1.1. About this Manual

This manual will help you to handling the device. Please read the manual completely and follow the advice. Acquaint yourself to the control elements and their functionality by studying the figures. This way, an adequate use of the device, is ensured. If you have problems, feel free to contact the competent MEDITE contact person.

Working with the machine contents crushing hazard from the hand wheel and the specimen clamp.

Crushing hazard: Working on moveable components of the machine should be avoided. These warnings are marked with a yellow triangle and a crushing hand inside.

Warning: This symbol warns you of risks for machine, material or environment. Warnings are marked with a yellow warning triangle.

Working with this machine contents cutting hazards from the knife carrier

and specimen clamp (moving).

Cutting hazard: Working with this machine contents cutting hazards for

fingers and hand. These warning are marked with a yellow triangle and a

cutting hand inside.

Notice: Notices, meaning important information for the user, that are not classified as danger or warning, are marked with an information symbol.

Danger warning: This symbol warns you of risks for the life or health of persons. Danger warnings are marked by a red framed triangle.

1.4. Personal protective equipment

When working with the Microtome M530, it is necessary, according to you security policies applicable in any case, to wear protective clothings to avoid direct contact with the biologic samples. The same applies, for example, when cleaning the unit.

1.5. Transport and stocking

Observe the following notes for transport and stocking the Microtome M530.

1.6. Environmental conditions

The permissible ambient conditions for operation of the device are listed below. Operation at other ambient conditions is not permitted.

• Operation is only allowed in enclosed rooms,

• Ambient temperature from +15°C to +40°C (without strong fluctuations),

• Only set it on a vibration-free, the weight of the device corresponding laboratory bench,

• Relative air humidity max 80% non-condensing,

• The unit must not be exposed to direct sunlight,

• Maximum height of 2000 m (according to EN 61010-1:2001),

• Mains voltage fluctuation is within + / - 10 % of rated voltage.

Always use the carrying device for lifting and carrying the Microtome M530 make sure to avoid a correct attitude about material things and especially physical damage when lifting!

The Microtome M530 is supplied in a safety transport box. Transporting them always upright and avoid shocks and extreme temperature fluctuations! For the storage of the device an ambient temperature of +5 ° C to +50 ° C is permissible.

Always wear protective clothing (e.g. lab coat, gloves,) to prevent accidental contact with biological samples.

„Top“: The packaged unit must always be transported and stored so that the arrows point upwards at any time.

"Protect from wetness": The packaged unit must be protected from excessive humidity and must be stored and transported in covered.

1.7. Safety measures on installation location

The Microtome M530 must be installed on a stable, level and horizontally oriented base to ensure the safe and reliable operation.

1.8. Protectiom measures of the device

1.9. Intended Use

The Microtome is a highly efficient multi-purpose microtome which can be used for any tasks

in paraffin, hard- and thin-sectioning techniques in biology, medicine, industry and research.

A use in food industries and similar facilities is strictly prohibited. Arbitrary rebuilding or

changing of this laboratory instrument is strictly prohibited due to safety reasons. Use only

original MEDITE spare parts in case of replacement issues.

Operation, maintenance or service conditions mentioned in this manual have to be strictly observed.

1.10. Approved Operators

Only persons who have been thoroughly trained by the operator and commissioned can handle the Microtome M530. The operator is responsible for other people in the work area. The operating company make the instruction manual accessible to the operator and make sure, that the operator have read and understand the instruction manual.

Unauthorized modifications to the device are forbidden due to safety reasons. When replacement damaged parts of the device only original spare parts must be used.

You should always activate these two safety precautions, when the

Microtome is not in use.

The handwheel brake – if activated – keeps the handwheel in its position

and protect the operator from crushing hazards.

Don’t use the device in an explosive area.

The knife carrier comes with a finger protection, which always should be

used to decrease the risk of injury.

2.3. Transport

When unpacking, carrying or moving the Microtome M530, use the recessed handles, which

are positioned at the front and at the back of the unit.

Do never use the knife holder or the section waste tray as handle for

carrying!

Before every transport remove the knife holder and the section waste tray!

3. Startup the Microtome M530

The Microtome M530 must be installed on a stable, level and horizontally

oriented base to ensure the safe and reliable operation.

Before start to work on the Microtome M530, leave the unit on the

designated workspace for at least 2 hours or more, to adjust to room

temperature.

Remove the microtome blade and activate the handwheel brake, when

dismounting the Microtome M530!

4. Operation

4.1. Power On

Check, if the on / off switch is in the „0“ (off) position. If not, press the switch to the „0“

position. Plug in the power cord and press the on / off switch in the „I“ (on) position. The

Microtome M530 is ready for operation, if the display of the film tastatur light on.

4.2. Prepare the specimen

4.2.1. Object orientation

The angle of the cassette clamp can adjust to 8° in all directions. To adjust the angle, release

the cassette clamp by turning the lever (3) in your direction. After this you can rotate the

cassette clamp for 360° and you can adjust the angle by turning the angle-adjustment

screws (1, 2). Screw (1) is for the vertical adjustment, screw (2) for the horizontal

adjustment.

When finished the adjustment, fix the cassette clamp by turning the lever (3) in the fix-

position.

Always release the lever (3) before adjusting the angle adjustment. If not,

the object clamp can be damaged.

Make sure the contact surfaces of the cassette are free from paraffin to

achieve an optimal clamping.

After a long period of non-use, pull of the plug in to protect the device by lightning strike or something near it.

4.2.2. Insert the specimen

To insert a specimen, pull the lever (1) in your direction, and then insert the specimen in the

cassette clamp (2).

To remove the specimen, press the lever (1) in your direction and remove the specimen.

4.2.3. Remove the Cassette clamp

If you want to install another cassette clamp, turn the lever (1) in your direction.

After this remove the complete lever by pulling out the the right side. Now you can remove

the cassette clamp. Notice that the cassette clamp must be in center before you remove it.

Insert the new cassette clamp, notice, that the hole of the adapter need to fit with the hole

from the object clamp. After this, insert the lever in the object clamp and fix it by pressing the

lever in the fix-position.

Turn the cassette clamp in the center position, before you start to remove

the cassette clamp.

The screws of the angle adjustment have a lock function for center the

cassette clamp. If you will center the cassette clamp turn the both screws as

long as they locked. When both screws locked, the cassette clemp is in the

center.

4.3. Prepare knife carrier

4.3.1. Inserting the microtome blade

To insert the blade, turn the finger guard to the front and lose the insertion for the blade by

pressing the lever (1) to the front. Now the insertion for the blade will open.

The gap from the blade carrier will open.

Now insert the microtome blade carefully in the gap (2)! Press the lever (1) back to fix the

microtome blade.

The knife carrier has ergonomic grips, which can be set in each position. To

positioning the grips pull the grips careful out and turn it in the new position.

Lose the grips for lock.

After every operation or non-use, put the finger guard over the microtome

blade and activate the brake for the handwheel!

4.3.2. Adjustment of the knife carrier

The complete knife carrier can be moved forward and backward on the rails or taken off the

rails. Pull up the lever (1) to release the knife carrier. Turn the knife carrier in the new

position and pull down the lever (1) to fix the knife carrier.

4.3.3. Adjustment of the blade holder

The blade holder can be moved in the right or left direction. Pull down the lever (1) to release

the blade holder. Turn the blade holder in the new position and pull up the lever (1) to fix the

blade holder.

4.3.4. Adjusting The Knife Carrier Angles

The knife carrier has a scala for the angle-adjustment on the left side (2). The angle can

adjust from 6 to 12 degree.

Pull down the lever (1) to release the knife carrier. Turn the knife carrier in the new position

and pull up the lever (1) to fix the blade holder.

4.3.5. Change the microtome blade

To insert the blade, turn the finger guard to the front and lose the insertion for the blade by

pressing the lever (1) to the front. Now the insertion for the blade will open.

Take the cleaning brush - the brush has an integrated magnet on the back, which simplifies

the removal of the microtome blade. Move the back of the cleaning brush to the microtome

blade until the magnet adheres to the blade and pull the blade carefully out of the blade

holder head.

Put the microtome blade in the knife carrier head and press up the lever (1).

4.4. Use the handwheel brake

Tip the lever from “open“ position to “close” position. The lever will automatically move to

“close“ position. Avoid to overwinding the lever to the mechanical end position.

Turn down the lever to release the handwheel.

Do not rotate the handwheel while the brake is activated. The device can be

damaged!

4.5. Start the cutting process

4.5.1. Trimming the specimen

Without turning the handwheel tip button (8) to approach the specimen to the blade in min

5µm steps or in actual user selected trim value (example 20µm) . One tip one movement to

the specimen. Acustic signal confirmed action.

Hold pressing button (8) and turn the handwheel each turn moves the specimen min 5µm or

user selected trim value to the blade.

Use a different area of the cutting edge for trimming than for sectioning by

moving the blade holder left or right.

After every operation or non-use, put the finger guard over the microtome

blade and activate the brake for the handwheel!

Before the cutting process starts, the knife carrier and specimen clamp

needs to adjusting. For more Information, read chapter 4.2 Prepare the

specimen and chapter 4.3 Prepare the knife carrier.

If you want to move the specimen away from the blade tip button (6) each tip moves the

specimen 6µm away from the blade. Hold pressing button (6) until second acustic signal

appears moves the specimen automatically into home position. You can stop that

automatically movement by again tip button (6).

Check if "Trim" is shown on the display. Set the slice thickness by turning the multiple

functional knob (Example 50 µm). The display shows the currently set cut thickness.

Cut the sample by turning the hand wheel until the desired object area is reached.

4.5.2. Sectioning the specimen

Check if "Fine" is shown on the display. Set the section thickness by turning the multiple

functional knob (Example 5 µm). The display shows the currently set cut thickness.

Cut the sample by turning the hand wheel until the desired object area is reached.

4.5.3. Automatic Retraction

To prevent the knife and the specimen from damage when the object clamp is moving up again, the specimen can be pulled back during upward movement by activating the automatic “Retraction”. The “Retraction” can be switched on and off by pressing button (2).

4.5.4. Position Memory

The M530 offers the possibility to store 3 values for trimming positions. These values can be used to recover certain positions within a short time. A position can be stored by pushing one of the button (3), (4) or (5) for more than one second until a signal appears. The value of the actual positioning then is stored in your chosen memory and can be approached any time later on by just pushing the memory key again. Occupied memory numbers are shown “M1 … M3” including their values in the display to avoid an overwriting of your positions. Of course you can overwrite your values just by proceding the same way again.

If the function is activated there is an “R” appearing in the display.

4.6. Troubleshooting

4.6.1. Change the Fuse

To change the fuses switch-off the device. Press the on / off switch in the "0" position and

remove the power cord. Then squeeze the two pins (1) and pull the drawer (2) with the

fuses out carefully.

In chapter 2.2 - Technical Data are the data of the fuse. Replace the defect fuse (3) with a

new fuse and slide the tray back into the appropriate slot until you hear a clear click.

After each restart of the Microtome these values are deleted for safety.

5. Cleaning, Maintenance and Recycling

5.1. Cleaning Instructions

The Microtome M530 is a precision instrument. Of course it is necessary to keep it clean to

guarantee a long timespan between maintenance checks. So, after each use, you have to

clean the specimen clamp, the knife carrier, the section waste tray and all visible guiding.

The surface of the Microtome may be cleaned with standardized, not scratching cleaning

agents, if needed. To remove sticking deposits of sections, use a brush. Don’t use sharp-

edged blades or tools!! These will destroy the surface of the Microtome. The whole

Microtome has to be checked regularly by MEDITE service.

5.2. Timing of Cleaning And Maintenance

Before starting the cleaning, always switch-off the device and pull of the power cord.

A regularly maintenance of the Microtome M530 (cleaning and greasing of

the relevant mechanical parts, cleaning of parts inside and exchange of

expandable parts) by MEDITE Service is the main requirement for a well

working microtome! Ask your local MEDITE representative for a

maintenance check.

MEDITE recommends you to include a daily cleaning procedure – after

finish of routine work - into your cleaning and maintenance plan.

5.3. Abandonment / Recycling

After expiration of instrument’s life span please transfer unit into professionally disposal or

send it back to MEDITE. As a manufacturer / distributor we take back the used equipment,

requiring a payment for disposal.
