career, heserves the toxicology, pathology, and cancer researchcommunities as a mentor, reviewer, and advisor. He is agreat scholar and teacher who has encouraged and

mentored many young investigatorsin these fields and is highly deservingof the Distinguished Life t i m eToxicology Scholar Award.

EducationDr. Frederick W. Oehme, Professor ofTox i c o l o g y, Pa t h o b i o l o g y, Medicineand Physiology in the College ofVeterinary Medicine at Kansas StateUniversity, has been selected as the2003 Educator Award winner.Dr. Oehme has been an important

and continuing part of toxicological education for manyyears. His efforts extend from his university, to SOT, andbeyond to the international arena. His early efforts inbuilding the graduate training program in toxicology atKansas State are noteworthy as is his mentoring of oversixty graduate and post-doctoral students. The success ofthese students in examinations for accreditation by theAmerican Board of Toxicology and/or the American Boardof Veterinary Toxicology as well as in their subsequentcareers is a fitting tribute to his efforts.

Dr. Oehme's strong interest in continuing education and inSOT led him in 1980 to propose, organize, and present in1980 the first SOT Continuing Education course. In the 23years since then the Society's Continuing EducationCourses have become a valuable source of information fort oxicologists and an important part of SOT Annu a lM e e t i n g s. C o n c e rn with professional standards int oxicology led to Dr. Oehme's invo l vement in theestablishment of the American Board of Toxicology,

including chairing the initial Constitution and By-LawsCommittee and subsequent service on the ExecutiveC o m m i t t e e. In the international arena he has beeninvolved with training activities in many countries and hasserved as a toxicology consultant to FAO and WHO for thedevelopment of poisoning prevention programs around theworld.

In summary, it can be said that Dr. Oehme's contributionsto toxicology education have been many and varied andthe result of a deep and abiding commitment. His impacton toxicology has been, and continues to be, importantand valuable.

Enhancement of AnimalWelfare

The 2003 Enhancement of Animal Welfare Award hasbeen awarded to Dr. Ian Kimber and Dr. G . Fra n kGerberick. They have been active in the field of toxicologyfor over 20 years, and they are both the authors or co-authors of over 300 publications on the subject ofpredictive testing in immunology.

G. Frank Gerberick and Ian Kimber recently sponsored theMurine Local Lymph Node Assay for review by the Inter-Agency Coordinating Committee for the Validation ofA l t e rn a t i ve Methods Peer Rev i ew Panel The panelunanimously recommended that, with appropri a t equalifications, the Local Lymph Node Assay was at leastas accurate as current guinea pig methods for the hazardidentification of strong to moderate chemical sensitizingagents and offered “several advantages with respect toanimal use refinement compared to conventional guineapig methods in that it involves less pain and distress.” As ac o n s e q u e n c e, this assay is now accepted by manyregulatory bodies, including the U.S. Food and DrugA d m i n i s t ration, as a valid predictor of immu n esensitization.

The Local Lymph Node Assay is an in vivo assay, whichenhances the welfare of the laboratory animal by reductionof distress without compromise of data quality. This is akey goal of all scientists truly interested in animal welfare, while recognizing the need for their continued use.

MeritD r. M . W. " D rag" Anders is therecipient of the 2003 Merit Award.Drag is considered by his peers to beamong a select group whoseresearch in the field of drug andchemical bioactivation can beconsidered "distinguished." His workon the bioactivation, cova l e n tbinding, and toxicity of halogenatedhydrocarbons is arguably thedefinitive body of work. When Dragentered research, the cova l e n tbinding hypothesis of toxicity was in

its infancy, and the focus of the field was to elucidate thenature of the biologically reactive intermediates. In the1970s, he carried out some of the earliest work onchloroform, dichloromethane, and carbon tetrachloridethat actually proved the hypothesis by linking the biologicalresponse of the parent compound to the chemistrythrough the unequivocal structural identification of thereactive intermediates. He used his chemical knowledgeto push electrons and with various techniques proved thenature of the involvement of reactive species in toxicity. This rigorous proof of mechanism through elegant andoriginal experimental science coupled to a commitment toexcellence in research is what has distinguished Drag'swork throughout his career in toxicology. Drag hasreceived a number of prestigious awards including theBernard B. Brodie Award in Drug Metabolism and aMERIT (Method to Extend Research in Time) award fromNIEHS. He has received and held distinguished chairs andlectureships positions as well as serving as the Chair ofthe Department of Pharmacology and then theDepartment of Pharmacology and Physiology after mergerof these two departments at the University of Rochester. He has served in va rious advisory capacities inrecognition of his stature in the field of toxicology. Drag'scareer is also distinguished by his record of training manyoutstanding students. T h u s, Drag's career is tru l ydistinguished, in his research, training, and service andmost importantly in his character and personality.

Award inInhalationToxicology

The 2003 Early Career Award inInhalation Toxicology is presented toDr. Ilona Jaspers, Assistant Professorin the Department of Pe d i a t ri c s,Division of Infectious Diseases,U n i versity of North Carolina atChapel Hill. D r. Jaspers' Awa r d ,which is the second of its kind to beawarded through SOT, will be used to

s u p p o rt wo rk under her proposal "Diesel-InducedAlterations of Influenza Pathogenesis." This work will seekto establish whether exposure to diesel exhaust particlesrenders airway epithelial cells more susceptible toinfluenza-induced pro-inflammatory responses and thusenhances the pathogenesis associated with influenzainfections. In addition, this project will determine whetherantiviral defense responses of airway epithelial cells arecompromised by prior exposure to diesel exhaust particles, leading to an advantage of the invading virus over the hostdefense system. Dr. Jaspers obtained her Bachelor ofScience degree in Biology at Seton Hall University, andwas an NIEHS Pre-Doctoral Fellow at New York Universitywhere she earned her Ph.D. in Toxicology in 1997 under Dr.Lung Chi Chen.
